Data from: Comparing biocontrol and herbicide for managing an invasive non-native plant species: efficacy, non-target effects and secondary invasion

<p>1. Globally, invasive non-native plants are an increasing threat to indigenous biodiversity and ecosystems, but management can be compromised by poor efficacy of control methods, harmful non-target effects or secondary invasions by other non-native plant species.</p> <p>2. A 5-year field trial compared two stakeholder-selected control methods for heather, a European plant invading native ecosystems in and adjoining Tongariro National Park in New Zealand. The control methods were a selective herbicide (Pasture Kleen®; 2,4-D ester) and biocontrol with an introduced beetle Lochmaea suturalis (Coleoptera: Chrysomelidae).</p> <p>3. Biocontrol reduced mean heather cover by 97%, slightly more than herbicide at 87%, compared with a 20% increase in heather under no management.</p> <p>4. Cover of native dicots, the most species-rich plant group, increased following biocontrol. In contrast, herbicide application had major non-target effects on native dicots, reducing their percentage cover and species richness. Native monocot cover and species richness increased following both herbicide and biocontrol treatments.</p> <p>5. A similar 8-fold increase in non-native monocots occurred following both biocontrol and herbicide treatments. Overall, secondary invasion was greatest with biocontrol because non-native dicot cover also increased, whereas herbicide almost eliminated non-native dicots. 6. Synthesis and applications. Biocontrol and herbicide treatments both controlled heather but herbicide application was associated with severe non-target impacts on native dicots. Benefits to the native flora were consequently greatest in the biocontrol treatment, despite greater secondary invasion. Control strategies for management of widespread non-native plants to optimize ecosystem outcomes should include more consideration of biocontrol.</p>

Comparing three screen-based sedentary behaviours' effect upon adolescents' participation in physical activity: The ESSENS study

<p class="Arthurstyle1">Background: Literature focusing on the association between sedentary behaviours and physical activity has provided equivocal results and has been dominated by TV viewing as the indicator of sedentary behaviour. There is a need for more studies exploring the association between contemporary screen activities and physical activity among youth.</p> <p class="Arthurstyle1">Methods: A cross-sectional study including 742 adolescents was conducted in 2016. Data were collected at school through an online questionnaire. Regression analyses were used to explore the association between different screen-based sedentary behaviours and participation in physical activity.</p> <p class="Arthurstyle1">Results: The results showed that those with lower (vs higher) time spent on TV/movie streaming and electronic game playing both on weekdays and weekend days had significantly higher odds of participating in physical activity. There were no significant associations between socializing/surfing online both on weekdays and weekend days and physical activity in adjusted models.</p> <p class="Arthurstyle1">Conclusions: TV/movie streaming and electronic game playing during both weekdays and weekend days were significantly inversely related with participating in physical activity. Initiatives aimed at reducing screen-based sedentary activities might result in favourable effects on physical activity levels among adolescents.</p>

Data from: Comparative analyses of effective population size within and among species: ranid frogs as a case study

It has recently become practicable to estimate the effective sizes (Ne) of multiple populations within species. Such efforts are valuable for estimating Ne in evolutionary modeling and conservation planning. We used microsatellite loci to estimate Ne of 90 populations of four ranid frogs (20 to 26 populations per species, mean n per population = 29). Our objectives were to determine typical values of Ne for populations of each species, compare Ne estimates among the species, and test for correlations between several geographic variables and Ne within species. We used single-sample linkage disequilibrium, approximate Bayesian computation, and sibship assignment methods to estimate contemporary Ne for each population. Three of the species—Rana pretiosa, R. luteiventris, and R. cascadae— have consistently small effective population sizes (&lt;50). Ne in Lithobates pipiens spans a wider range, with some values in the hundreds or thousands. There is a strong east-to-west trend of decreasing Ne in Lithobates pipiens. The smaller effective sizes of western populations of this species may be related to habitat fragmentation and population bottlenecking.

