3D EBSD Dataset of the Alpha and Beta Phase Orientations for a Hot-Rolled Model Zircaloy-4 with 7 wt.% Nb Alloy

<p>A set of serial-section&nbsp;electron backscatter diffraction (EBSD) data&nbsp;files and a&nbsp;3D reconstruction&nbsp;of&nbsp;a&nbsp;model&nbsp;Zircaloy-4 with 7 wt.% Nb addition&nbsp;alloy following hot-rolling.</p> <p>The 3D data set contains measurements of the material rolled at 725C to 75% reduction. The measurements&nbsp;include&nbsp;indexing of both the alpha and the beta phases in 441 sequential slices, each&nbsp;of 0.1&nbsp;&mu;m, through a small section of the material, taken using the&nbsp;dual beam Thermo Scientific Helios Xe<sup>+</sup>&nbsp;plasma focused ion-beam scanning electron microscope&nbsp;(PFIB-SEM). The 3D EBSD data set includes&nbsp;EBSD measurements&nbsp;in the form of ctf files, binary data files, an Aztec project file,&nbsp;and accompanying&nbsp;images for&nbsp;each slice.</p> <p>A 3D volume was&nbsp;reconstructed from the 3D EBSD data set&nbsp;using a customised&nbsp;pipeline within&nbsp;the DREAM.3D software. <p>Geothermal hot springs, widely distributed on Earth, are important geological green-house gas sources which contribute large amounts of CO<sub>2</sub> emission to the atmosphere. Exploring microbial carbon fixation in these springs is vital to fully understand the carbon budget in these terrestrial aquatic systems. In this study, carbon fixation rate by photosynthetic mats was determined with a <sup>13</sup>C labeling approach along a gradient of temperature (~69 - ~75 <sup>o</sup>C) in a geothermal pool at Rehai Park in Tengchong, Yunnan Province, China. Light experiments were performed to determine carbon fixation rate by both photosynthesis and chemosynthesis, while dark experiments were performed by chemosynthesis. The results showed that the highest carbon fixation rate (about 4,637 μg C/g TOC/h) was obtained in light at the lowest temperature (~69 <sup>o</sup>C) in the afternoon (1:00 to 6:00 pm), most of which (96.7%) was contributed by photosynthesis. The lowest carbon fixation rate (138 μg C/g TOC/h) was measured at the highest temperature (~75 <sup>o</sup>C), where chemosynthesis dominated over photosynthesis in carbon fixation. At these rates, the photosynthetic mats would fix 80% CO<sub>2</sub> emission during the daytime. At the end of daytime, the photosynthetic mats respired 34% -70% newly fixed organic carbon in one hour. Interestingly, both the carbon fixation rate and respiration rate decreased exponentially with increasing temperature, which may be ascribed to a temperature dependence of both microbial community composition and enzymatic activity. In summary, our study revealed that biological carbon fixation by photosynthetic mats is highly temperature dependent and significantly affects carbon cycling in hot springs.</p>

Data from: Prevalence of afebrile malaria and development of risk-scores for gradation of villages: a study from a hot-spot in Odisha

Introduction: Malaria is a public health emergency in India and Odisha. The national malaria elimination programme aims to expedite early identification, treatment and follow-up of malaria cases in hot-spots through a robust health system, besides focusing on efficient vector control. This study, a result of mass screening conducted in a hot-spot in Odisha, aimed to assess prevalence, identify and estimate the risks and develop a management tool for malaria elimination. Methods: Through a cross-sectional study and using WHO recommended Rapid Diagnostic Test (RDT), 13221 individuals were screened. Information about age, gender, education and health practices were collected along with blood sample (5 µl) for malaria testing. Altitude, forestation, availability of a village health worker and distance from secondary health center were captured using panel technique. A multi-level poisson regression model was used to analyze association between risk factors and prevalence of malaria, and to estimate risk scores. Results: The prevalence of malaria was 5.8% and afebrile malaria accounted for 79 percent of all confirmed cases. Higher proportion of Pv infections were afebrile (81%). We found the prevalence to be 1.38 (1.1664 - 1.6457) times higher in villages where the Accredited Social Health Activist (ASHA) didn't stay; the risk increased by 1.38 (1.0428 - 1.8272) and 1.92 (1.4428 - 2.5764) times in mid- and high-altitude tertiles. With regard to forest coverage, villages falling under mid- and highest-tertiles were 2.01 times (1.6194 - 2.5129) and 2.03 times (1.5477 - 2.6809), respectively, more likely affected by malaria. Similarly, villages of mid tertile and lowest tertile of education had 1.73 times (1.3392 - 2.2586) and 2.50 times (2.009 - 3.1244) higher prevalence of malaria. Conclusion: Presence of ASHA worker in villages, altitude, forestation, and education emerged as principal predictors of malaria infection in the study area. An easy-to-use risk-scoring system for ranking villages based on these risk factors could facilitate resource prioritization for malaria elimination.

