Three-dimensional reconstructed canopies of apple cultivars grafted on different interstocks
<p>Canopy architecture is important for a range of orchard ecosystem processes, including radiation interception and distribution within canopy, flowering, carbon assimilation, fruit yield and quality and pest and pathogen attacks in fruit trees. The canopy of fruit tree varies greatly from cultivar to cultivar, and can be manipulated by horticultural practices, such as rootstock, pruning, training etc. We collected canopy architectural data in ‘Fuji’ and ‘Gala’ apple cultivars which were grafted onto two rootstock/interstock combinations: <em>M. micromalus</em>/<em>M. domestica</em> cv. ‘Qinguan’, a vigorous interstock and <em>M. micromalus</em>/M26, a dwarf interstock. Allometric relationships at leaf and shoot scale were determined for all shoot types before harvests in 2011 and 2012, including bourse, bourse short shoot, bourse long shoot, vegetative short shoot, and vegetative long shoot. A dataset of 4351 records of leaf Euler angles were digitized for all shoot types. For 19 trees, their 3D distribution of the foliage within canopy was reconstructed by combining both the spatial locations of shoots (as measured from 3D digitizing) and foliage reconstruction based on allometric relationships and leaf angle distributions. We believe this dataset is a valuable resource for studies and models that require leaf angles and detailed 3D virtual canopies.</p>
Improved antibacterial activity of hemp fibres by covalent grafting of quaternary ammonium groups
<p><span><span>In this study, novel antibacterial hemp fibres grafted with </span></span>quaternary<span><span> ammonium groups, named HF-GTA, were prepared by alkalisation, oxidation, amination, and quaternisation multistage reactions. The chemical structure and micromorphology of the fibres were characterised by Fourier-transform infrared spectroscopy, scanning electron microscopy, X-ray photoelectron spectroscopy, thermogravimetric analysis, and X-ray diffraction. The grafting and reaction mechanisms were proved to be successful, which indicated that the grafting reaction primarily occurred on the </span></span>hydroxyl <span><span>groups of cellulose and hemicellulose in the hemp fibres, where they retained good fibrous morphology, thermal stability and hygroscopicity. The antibacterial activity of HF-GTA exhibited the best antibacterial activity, where the antibacterial ratios against <i><span>E. coli </span></i>and <i><span>S. aureus</span></i> were 95.41% and 99.64%, respectively. <span>E</span><span>ven </span><span>a</span><span>fter 30 times of washing</span><span>, </span><span>the antibacterial activity </span><span>wa</span><span>s</span><span> retained</span><span> at </span><span>89.78% and 91.12%</span><span>,</span><span> indicating that HF-GTA </span><span>wa</span><span>s</span><span> endowed with good washing resistance. </span>The antibacterial mechanism occurred because the electrostatic reaction reduced the electrochemical potential on the cell membrane, leading to the release of cytoplasmic substances and the dissolution of cells, thereby producing antibacterial activity. It would be significantly important to guarantee textile quality and prevent disease transmission. </span></span></p>
Data from: Crossing phenotype heritability and candidate gene expression in grafted black-lipped pearl oyster Pinctada margaritifera, an animal chimera
Grafting mantle tissue of a donor pearl oyster into the gonad of a recipient oyster results in the formation of a chimera, the pearl sac. The phenotypic variations of this chimera are hypothesized to be the result of interactions between the donor and recipient genomes. In this study, the heritability of phenotypic variation and its association with gene expression were investigated for the first time during P. margaritifera pearl production. Genetic variance was evaluated at different levels, 1) before the graft operation (expression in graft tissue), 2) after grafting (pearl sac tissue expression in chimera) and 3) on the product of the graft (pearl phenotype traits) based on controlled bi-parental crosses and the F1 generation. Donor related genetic parameter estimates clearly demonstrate heritability for nacre weight and thickness, darkness and colour, surface defects and grade, which signifies a genetic basis in the donor oyster. In graft relative gene expression, the value of heritability was superior to 0.20 in for almost all genes, while in pearl sac, heritability estimates were low (h² < 0.10) (except for CALC1 and Aspein). Pearl sac expression seems to be more influenced by residual variance than the graft, which can be explained by environmental effects that influence pearls sac gene expression and act as a recipient additive genetic component. The interactions between donor and recipient are very complex and further research is required to understand the role of the recipient oysters on pearl phenotypic and gene expression variances.

