PEW01 Plant species composition and edaphic properties in wallow and non-wallow plots at Konza Prairie

Bison wallows—bare depressions formed by repeated trampling and dust-bathing—introduce fine-scale heterogeneity to tallgrass prairie soils and plant communities. We compared plant communities and edaphic properties of wallows to adjacent non-wallow plots and cattle-grazed plots. Wallows had reduced organic matter and nitrogen but elevated clay and sodium. Non-fenced wallows had lower plant species richness than fenced wallows and all non-wallows. However, wallows supported distinct plant communities, including wetland species in temporary pools. While bison-grazed areas generally had higher plant diversity, wallows added localized variation not found in cattle or fenced plots. Our results suggest wallows function as small-scale ecosystem engineering features, shaping soil conditions and plant composition.

Hyperspectral reflectance values and biophysicochemical properties of biocrusts and soils in the Fryxell Basin, McMurdo Dry Valleys, Antarctica (2019)

This data package includes ecological parameters of biocrust and soil from samples collected in-situ within the Lake Fryxell Basin of the McMurdo Dry Valleys, Antarctica during December of 2019. Parameters include biological (ash-free dry mass, pigment concentration, and counts of soil invertebrates), physical (water content, electrical conductivity, and pH), and chemical properties (inorganic nitrogen, inorganic phosphorous, total nitrogen, and total organic carbon) of the surface soil, biocrust, and underlying soil. This data package also contains reflectance measurements of biocrust, soil, and granite samples acquired in a laboratory using a hyperspectral spectrometer. Included are hyperspectral reflectance measurements of a laboratory study using a variety of mixtures of soil and biocrust (0 – 100% biocrust), as well as reflectance measurements of individual grab samples collected from each of the field plots. These data aid our understanding of the ecological structure and functioning of biocrust microhabitats as well as the location of these communities throughout the Lake Fryxell Basin. This work also aims to assist in understanding the carbon budget of the basin and highlight the importance of these snowpack-fed biocrust communities, which are understudied but likely an important piece of the overall carbon budget in this region.

Biophysicochemical properties and hyperspectral reflectance values of biocrust and soil samples, Fryxell Basin, McMurdo Dry Valleys, Antarctica (2022)

This data package includes biophysicochemical properties of biocrust and soil samples collected in-situ within the Lake Fryxell basin of the McMurdo Dry Valleys, Antarctica during December 2022 at 64 different terrestrial locations. These parameters include biological (ash-free dry mass, pigment concentration, soil invertebrate counts), physical (gravimetric water content, electrical conductivity, pH), and chemical (inorganic nitrogen, inorganic phosphorous, total nitrogen, soil organic carbon) properties of the surface soil, biocrust, and underlying soil. This package also contains reflectance measurements of individual grab samples of biocrust, soil, and rock collected from each of the field sites, acquired in a laboratory using a hyperspectral spectrometer. Additionally, this package contains parameters derived solely from geospatial data for each of the field sites, including aspect, slope, elevation, gravimetric water content, and snow frequency. These data were used to model habitat suitability of biocrusts in the Fryxell Basin using both in-situ field survey data and geospatial parameters. They aid in our understanding of the ecology of biocrust communities, where they are located throughout the Fryxell basin, and the structure and functioning of these microhabitats as well as improving understanding of the regional carbon budget. These data also highlight the importance of snow patch-associated biocrust communities, which are understudied but likely an important piece of the overall carbon budget in this region.

Biophysicochemical properties, pigment concentrations, and nif gene counts of microbial mats and soils from Taylor and Beacon Valleys, McMurdo Dry Valleys, Antarctica (2019-2020)

This data package includes biophysicochemical properties, pigment concentrations, and nif gene counts from microbial mat and soil samples collected from the McMurdo Dry Valleys of Antarctica during the 2019-20 austral summer. Specifically, data include physicochemical properties of the collected soils (gravimetric water content, pH, electrical conductivity, inorganic nitrogen, inorganic phosphorous, sulfate, chloride, soil organic carbon, and total nitrogen), biological properties of the microbial mats (ash-free dry mass and pigment concentrations – scytonemin, scytonemin-red, myxoxanthophyll, zeaxanthin, chlorophyll-a, chlorophyll-b, β-carotene, canthaxanthin, echinenone), and nif gene counts (overall nif and nifH) of the soils and microbial mats. These data were collected to infer the functioning and nitrogen cycling abilities of microbial mat and soil communities from areas of differing landscape histories and geochemical legacies (Ross and Taylor tills in the Taylor Valley, and Beacon Cirque in Beacon Valley).

Geum and Kobresia soil inorganic and organic property data for Saddle and North of Tvan, 1993.