Data from: Hot spring frogs (Buergeria japonica) prefer cooler water to hot water

<p>"Hot spring frog" is an informal name used for the Japanese stream tree frog (<i>Buergeria japonica</i>), which is widely distributed in Taiwan and the Ryukyu Archipelago in Japan. Some populations of the species are known to inhabit hot springs. However, water temperature can be extremely high around the sources of hot springs. Thus, it is questionable whether <i>B. japonica</i> selectively inhabits such dangerous environments. To address this question, we conducted a series of observations of water temperature preferences of a hot spring population of <i>B. japonica</i> in Kuchinoshima Island in Japan: (1) a field observation of tadpole density in water pools of different temperatures, (2) a field observation of water temperatures where adult males appear for breeding, and (3) an indoor observation of water temperatures selected by adult females for oviposition. As a result, tadpoles showed a higher density in cooler water. Adult males avoided water pools hotter than 37 °C, and adult females selected cooler pools for oviposition. Camera records also showed that adult individuals tend to appear around cooler pools. Thus, we did not find any support for the hypothesis that hot spring frogs prefer hot water. Conversely, they apparently tended to prefer cooler water if it was available. Water temperatures around the sources of the hot spring exceed thermal tolerances of the species and could be a strong selective pressure on the population. Thus, the ability to sense and avoid lethal temperatures may be a key ecological and physiological characteristic for the species that inhabit hot springs.</p>

Data from: Night warming on hot days produces novel impacts on development, survival and reproduction in a small arthropod

1. An asymmetric increase of nighttime temperatures on hot days is one of the main features of global climate change. But the biological effects of an increased nighttime temperature combined with high daytime temperature are unclear. 2. We used six thermal regimens to simulate nighttime temperatures on hot days and investigated the effects of night warming on life history traits of the English grain aphid Sitobion avenae. Experimental temperatures fluctuated in continuous diurnal cycles, increasing from 27 °C to a maximum 35 °C and then declining to 27 °C gradually before further dropping to different minima (13, 16, 19, 21, 23 or 25 °C) representing nighttime temperatures. 3. When compared to expectations based on constant temperatures, night warming raised the optimum temperature for development by 3 °C, in contrast to results from experiments where temperature variability was altered symmetrically or in a parallel manner. Night warming also reduced aphid survival under heat from 75% to 37%, and depressed adult performance by up to 50%. Overall, night warming exacerbated the detrimental effects of hot days on the intrinsic rate of population increase, which was predicted to drop by 30% when nighttime minimum temperatures exceeded 20 °C. 4. Our novel findings on development challenge the 'Kaufmann effect', suggesting this is inapplicable to night warming likely to be encountered in nature. Although many average temperature models predict increasing pest outbreaks, our results suggest that outbreaks of some species might decrease due to the effects of night warming on population dynamics.

