Data from: Are thyroid hormones mediators of incubation temperature-induced phenotypes in birds?

Incubation temperature influences a suite of traits in avian offspring. However, the mechanisms underlying expression of these phenotypes are unknown. Given the importance of thyroid hormones in orchestrating developmental processes, we hypothesized that they may act as an upstream mechanism mediating the effects of temperature on hatchling phenotypic traits such as reduced growth and thermoregulation. We found that plasma T3, but not T4 concentrations, differed among newly-hatched wood ducks (Aix sponsa) from different embryonic incubation temperatures. T4 at hatching correlated with time spent hatching, and T3 correlated with hatchling body condition, tarsus length, time spent hatching, and incubation period. In addition, the T3:T4 ratio differed among incubation temperatures up at hatch. Our findings are consistent with the hypothesis that incubation temperature modulates plasma thyroid hormones which in turn influences multiple aspects of duckling phenotype.

Incubation temperature and physiological aging in the zebra finch

<p>In birds, incubation temperature has received increased attention as an important source of phenotypic variability in offspring. A lower than optimal incubation temperature may negatively affect aspects of nestling physiology, such as body growth and energy metabolism. However, the long-term effects of sub-optimal incubation temperature on morphology and physiology are not well understood. In a previous study, we showed that zebra finches from eggs incubated at a low temperature (35.9°C) for 2/3 of the total incubation time suffered a lower post-fledging survival compared to individuals that had been incubated at higher temperatures (37.0 and 37.9°C). In the present study, we investigated whether these variations in incubation temperature could cause permanent long-lasting differences in body mass, body size, or basal metabolic rate. Furthermore, we tested whether the observed differences in survival between treatment groups would be reflected in the rate of physiological deterioration, assessed through oxidative damage and decreased metabolic rate with age (i.e. 'metabolic aging'). Incubation temperature did not significantly affect embryonic or nestling body growth and did not influence final adult body mass or body size. Nor was there any long-term effect on basal metabolic rate. Birds from eggs incubated at the lowest temperature experienced an accumulation of oxidative damage with age, although this was not accompanied by an accelerated rate of metabolic aging. The present results suggest that the low survival in these birds was possibly mediated by increased oxidative stress, but independent of body growth and the basal metabolic rate.</p>

Data from: Plasticity of snowy plover incubation behaviors in response to risks of nest predation

<p>Nest predation influences population dynamics and is thought to exert strong selection on the evolution of avian life history. Because parental behaviors can attract the attention of nest predators, incubating birds are predicted to decrease conspicuous behaviors at the nest-site and increase incubation constancy when risks of nest predation are high. We examined whether snowy plovers Charadrius nivosus responded to predator-specific risks of nest predation, using the number of off bouts and daily nest attendance (proportion of time spent incubating) as responses. We quantified risks using predator-specific hazard rates of nest mortality, which varied daily and were based on habitat characteristics at each nest. We assessed the influence of predator-specific risks of nest mortality on incubation behaviors using an individual-centering approach, allowing us to explain variation in incubation behaviors within and among breeding pairs. We found increased number of off bouts and nest attendance within breeding pairs in response to increasing risks of nest predation by foxes (Vulpes spp.) and gulls (Larus spp.), but not coyotes (Canis latrans) and common ravens (Corvus corax). Among breeding pairs across habitats, we found increased nest attendance in response to higher risks of nest predation by foxes, but not coyotes, gulls, or ravens. Breeding pairs differed in the amount of behavioral plasticity they exhibited in response to risks of nest predation. Our results suggest that risks of nest predation differentially influence behavioral responses of snowy plovers depending on the predator species, and the amount of behavioral plasticity may depend on characteristics of breeding adults.</p>

Data from: Incubator-independent cell-culture perfusion platform for continuous long-term microelectrode array electrophysiology and time-lapse imaging

Most in vitro electrophysiology studies extract information and draw conclusions from representative, temporally limited snapshot experiments. This approach bears the risk of missing decisive moments that may make a difference in our understanding of physiological events. This feasibility study presents a simple benchtop cell-culture perfusion system adapted to commercial microelectrode arrays (MEAs), multichannel electrophysiology equipment and common inverted microscopy stages for simultaneous and uninterrupted extracellular electrophysiology and time-lapse imaging at ambient CO2 levels. The concept relies on a transparent, replica-casted polydimethylsiloxane perfusion cap, gravity- or syringe-pump-driven perfusion and preconditioning of pH-buffered serum-free cell-culture medium to ambient CO2 levels at physiological temperatures. The low-cost microfluidic in vitro enabling platform, which allows us to image cultures immediately after cell plating, is easy to reproduce and is adaptable to the geometries of different cell-culture containers. It permits the continuous and simultaneous multimodal long-term acquisition or manipulation of optical and electrophysiological parameter sets, thereby considerably widening the range of experimental possibilities. Two exemplary proof-of-concept long-term MEA studies on hippocampal networks illustrate system performance. Continuous extracellular recordings over a period of up to 70 days revealed details on both sudden and gradual neural activity changes in maturing cell ensembles with large intra-day fluctuations. Correlated time-lapse imaging unveiled rather static macroscopic network architectures with previously unreported local morphological oscillations on the timescale of minutes.

Advanced in vitro hemocompatibility assessment of biomaterials using a new flow incubation system

<p>We present a new setup that enables standardized whole blood incubation of biomedical materials under flow. A blood volume of 2 ml is recirculated over test surfaces in a custom-made parallel plate incubation system to determine the activation of hemostasis and inflammation. Controlled physiological shear rates between 125 s<sup>-1</sup> and 1,250 s<sup>-1</sup> and minimized contact to air are combined with a natural-like pumping process. A unique feature of this setup allows tracing adhesion of blood cells to test surfaces microscopically <em>in situ</em>. Validation testing was performed in comparison to previously applied whole blood incubation methodologies.</p>