This retrospective cohort study aims to compare the clinical efficacy and safety of amnio-graft and skin graft procedures performed in the Plastic Surgery Department of Al Mujtahid Hospital in Damascus, Syria.</p><p>II.<strong> Objectives</strong>:</p><p>To assess the clinical efficacy of amnio-graft and skin graft procedures.</p><p>To evaluate the safety profile of amnio-graft and skin graft procedures.</p><p>To compare the outcomes of patients undergoing plastic surgery with amnio-graft versus skin graft.</p><p>III. <strong>Study Design</strong>:</p><p>This study will adopt a retrospective cohort design, analyzing data collected from patient records in the Plastic Surgery Department during the specified study duration.</p><p>IV. Treatment with 3-aminobenzamide during ex vivo lung perfusion of damaged rat lungs reduces graft injury and dysfunction after transplantation
<p>Ex vivo lung perfusion (EVLP) with pharmacological reconditioning may increase donor lung utilization for transplantation (LTx). 3-Aminobenzamide (3-AB), an inhibitor of poly (ADP-ribose) polymerase (PARP), reduces ex vivo lung injury in rat lungs damaged by warm ischemia (WI). Here we determined the effects of 3-AB reconditioning on graft outcome after LTx. Three groups of donor lungs were studied: Control (Ctrl): 1 hour WI + 3 hours cold ischemia (CI) + LTx; EVLP: 1 hour WI + 3 hours EVLP + LTx; EVLP + 3-AB: 1 hour WI + 3 hours EVLP + 3-AB (1 mg<sup>.</sup>mL<sup>−1</sup>) + LTx. Two hours after LTx, we determined lung graft compliance, edema, histology, neutrophil counts in bronchoalveolar lavage (BAL), mRNA levels of adhesion molecules within the graft, as well as concentrations of interleukin-6 and 10 (IL-6, IL-10) in BAL and plasma. 3-AB reconditioning during EVLP improved compliance and reduced lung edema, neutrophil infiltration, and the expression of adhesion molecules within the transplanted lungs. 3-AB also attenuated the IL-6/IL-10 ratio in BAL and plasma, supporting an improved balance between pro- and anti-inflammatory mediators. Thus, 3-AB reconditioning during EVLP of rat lung grafts damaged by WI markedly reduces inflammation, edema, and physiological deterioration after LTx, supporting the use of PARP inhibitors for the rehabilitation of damaged lungs during EVLP.</p>
Data from: Grafting or pruning in the animal tree: lateral gene transfer and gene loss?
Background: Lateral gene transfer (LGT), also known as horizontal gene transfer, into multicellular eukaryotes with differentiated tissues, particularly gonads, continues to be met with skepticism by many prominent evolutionary and genomic biologists. A detailed examination of 26 animal genomes identified putative LGTs in invertebrate and vertebrate genomes, concluding that there are fewer predicted LGTs in vertebrates/chordates than invertebrates, but there is still evidence of LGT into chordates, including humans. More recently, a reanalysis of a subset of these putative LGTs into vertebrates concluded that there is not horizontal gene transfer in the human genome. One of the genes in dispute is an N-acyl-aromatic-L-amino acid amidohydrolase (ENSG00000132744), which encodes ACY3. This gene was initially identified as a putative bacteria-chordate LGT but was later debunked as it has a significant BLAST match to a more recently deposited genome of Saccoglossus kowalevskii, a flatworm, Metazoan, and hemichordate. Results: Using BLAST searches, HMM searches, and phylogenetics to assess the evidence for LGT, gene loss, and rate variation in ACY3/ASPA homologues, the most parsimonious explanation for the distribution of ACY3/ASPA genes in eukaryotes involves both gene loss and bacteria-animal LGT, albeit LGT that occurred hundreds of millions of years ago prior to the divergence of gnathostomes. Conclusions: ACY3/ASPA is most likely a bacteria-animal LGT. LGTs at these time scales in the ancestors of humans are not unexpected given the many known, well-characterized, and adaptive LGTs from bacteria to insects and nematodes.