Data from: Comparing ageing and the effects of diet supplementation in wild vs. captive antler flies, Protopiophila litigata

1) Few studies have simultaneously compared ageing within genetically similar populations in both laboratory and natural environments. Such comparisons are important for interpreting laboratory studies, because factors such as diet could affect ageing in environment-dependent ways. 2) Using a natural population of antler flies (Protopiophila litigata), we conducted separate factorial experiments in 2012 and 2013 that compared age-specific male survival and mating success in laboratory cages versus a natural field environment while supplementing their diets with protein or sugar. 3) We found consistent and substantial increases in both survival and mating rates in the laboratory compared to the field, but remarkably, despite these large differences actuarial ageing was only higher in the lab than in the field in 2012 and similar in the two environments in 2013. In both years, there was no difference between environments in reproductive ageing. 4) We found that males fed protein had a higher mortality rate than males fed sugar (strong and low support in 2012 and 2013 respectively). 5) In contrast, diet did not strongly impact average mating rates, actuarial ageing, or reproductive ageing in either experiment. 6) Our results provide the first evidence that the negative effect of protein on lifespan reported in many laboratory studies can also occur in wild populations, although perhaps less consistently. They also highlight how laboratory environments can influence life history traits and suggest caution when extrapolating from the laboratory to the field.

Data from: Effects of removing woody cover on long‐term population dynamics of a rare annual plant (Agalinis auriculata): a study comparing remnant prairie and oldfield habitats

1. Worldwide, grasslands are becoming shrublands/forests. In North America, eastern red cedar (Juniperus virginiana) often colonizes prairies. Habitat management can focus on woody removal, but we often lack long-term data on whether removal leads to population recovery of herbaceous plants without seeding. 2. We undertook a long-term study (17 years) of numbers of the rare annual plant Agalinis auriculata in a gridwork of 100 m2 plots in adjacent prairie and oldfield sites in Kansas, USA. We collected data before and after removal of Juniperus virginiana at the prairie. 3. Plant population sizes were highly variable at both sites and over time. High numbers of plants in a plot one year were often followed by low numbers the following year, suggesting negative density-dependence. Plant numbers were lowest with extensive woody cover and with low precipitation. After woody plant removal, A. auriculata increased dramatically in abundance and occupancy in most years; increases were also seen at the oldfield, suggesting later survey years were overall more favorable. 4. Synthesis and applications: Removal of woody plants led to increased numbers of a rare annual prairie plant, without seeding. Multiple years of data were essential for interpretation given extreme temporal variability in numbers. The largest prairie population was seven years following tree removal, showing that positive effects of management can last this long. This species also fared well in oldfield habitat, suggesting restoration opportunities. Given that land managers are busy, time-efficient field methods and data analysis approaches such as ours offer advantages. In addition to general linear models, we suggest Rank Occupancy Abundance Profiles (ROAPs), a simple-to-use data visualization and analysis method. Creation of ROAPs for sites before and after habitat management helps reveal the degree to which plant populations are responding to management with changes in local density, changes in occupancy, or both.

Comparing the strength of modular signal, and evaluating alternative modular hypotheses, using covariance ratio effect sizes with morphometric data

<p>The study of modularity is paramount for understanding trends of phenotypic evolution, and for determining the extent to which covariation patterns are conserved across taxa and levels of biological organization. However, biologists currently lack quantitative methods for statistically comparing the strength of modular signal across datasets, and a robust approach for evaluating alternative modular hypotheses for the same dataset. As a solution to these challenges, we propose an effect size measure (Z_CR) derived from the covariance ratio, and develop hypothesis-testing procedures for their comparison. Computer simulations demonstrate that Z_CR displays appropriate statistical properties and low levels of misspecification, implying that it correctly identifies modular signal, when present. By contrast, alternative methods based on likelihood (EMMLi) and goodness of fit (MINT) suffer from high false positive rates and high model misspecification rates. An empirical example in sigmodontine rodent mandibles is provided to illustrate the utility of Z_CR for comparing modular hypotheses. Overall, we find that covariance ratio effect sizes are useful for comparing patterns of modular signal across datasets or for evaluating alternative modular hypotheses for the same dataset. Finally, the statistical philosophy for pairwise model comparisons using effect sizes should accommodate any future analytical developments for characterizing modular signal.</p>

Data from: Effective population size of natural populations of Drosophila buzzatii, with a comparative evaluation of nine methods of estimation