Data from: A multispecies approach reveals hot-spots and cold-spots of diversity and connectivity in invertebrate species with contrasting dispersal modes

Genetic diversity is crucial for species' maintenance and persistence, yet is often overlooked in conservation studies. Species diversity is more often reported due to practical constraints, but it is unknown if these measures of diversity are correlated. In marine invertebrates, adults are often sessile or sedentary and populations exchange genes via dispersal of gametes and larvae. Species with a larval period are expected to have more connected populations than those without larval dispersal. We assessed the relationship between measures of species and genetic diversity, and between dispersal ability and connectivity. We compiled data on genetic patterns and life history traits in nine species across five phyla. Sampling sites spanned 600 km in the northwest Mediterranean Sea and focused on a 50 km area near Marseilles, France. Comparative population genetic approaches yielded three main results. (1) Species without larvae showed higher levels of genetic structure than species with free-living larvae but the role of larval type (lecithotrophic or planktotrophic) was negligible. (2) A narrow area around Marseilles, subject to offshore advection, limited genetic connectivity in most species. (3) We identified sites with significant positive contributions to overall genetic diversity across all species, corresponding with areas near low human population densities. In contrast, high levels of human activity corresponded with a negative contribution to overall genetic diversity. Genetic diversity within species was positively and significantly linearly related with local species diversity. Our study suggests that local contribution to overall genetic diversity should be taken into account for future conservation strategies.

Data from: Subtype diversity and reassortment potential for co-circulating avian influenza viruses at a diversity hot spot

1. Biological diversity has long been used to measure ecological health. While evidence exists from many ecosystems that declines in host biodiversity may lead to greater risk of disease emergence, the role of pathogen diversity in the emergence process remains poorly understood. Particularly, because a more diverse pool of pathogen types provides more ways in which evolutionary innovations may arise, we suggest that host–pathogen systems with high pathogen diversity are more prone to disease emergence than systems with relatively homogeneous pathogen communities. We call this prediction the diversity-emergence hypothesis. 2. To show how this hypothesis could be tested, we studied a system comprised of North American shorebirds and their associated low-pathogenicity avian influenza (LPAI) viruses. These viruses are important as a potential source of genetic innovations in influenza. A theoretical contribution of this study is an expression predicting the rate of viral subtype reassortment to be proportional to both prevalence and Simpson's Index, a formula that has been used traditionally to quantify biodiversity. We then estimated prevalence and subtype diversity in host species at Delaware Bay, a North American AIV hotspot, and used our model to extrapolate from these data. 3. We estimated that 4 to 39 virus subtypes circulated at Delaware Bay each year between 2000 and 2008, and that surveillance coverage (percentage of co-circulating subtypes collected) at Delaware Bay is only about 63·0%. Simpson's Index in the same period varied more than fourfold from 0·22 to 0·93. These measurements together with the model provide an indirect, model-based estimate of the reassortment rate. A proper test of the diversity-emergence hypothesis would require these results to be joined to independent and reliable estimates of reassortment, perhaps obtained through molecular surveillance. 4. These results suggest both that subtype diversity (and therefore reassortment) varies from year to year and that several subtypes contributing to reassortment are going undetected. The similarity between these results and more detailed studies of one host, ruddy turnstone (Arenaria interpres), further suggests that this species may be the primary host for influenza reassortment at Delaware Bay. 5. Biological diversity has long been quantified using Simpson's Index. Our model links this formula to a mechanistic account of reassortment in multipathogen systems in the form of subtype diversity at Delaware Bay, USA. As a theory of how pathogen diversity may influence the evolution of novel pathogens, this work is a contribution to the larger project of understanding the connections between biodiversity and disease.

Data from: Hot dogs: high ambient temperatures impact reproductive success in a tropical carnivore

