Assessing microbiome population dynamics using wild-type isogenic standardized hybrid (WISH)-tags
<p><span>Microbiomes feature recurrent compositional structures under given environmental conditions. </span><span>However, these patterns may conceal diverse underlying population dynamics that require intra-strain resolution. Here, we developed a genomic tagging system, termed wild-type isogenic standardized hybrid (WISH)-tags, that can be combined with quantitative PCR and next-generation sequencing for microbial strain enumeration. We experimentally validated the performance of 62 tags and showed they can be differentiated with high precision. WISH-tags were introduced into model and non-model bacterial members of the mouse and plant microbiota. Intra-strain priority effects were tested using one species of isogenic barcoded bacteria in the murine gut and the <em>Arabidopsis</em> phyllosphere, both with and without microbiota context. We observed colonization resistance against late arriving strains of <em>Salmonella</em> typhimurium in the mouse gut, whereas the phyllosphere accommodated <em>Sphingomonas</em> latecomers in a proportional manner to their presence at the late inoculation timepoint. This demonstrates that WISH-tags are a resource for deciphering population dynamics underlying microbiome assembly across biological systems. </span></p>

<p><em>Obovaria olivaria</em> is a species of freshwater mussel native to the Mississippi River and Laurentian Great Lakes-St. Lawrence River drainages of North America. This mussel has experienced population declines across large parts of its distribution and is imperiled in many jurisdictions. <em>Obovaria olivaria </em>uses the similarly imperiled <em>Acipenser fulvescens</em> (Lake Sturgeon) as a host for its glochidia. We employed mitochondrial DNA sequencing and Restriction-site Associated DNA sequencing (RAD-seq) to assess patterns of genetic diversity and population structure of <em>O. olivaria</em> from 19 collection locations including the St. Lawrence River drainage, the Great Lakes drainage, the Upper Mississippi River drainage, the Ohioan River drainage and the Mississippi Embayment. Heterozygosity was highest in Upper Mississippi and Great Lakes populations, followed by a reduction in diversity and relative effective population size in the St. Lawrence populations. Pairwise <em>F</em><sub>ST</sub> ranged from 0.00 to 0.20, and analyses of genetic structure revealed two major ancestral populations, one including all St. Lawrence River/Ottawa River sites and the other including remaining sites; however, significant admixture and isolation by river distance across the range were evident. The genetic diversity and structure of <em>O. olivaria</em> is consistent with the existing literature on <em>Acipenser fulvescens</em> and suggest that, although northern and southern <em>O. olivaria</em> populations are genetically distinct, genetic structure in <em>O. olivaria</em> is largely clinal rather than discrete across its range. Conservation and restoration efforts of <em>O. olivaria</em> should prioritize the maintenance and restoration of locations where <em>O. olivaria </em>remain, especially in northern rivers, and to ensure connectivity that will facilitate dispersal of <em>Acipenser fulvescens</em> and movement of encysted glochidia.</p>

Geolocator tagging of east Siberian bluethroats
<p>Many long-distance migratory bird species seem to retain the winter quarters their ancestors used during the last glacial maximum. Post-glacial colonization following the retreat of the ice sheets have then resulted in apparent suboptimal migration routes, since the expanding populations have failed to use more nearby, alternative and climatically suitable, wintering areas. The bluethroat subspecies <em>Cyanecula svecica svecia </em>occurs in the northern areas of the Palearctic from the Atlantic to Alaska. Because it is monotypic and lacks population structure in mitochondrial DNA it is assumed to have colonized its present range from one glacial refuge population. Geolocator tracks from birds in its western range have shown that these migrates to the Indian subcontinent following a route west of the Himalaya. In the present study we fit geolocators of bluethroats breeding in their eastern range (Chukotka) to test whether they also use a route west of the Himalaya to the same wintering grounds as their European conspecifics, or shortcut east of the Himalaya. We retrieved one of the 30 bluethroats tagged which clearly showed that it migrated east of the Himalaya to a wintering area tentatively located in Myanmar. By assuming that this bird is representative for the migration of eastern <em>Cyanecula s. svecia</em>, we can reject our initial hypothesis of a common migration route of all <em>Cyanecula s. svecia</em>. The different routes and wintering areas of western and eastern <em>Cyanecula s. svecia</em> implicate the presence of a migratory divide somewhere north of the Himalaya, and further that the populations were in different refuge populations during the last glaciation.</p>

COURAGE - SwipeIt - Study 1 & 2 - SwipeIt-Tagging data
<p>We have designed a serious mini-game (the mobile app “SwipeIt”) that allows for a playful and controllable interaction with potentially toxic content items. <br> The app was designed to be used in a classroom context to support classroom discussions around example social media items. Given that we know that the classification of social media items regarding different types of discrimination and toxic content varies between individuals, we first wanted to capture such differences and make them available as a resource for classroom discussions. In addition to triggering classroom discussions, this collection of data will also serve as a resource for further analyses using machine learning techniques.</p> <p> </p> <p>In study 1, we provided discrimination, hate speech, cyberbullying (cybermobbing) and verbal violence as labels.<br> In study 2, we provided discrimination, hate speech, cyberbullying (cybermobbing) and sexism as labels.</p>
