Development and validation of a machine learning model for use as an automated artificial intelligence tool to predict mortality risk in patients with COVID-19

Background

New York City quickly became an epicenter of the COVID-19 pandemic. Due to a sudden and massive increase in patients during COVID-19 pandemic, healthcare providers incurred an exponential increase in workload which created a strain on the staff and limited resources. As this is a new infection, predictors of morbidity and mortality are not well characterized.

Methods

We developed a prediction model to predict patients at risk for mortality using only laboratory, vital and demographic information readily available in the electronic health record on more than 3000 hospital admissions with COVID-19. A variable importance algorithm was used for interpretability and understanding of performance and predictors.

Findings

We built a model with 84-97% accuracy to identify predictors and patients with high risk of mortality, and developed an automated artificial intelligence (AI) notification tool that does not require manual calculation by the busy clinician. Oximetry, respirations, blood urea nitrogen, lymphocyte percent, calcium, troponin and neutrophil percentage were important features and key ranges were identified that contributed to a 50% increase in patients’ mortality prediction score. With an increasing negative predictive value (NPV) starting 0.90 after the second day of admission, we are able more confidently able identify likely survivors. This study serves as a use case of a model with visualizations to aide clinicians with a better understanding of the model and predictors of mortality. Additionally, an example of the operationalization of the model via an AI notification tool is illustrated.

On The Development of Multidimensional Progenitor Models For Core-collapse Supernovae

Eight multi-dimensional core-collapse supernova progenitor models from work, 'On The Development of Multidimensional Progenitor Models For Core-collapse Supernovae'. Works that utilize these progenitor models are required to cite article. All models are given at 424 seconds, less than a second before iron core-collapse. The 3D 4pi models were evolved to 424.31 seconds, the collapse time according to the 1D MESA model. All data are in FLASH4 format using the HDF5 data structure.

3D models of mouse embryonic development (stl, converted from EMAP)

A collection of readily-usable volumetric models for all tissues and stages of mouse peri-implantation development as extracted from the eMouse Atlas Project (E5.0 to E9.0), as well as custom-made models of all pre-implantation stages (E0 to E4.0). These models have been converted to a commonly used 3D format (.stl), and are provided in ready-made files for digital exploration and illustration.

Specifically:

individual .stl models of embryonic subcomponents for each developmental stage (e.g. epiblast, visceral endoderm; 1 folder per EMAP embryonic stage)

ready-made .blend files where such components have been reassembled as a full embryo model, and in a scene with pre-prepared light sources and aligned camera (1 file per embryonic stage)

a .blend file with all embryonic models provided arranged in a temporal lineup; and a .blend file with a "starter-pack" of pre-made materials to use for easy illustration.

Also included are two data tables:

"EMAP_key.xlsx" indicating the source of its model, and the embryonic stage corresponding to each Theiler Stage

"EMAP_partslist.xlsx" containing the original name of each embryonic component provided (as used in EMAP), and the corresponding name used in this dataset.

Raw data for the article: Two Sides of The Same Coin: Normal and Tumoral Stem Cells, The Relevance of In Vitro Models and Therapeutic Approaches: The Experience with Zika Virus in Nervous System Development and Glioblastoma Treatment

Neural stem cells (NSCs) were described for the first time more than two decades ago for their ability to differentiate into all neural cell lineages. The isolation of NSCs from adults and embryos was carried out by various laboratories and in different species, from mice to humans. Similarly, no more than two decades ago, cancer stem cells were described. Cancer stem cells, previously identified in hematological malignancies, have now been isolated from several solid tumors (breast, brain, and gastrointestinal compartment). Though the origin of these cells is still unknown, there is a wide consensus about their role in tumor onset, propagation and, in particular, resistance to treatments. Normal and neoplastic neural stem cells share common characteristics, and can thus be considered as two sides of the same coin. This is particularly true in the case of the Zika virus (ZIKV), which has been described as an inhibitor of neural development by specifically targeting NSCs. This understanding prompted us and other groups to evaluate ZIKV action in glioblastoma stem cells (GSCs). The results indicate an oncolytic activity of this virus vs. GSCs, opening potentially new possibilities in glioblastoma treatment.