Climate change imposes an urgent need to recognise and conserve the species likely to be worst affected. However, while ecologists have mostly explored indirect effects of rising ambient temperatures on temperate and polar species, physiologists have predicted direct impacts on tropical species. The African wild dog (Lycaon pictus), a tropical species, exhibits few of the traits typically used to predict climate change vulnerability. Nevertheless, we predicted that wild dog populations might be sensitive to weather conditions, because the species shows strongly seasonal reproduction across most of its geographical range. We explored associations between weather conditions, reproductive costs, and reproductive success, drawing on long-term wild dog monitoring data from sites in Botswana (20°S, 24 years), Kenya (0°N, 12 years), and Zimbabwe (20°S, 6 years). High ambient temperatures were associated with reduced foraging time, especially during the energetically costly pup-rearing period. Across all three sites, packs which reared pups at high ambient temperatures produced fewer recruits than did those rearing pups in cooler weather; at the non-seasonal Kenya site such packs also had longer inter-birth intervals. Over time, rising ambient temperatures at the (longest-monitored) Botswana site coincided with falling wild dog recruitment. Our findings suggest a direct impact of high ambient temperatures on African wild dog demography, indicating that this species, which is already globally endangered, may be highly vulnerable to climate change. This vulnerability would have been missed by simplistic trait-based assessments, highlighting the limitations of such assessments. Seasonal reproduction, which is less common at low latitudes than at higher latitudes, might be a useful indicator of climate change vulnerability among tropical species.

Data from: Eco-morphological differentiation in Lake Magadi tilapia, an extremophile cichlid fish living in hot, alkaline and hypersaline lakes in East Africa

Ecological diversification through divergent selection is thought to be a major force during the process of adaptive radiations. However, the large sizes and complexity of most radiations such as those of the cichlids in the African Great Lakes make it impossible to infer the exact evolutionary history of any population divergence event. The genus Alcolapia, a small cichlid lineage endemic to Lakes Magadi and Natron in East Africa, exhibits phenotypes similar to some of those found in cichlids of the radiations of the African Great Lakes. The simplicity within Alcolapia makes it an excellent model system to investigate ecological diversification and speciation. We used an integrated approach including population genomics based on RAD-seq data, geometric morphometrics, and stable isotope analyses to investigate the eco-morphological diversification of tilapia in Lake Magadi and its satellite lake Little Magadi. Additionally, we reconstructed the demographic history of the species using coalescent simulations based on the joint site frequency spectrum. The population in Little Magadi has a characteristically upturned mouth - possibly an adaptation to feeding on prey from the water surface. Eco-morphological differences between populations within Lake Magadi are more subtle, but are consistent with known ecological differences between its lagoons such as high concentrations of nitrogen attributable to extensive guano deposits in Rest of Magadi relative to Fish Springs Lagoon. All populations diverged simultaneously only about 1,100 generations ago. Differences in levels of gene flow between populations and the effective population sizes have likely resulted in the inferred heterogeneous patterns of genome-wide differentiation.

Data from: A novel case of autogamy and cleistogamy in Dendrobium wangliangii: a rare orchid distributed in the dry-hot valley

Dendrobium wangliangii is an epiphytic orchid distributed in the Jinshajiang dry-hot valley in Luquan County, Yunnan Province, China. Most Dendrobium spp. typically have a low fruit set, but this orchid shows a higher fruit set under natural conditions despite the lack of effective pollinators. The pollination biology of the critically endangered D. wangliangii was investigated in this study. A fruit set rate of 33.33±4.71% was observed after bagging treatment in 2017 and a high fruit set rate (65.72±4.44% in 2011; 50.79±5.44% in 2017) was observed under natural conditions, indicating that D. wangliangii is characterized by spontaneous self-pollination. The anther cap prevented the growing pollinium, thus the pollinium slid down and reached the stigmatic cavity, leading to autogamous self-pollination. Specifically, 51.50% of 162 unopened flowers (total 257 flowers) of this Dendrobium species under extreme water-deficit conditions developed into fruits, suggesting the presence of cleistogamy in D. wangliangii. Here, cleistogamy may represent the primary mode of pollination for this orchid. Spontaneous self-pollination and specific cleistogamous autogamy could represent major adaptions to the drought and pollinator-scarce habitat in the Jinshajiang dry-hot valley.

CAT# HHU2-734 The Hot Springs site is a massive village on the shore of Port Moller, on the Alaska Peninsula side of the southern Bering Sea. It was excavated by several different teams over the last 100 years. The main occupations are from 2000 BCE-1000 BCE, and from 100 CE to 800 CE. The Hot Springs artifacts are presented as a result of the research conducted under grants NSF 0137756, NSF 1204020, NSF 1139266, and NSF 1321411. H. Maschner, Principal Investigator.