Allozyme and microsatellite data from numerous populations of Drosophila buzzatii have been used (i) to determine to what degree Ne varies among generations within populations, and among populations, and (ii) to evaluate the congruence of four temporal and five single sample estimators of Ne. Effective size of different populations varied over two orders of magnitude, most populations are not temporally stable in genetic composition, and Ne showed large variation over generations in some populations. Short term Ne estimates from the temporal methods were highly correlated, but the smallest estimates were the most precise for all four methods, and the most consistent across methods. Except for one population, Ne estimates were lower when assuming gene flow than when assuming populations were closed. However, attempts to jointly estimate Ne and immigration rate were of little value because the source of migrants was unknown. Correlations among the estimates from the single sample methods generally were not significant although, as for the temporal methods, estimates were most consistent when they were small. These single sample estimates of current Ne are generally smaller than the short term temporal estimates. Nevertheless, population genetic variation is not being depleted, presumably due to past or ongoing migration. A clearer picture of current and short term effective population sizes will only follow with better knowledge of migration rates between populations. Different methods are not necessarily estimating the same Ne, they are subject to different bias, and the biology, demography and history of the population(s) may affect different estimators differently.

Bats generate lower affinity, but higher diversity antibody responses compared to mice, an effect that can be manipulated with diet

<p><span>Bats are reservoirs of many zoonotic viruses that are fatal in humans but do not cause disease in bats. Moreover, bats generate low neutralizing antibody titers in response to experimental viral infection, although more robust antibody responses have been observed in wild caught bats during times of food stress. Here we compared the antibody titers and B cell receptor (BCR) diversity of Jamaican fruit bats (<em>Artibeus jamaicensis</em>; JFB) and BALB/c mice generated in response to T-dependent and T-independent antigens. We then manipulated the diet of JFBs and challenged them with H18N11 influenza A-like virus or a replication incompetent Nipah virus VSV (Nipah-riVSV). Under standard housing conditions, JFBs generated a lower avidity antibody response and possessed more BCR mRNA diversity compared to BALB/c mice. However, withholding protein from JFBs improved serum neutralization in response to Nipah-riVSV and improved serum antibody titers specific to H18 but reduced BCR mRNA diversity. </span></p>

The cat as a naturally occurring model of renal interstitial fibrosis: Characterisation of primary feline proximal tubular epithelial cells and comparative pro-fibrotic effects of TGF-β1

<p>Chronic kidney disease (CKD) is common in both geriatric cats and aging humans, and is pathologically characterised by chronic tubulointerstitial inflammation and fibrosis in both species. Cats with CKD may represent a spontaneously occurring, non-rodent animal model of human disease, however little is known of feline renal cell biology. In other species, TGF-&beta;1 signalling in the proximal tubular epithelium is thought to play a key role in the initiation and progression of renal fibrosis. In this study, we first aimed to isolate and characterise feline proximal tubular epithelial cells (FPTEC), comparing them to human primary renal epithelial cells (HREC) and the human proximal tubular cell line HK-2. Secondly, we aimed to examine and compare the effect of human recombinant TGF-&beta;1 on cell proliferation, pro-apoptotic signalling and genes associated with epithelial-to-mesenchymal transition (EMT) in feline and human renal epithelial cells.</p> <p>FPTEC were successfully isolated from cadaverous feline renal tissue, and demonstrated a marker protein expression profile identical to that of HREC and HK-2. Exposure to TGF-&beta;1 (0-10 ng/ml) induced a concentration-dependent loss of epithelial morphology and alterations in gene expression consistent with the occurrence of partial EMT in &nbsp;all cell types. This was associated with transcription of downstream pro-fibrotic mediators, growth arrest in FPTEC and HREC (but not HK-2), and increased apoptotic signalling at high concentrations of TGF- &beta;1. These effects were inhibited by the ALK5 (TGF-&beta;1RI) antagonist SB431542 (5 &micro;M), suggesting they are mediated via the ALK5/TGF-&beta;1RII receptor complex. Taken together, these results suggest that TGF-&beta;1 may be involved in epithelial cell dedifferentiation, growth arrest and apoptosis in feline CKD as in human disease, and that cats may be a useful, naturally occurring model of human CKD.</p>

Dataset from The effect of visitors on stress-related behaviour of zoo-housed red-faced spider monkeys (Ateles Paniscus) compared in the inside and outside compartment of the enclosure