<p>This record contains raw data related to the article “LUMINAL ENDOTHELIALIZATION OF SMALL CALIBER SILK TUBULAR GRAFT FOR VASCULAR CONSTRUCTS ENGINEERING"</p> <p>The constantly increasing incidence of coronary artery disease worldwide makes necessary to set advanced therapies and tools such as tissue engineered vessel grafts (TEVGs) to surpass the autologous grafts [(i.e., mammary and internal thoracic arteries, saphenous vein (SV)] currently employed in coronary artery and vascular surgery. To this aim, <em>in vitro</em> cellularization of artificial tubular scaffolds still holds a good potential to overcome the unresolved problem of vessel conduits availability and the issues resulting from thrombosis, intima hyperplasia and matrix remodeling, occurring in autologous grafts especially with small caliber (<6 mm). The employment of silk-based tubular scaffolds has been proposed as a promising approach to engineer small caliber cellularized vascular constructs. The advantage of the silk material is the excellent manufacturability and the easiness of fiber deposition, mechanical properties, low immunogenicity and the extremely high <em>in vivo</em> biocompatibility. In the present work, we propose a method to optimize coverage of the luminal surface of silk electrospun tubular scaffold with endothelial cells. Our strategy is based on seeding endothelial cells (ECs) on the luminal surface of the scaffolds using a low-speed rolling. We show that this procedure allows the formation of a nearly complete EC monolayer suitable for flow-dependent studies and vascular maturation, as a step toward derivation of complete vascular constructs for transplantation and disease modeling.</p>
Reimagined MPFL Reconstruction: Retinacular fixation of the doubled hamstring graft at the patella and suture anchor-based femoral fixation
<p class="MsoNormal"><strong>Background.</strong> Lateral patellar dislocation is frequently observed among teenagers and young adults. There is no consensus on the best type of graft or fixation strategy for the femur and patella, and complications such as iatrogenic patella fracture, tunnel malposition, and grafting failure are common. The objective of our research is to outline a new method of medial patellofemoral ligament (MPFL) reconstruction, which involves two key components: (1) Patellar fixation is accomplished by suturing the two limbs of the looped doubled hamstring graft in a divergent fashion to the retinaculum at the medial border of the upper half of patella and, (2) the placement of a suture anchor tied to the graft at the isometric point on the medial femur condyle.</p> <p class="MsoNormal"><strong>Methods.</strong> This study is a retrospective assessment of patients who underwent MPFL reconstruction at our hospital during the period between September 2018 and August 2020. Patients were monitored for at least 2 years after the initial procedure until August 2022.</p> <p class="MsoNormal"><strong>Results.</strong> A total of 29 patients were recruited for the study, with 22 being females and the average age being 30.38 years. During the postoperative period, none of the participants experienced instability, re-dislocation, or patellar/femoral fractures. The Tegner-Lysholm Knee Score was good to excellent for 17 (58.6%) participants, fair for 10 (34.5%) participants, and poor for 2 (6.9%) participants. The Kujala Anterior Knee pain score was more than 80 for 19 (65.5%) participants. No cases of abnormal growth were reported during a follow-up period of 30.6 months.</p> <p class="MsoNormal"><strong>Conclusion</strong>. The outcomes observed with this technique in terms of function were comparable to those reported with alternative surgical methods.</p>

Secondary Graft Loss After Third-party "Off-The-Shelf" Antigen-Specific T cell Infusion
<p>Here, we describe a severe adverse event of virus-specific T cell (VST therapy) in an infant with severe combined immunodeficiency who received VSTs for treatment of CMV viremia post-transplant and developed secondary graft rejection at 1-month post-VST infusion associated with the expansion of VST donor-derived T cells. Deposited here are the bulk repertoire, single-cell gene expression and TCR, and flow cytometry data from the study.</p><p><strong>bulk_rep.zip contains tables of alpha/beta TCR sequences for post-infusion, VST donor, and VST product samples, and samples stimulated with neoepitope peptides. </strong></p><p><strong>VST_agg_wTCR_SS.rds is an R Data Serialization file containing a Seurat object of the processed 10X single-cell gene expression and TCR profiling for the post-infusion, VST donor, and VST product samples.