This study was initiated to examine bulk density and organic matter content differences between soils where Geum (Acomastylis) rossii and Kobresia myosuroides were present. A subset of samples were also measured for total C, N, and P. Paired plots were randomly selected in locations where Kobresia and Geum populations were adjacent to one another. Soil samples were collected from 35 plots, 22 and 13 of which had southerly and northerly aspects, respectively. Soil cores were removed with 3.5-cm interior diameter PVC pipe that was driven into the tundra by use of a rubber mallet. The minimum depth of individual cores was 9 cm, and these depths were recorded at the time of removal (15 July, 19 July, 22 July, and 9 August 1993).

Krummholz island soil inorganic and organic property data for East of Tvan, 1995 - 1996.

Previous work has shown that passage of Engelmann spruce (Picea engelmannii) and subalpine fir (Abies lasiocarpa) tree islands across tundra lowers the soil carbon and nitrogen storage capacity of the top 15cm of soil (A horizon) (Pauker and Seastedt 1996). This study shows that levels of KCl extractable ammonium and percent organic matter were also significantly higher in the A horizon of undisturbed tundra sites compared with soils underneath or immediately adjacent to (windward or leeward) the krummholz. The response of soil KCl extractable NO3- also showed this trend but was not statistically significant. Holtmeier and Broll (1992) suggested that the depletion of organics and nutrients following the passage of tree island may be associated with a reduced clay content. We analyzed a subsample of these soils for cation exchange capacity (CEC) and texture. We did not find significantly lower clay content in soils under or adjacent to krummholz compared with those from undisturbed tundra. In fact, percent clay was greater in krummholz and windward sites than in tundra sites. The percent clay of windward sites was significantly greater in windward soils than either krummholz or tundra soils. Clearly, depletion of organics following the passage of tree islands does not appear to be associated with a depleted clay content. We found that CEC was very highly significantly correlated with percent organic content (using percent organic data only from the subset of soils on which CEC was measured). Therefore, the CEC content of these soils would appear to be strongly associated with the organic content but not with the clay content. Percent soil moisture was significantly higher directly underneath the krummholz compared with the other sites. There was no effect of treatment on pH. Soil inorganic and organic property data for subalpine forest, treeline, and alpine zone, 1999.

This study was initiated to examine the nitrogen content of three montane soils: subalpine, treeline and alpine; and to determine if the differences in soil nitrogen content were attributed to plant community and elevation. Soil organic matter, soil carbon, bulk density, pH and soil moisture were also measured for each site. Soil samples were collected from 64 total plots [22 subalpine,15 treeline and 27 alpine sites]. The subalpine site plots included aspen, fir, lodgepole, spruce and meadow vegetation cover. The treeline site plots included fir, spruce and meadow vegetation cover. The alpine site plots included dry meadow and mesic meadow fertilization (control, N, P, NP) plots. Soil cores were removed with 3.5-cm interior diameter PVC pipe that was driven into the soil by use of a rubber mallet. The minimum depth of individual cores was 10 cm. Cores were taken at each site three times over the period between 29 June 1999 and 29 July 1999.

Baseline soil inorganic and organic property data for Saddle snowfence, 1993.

Soil cores were collected during the construction of the 100+year snowfence on the Niwot Ridge Saddle in the autumn of 1993. Organic matter determinations were made on the samples in January 1994. The samples were also measured for total phosphorus, nitrogen, and carbon. These 1993 samples were representative of the baseline (pre-snowfence) soil conditions.

Krummholz island size, soil inorganic, and organic property data for Saddle, S slope of Niwot Ridge, 1994.

Engelmann spruce (Picea engelmannii) and subalpine fir (Abies lasiocarpa) tree islands modify the characteristics of surface soils in alpine tundra. Soil C content of the approximate A horizon (top 15 cm) of soil was measured during the summer of 1994 on windward, leeward, upslope and downslope sides, and interiors of tree islands on Niwot Ridge, Colorado, USA. A subset of samples from these sites were also used for CHN analysis and were measured for total phosphorus using persulfate digestions and colorimetric measurements. Results indicate significant (p<.0001) reductions of percent of dry mass represented by C in soil and significant (p<.04) declines in absolute C storage among soils on the windward sides of tree islands as compared to the upslope and downslope controls, and a tendency for reduced C on the leeward sides as well. Surface organic matter (O horizon) accumulations averaging 9.6 +/- 1.02 kg/m^2 are found in the interior of tree islands, but this material, in addition to roots, is not stabilized in the A horizons of soil. The movement of tree islands can therefore be regarded as disturbances to soil building processes in alpine tundra. Timberline forest and adjacent tundra patches of similar aspect and slope were also sampled for comparisons of soil C content. Results indicated similar C storage beneath trees and tundra at this lower elevation. The wind-induced movement of tree islands across the tundra creates enhanced snowpack within the trees and on their leeward sides. Shading and moisture conditions of the soil are altered, leading to C deposition and decomposition dynamics which differ from that of unimpacted tundra surface soils. However, at timberline, adjacent tundra lacks the ability to exhibit the enhanced C storage of alpine tundra at higher elevations. Snowpack within trees and adjacent tundra at timberline may be relatively constant such that biophysical factors affecting soil characteristics are relatively unchanged by plant life-form.