<p>Animals held in captivity often endure more stress than wild congeners do, which might be harmful for their health and wellbeing. This stress is often expressed by showing stereotypic behaviour. Previous studies showed that visitors can have a negative impact on the stress levels of zoo-housed animals, referred to by the term &lsquo;visitor effect&rsquo;. The visitor effect is found to be strong in arboreal primate species, such as spider monkeys.</p> <p>The current study examined the effect of visitors on the stress-related behaviour of red-faced spider monkeys (<em>Ateles Paniscus</em>) housed at ARTIS Amsterdam Royal Zoo. Two visitor variables were assessed; number of visitors and sound volume. Additionally, this study examined whether there was a difference in visitor effect in the inside and outside compartment of the enclosure since these exhibits differ strongly in design.</p> <p>Behavioural observations were conducted for 3 weeks daily, during which all behaviour displayed by the spider monkeys was noted, along with the number of visitors present, the level of sound (in decibel) and the location of observation.</p> <p>This study demonstrated that an increase in average level of decibel led to an increase in stereotypic behaviour. Additionally, spider monkeys were found to display more stereotypic behaviour in the outside compartment of their enclosure. However, this study did not find the number of visitors to have an influence on the stress-related behaviour. These results suggest that visitor noise can have a negative effect on the stress levels of red-faced spider monkeys and that this effect is dependent on the exhibit design.</p> <p>This study hopefully provides more insight into the visitor effect by assessing the influence of two important visitor variables. These insights might improve future designs of zoo enclosures, thus enhancing animal welfare. However, future research is needed to further assess the cause of stress in zoo-housed animals.&nbsp;</p>

Comparative Study of Field-Effect Transistors Based on Graphene Oxide and CVD Graphene in Highly Sensitive NT-proBNP Aptasensors

<p>Graphene-based materials are actively being investigated as sensing elements for the detection of different analytes. Both graphene grown by chemical vapor deposition (CVD) and graphene oxide (GO) produced by the modified Hummers&rsquo; method are actively used in the development of biosensors. The production costs of CVD graphene- and GO-based sensors are similar; however, the question remains regarding the most efficient graphene-based material for the construction of point-of-care diagnostic devices. To this end, in this work, we compare CVD graphene aptasensors with the aptasensors based on reduced GO (rGO) for their capabilities in the detection of NT-proBNP, which serves as the gold standard biomarker for heart failure. Both types of aptasensors were developed using commercial gold interdigitated electrodes (IDEs) with either CVD graphene or GO formed on top as a channel of liquid-gated field-effect transistor (FET), yielding GFET and rGO-FET sensors, respectively. The functional properties of the two types of aptasensors were compared. Both demonstrate good dynamic range from 10 fg/mL to 100 pg/mL. The limit of detection for NT-proBNP in artificial saliva was 100 fg/mL and 1 pg/mL for rGO-FET- and GFET-based aptasensors, respectively. While CVD GFET demonstrates less variations in parameters, higher sensitivity was demonstrated by the rGO-FET due to its higher roughness and larger bandgap. The demonstrated low cost and scalability of technology for both types of graphene-based aptasensors may be applicable for the development of different graphene-based biosensors for rapid, stable, on-site, and highly sensitive detection of diverse biochemical markers.</p>

Data from: A comparative study on genetic effects of artificial and natural habitat fragmentation on Loropetalum chinense (Hamamelidaceae) in Southeast China

Elucidating the demographic and landscape features that determine the genetic effects of habitat fragmentation has become fundamental to research in conservation and evolutionary biology. Land-bridge islands provide ideal study areas for investigating the genetic effects of habitat fragmentation at different temporal and spatial scales. In this context, we compared patterns of nuclear microsatellite variation between insular populations of a shrub of evergreen broad-leaved forest, Loropetalum chinense, from the artificially created Thousand-Island Lake (TIL) and the Holocene-dated Zhoushan Archipelago of Southeast China. Populations from the TIL region harboured higher levels of genetic diversity than those from the Zhoushan Archipelago, but these differences were not significant. There was no correlation between genetic diversity and most island features, excepting a negative effect of mainland–island distance on allelic richness and expected heterozygosity in the Zhoushan Archipelago. In general, levels of gene flow among island populations were moderate to high, and tests of alternative models of population history strongly favoured a gene flow-drift model over a pure drift model in each region. In sum, our results showed no obvious genetic effects of habitat fragmentation due to recent (artificial) or past (natural) island formation. Rather, they highlight the importance of gene flow (most likely via seed) in maintaining genetic variation and preventing inter-population differentiation in the face of habitat 'insularization' at different temporal and spatial scales.

Data from: Exposure to opposing temperature extremes causes comparable effects on Cardinium density but contrasting effects on Cardinium-induced cytoplasmic incompatibility

Terrestrial arthropods, including insects, commonly harbor maternally inherited intracellular symbionts that confer benefits to the host or manipulate host reproduction to favor infected female progeny. These symbionts may be especially vulnerable to thermal stress, potentially leading to destabilization of the symbiosis and imposing costs to the host. For example, increased temperatures can reduce the density of a common reproductive manipulator, Wolbachia, and the strength of its crossing incompatibility (cytoplasmic incompatibility, or CI) phenotype. Another manipulative symbiont, Cardinium hertigii, infects ~ 6-10% of Arthropods, and also can induce CI, but there is little homology between the molecular mechanisms of CI induced by Cardinium and Wolbachia. Here we investigated whether temperature disrupts the CI phenotype of Cardinium in a parasitic wasp host, Encarsia suzannae. We examined the effects of both warm (32°C day/ 29°C night) and cool (20°C day/ 17°C night) temperatures on Cardinium CI and found that both types of temperature stress modified aspects of this symbiosis. Warm temperatures reduced symbiont density, pupal developmental time, vertical transmission rate, and the strength of both CI modification and rescue. Cool temperatures also reduced symbiont density, however this resulted in stronger CI, likely due to cool temperatures prolonging the host pupal stage. The opposing effects of cool and warm-mediated reductions in symbiont density on the resulting CI phenotype indicates that CI strength may be independent of density in this system. Temperature stress also modified the CI phenotype only if it occurred during the pupal stage, highlighting the likely importance of this stage for CI induction in this symbiosis.

The relative effects of pace of life and habitat characteristics on the evolution of sexual ornaments: a comparative assessment

<p><span>Selection may favor greater investment into sexual ornaments when opportunities for future reproduction are limited (e.g., due to high adult mortality). However, a key driver of mortality, predation, typically selects against elaborate sexual ornaments. Here, we examine the evolution of sexual ornaments in a group of killifishes, which have marked contrasts in life-history strategy between species and inhabit environments that differ in their accessibility to aquatic predators. We first assessed if the size of sexual ornaments (unpaired fins) influenced swimming performance and found that larger fins negatively affected swimming performance. Second, we investigated whether the evolution of larger ornamental fins is driven primarily by the pace of life-history (i.e., investment into current vs future reproduction) or habitat type (as a proxy for predation risk). We found that males from species inhabiting ephemeral habitats with lower predation risk had both larger fins and more pronounced sexual dimorphism in fin size, compared to males from more accessible permanent habitats. Our results indicate that enlarged ornamental fins impair locomotion and evolve more frequently in environments that are less accessible to predators, but with no clear association to the pace of life-history. We provide a rare link between the evolution of sexual ornaments, decreases in locomotion, and natural selection on ornaments through vulnerability to predation.</span></p>

Assessing the comparative effects of storm-relative helicity components within right-moving supercell environments

<p>Supercell thunderstorms develop low-level rotation via tilting of environmental horizontal vorticity (ω<sub>h</sub>) by the updraft. This rotation induces dynamic lifting that can stretch near-surface vertical vorticity into a tornado. Low-level updraft rotation is generally thought to scale with 0–500 m storm-relative helicity (SRH): the combination of storm-relative flow, |SRF|, |ω<sub>h</sub>|, and cosφ (where φ is the angle between SRF and ω<sub>h</sub>). It is unclear how much influence each component of SRH has in intensifying the low-level mesocyclone. This study surveys these three components using self-organizing maps (SOMs) to distill 15,906 proximity soundings for observed right-moving supercells. Statistical analyses reveal the component most highly correlated to SRH and to streamwise vorticity (ω<sub>s</sub>) in the observed profiles is |ω<sub>h</sub>|. Furthermore, |ω<sub>h</sub>| and SRF are themselves highly correlated due to their shared dependence on the hodograph length. The representative profiles produced by the SOMs were combined with a common thermodynamic profile to initialize quasi-realistic supercells in a cloud model. The simulations reveal that, across a range of real-world profiles, intense low-level mesocyclones are most closely linked to ω<sub>h</sub> and SRF, while the angle between them appears to be mostly inconsequential.</p>