Biogeochemical rate data and sediment properties of samples used for a controlled flow through experiment testing the effect of nitrate on organic matter decomposition, PIE LTER, Plum Island Sound estuary, Massachusetts.

In this dataset, we used a controlled flow-through reactor (FTR) experiment to test the role of nitrate as an electron acceptor, and its effect on organic matter decomposition and the associated microbial community in salt marsh sediments. Organic matter decomposition significantly increased in response to nitrate, even at sediment depths typically considered resistant to decomposition. The use of isotope tracers suggests this pattern was largely driven by stimulated denitrification. Nitrate addition also significantly altered the microbial community and decreased alpha diversity, selecting for taxa belonging to groups known to reduce nitrate and oxidize more complex forms of organic matter. Fourier Transform-Infrared Spectroscopy further supported these results, suggesting that nitrate facilitated decomposition of complex organic matter compounds into more bioavailable forms. Taken together, these results suggest the existence of organic matter pools that only become accessible with nitrate and would otherwise remain stabilized in the sediment. The existence of such pools could have important implications for carbon storage, since greater decomposition rates as N loading increases may result in less overall burial of organic-rich sediment. Given the extent of nitrogen loading along our coastlines, it is imperative that we better understand the resilience of salt marsh systems to nutrient enrichment, especially if we hope to rely on salt marshes, and other blue carbon systems, for long-term carbon storage.

<p>Mesenchymal stem cells (MSCs) are well established to have promising therapeutic properties. TNF-stimulated gene-6 (TSG-6), a potent tissue-protective and anti-inflammatory factor, has been demonstrated to be responsible for a significant part of the tissue-protecting properties mediated by MSCs. Nevertheless, current knowledge about the biological function of TSG-6 in MSCs is limited. Here, we demonstrated that TSG-6 is a crucial factor that influences many functional properties of MSCs. The transcriptomic sequencing analysis of wild-type (WT) and TSG-6<sup>-/-</sup> -MSCs shows that the loss of TSG-6 expression leads to the perturbation of several transcription factors, cytokines, and other key biological pathways. TSG-6<sup>-/-</sup> -MSCs appeared morphologically different with dissimilar cytoskeleton organization, significantly reduced size of extracellular vesicles, decreased cell proliferative rate, and loss of differentiation abilities compared with the WT cells. These cellular effects may be due to TSG-6-mediated changes in the extracellular matrix (ECM) environment. The supplementation of ECM with exogenous TSG-6, in fact, rescued cell proliferation and changes in morphology. Importantly, TSG-6-deficient MSCs displayed an increased capacity to release interleukin-6 conferring pro-inflammatory and pro-tumorigenic properties to the MSCs. Overall, our data provide strong evidence that TSG-6 is crucial for the maintenance of stemness and other biological properties of murine MSCs.</p> <p>&nbsp;</p> <p>Some dataset of this research are in prism format, to ensure open access we attach a pdf instruction about this format and a link where downoladed it</p>

leaf anatomy, vascular traits and nanomechanical cell-wall properties in European beech provenances

<p>The file contains leaf anatomical data (thickness of individual leaf parenchyma layers), vascular traits of leaf midrib (vessel area and density and derived parameters), and nanomechanical properties of xylem cell walls (modulus of elasticity, adhesion, energy dissipation and deformation), which were studied in 15 provenances of European beech, originating from sites distributed across the whole range of the species. The trial plot (locality Tale in central Slovakia) was established in 1998 with 2-years-old seedling within the international provenance experiment with beech coordinated by the Institute of Forest Genetics of Thuenen Institute Grosshansdorf. Leaf anatomy was studied using light microscopy, while fluorescent microscopy was used to acquire vascular traits and atomic-force microscopy for nanomechanical cell-wall traits. Sun leaves were collected from 4 trees per provenances, 1 leaf per tree was analyzed. AFM was done in a subset of 8 provenances. The aim of the study was assessing geographical trends of the studied traits and their association with climate at the sites of origin to reveal potential adaptive variation patterns.</p>
