Data from: Genetic architecture, biochemical underpinnings, and ecological impact of floral UV patterning

Floral attraction traits can significantly affect pollinator visitation patterns, but adaptive evolution of these traits may be constrained by correlations with other traits. In some cases, molecular pathways contributing to floral attraction are well-characterized, offering the opportunity to explore loci potentially underlying variation among individuals. Here, we quantify the range of variation in floral UV patterning (i.e., UV "bulls-eye" nectar guides) among crop and wild accessions of Brassica rapa. We then use experimental crosses to examine the genetic architecture, candidate loci, and biochemical underpinnings of this patterning as well as phenotypic manipulations to test the ecological impact. We find qualitative variation in UV patterning between wild (commonly lacking UV patterns) and crop (commonly exhibiting UV patterns) accessions. Similar to the majority of crops, recombinant inbred lines (RILs) derived from an oilseed crop × WI fast-plant® cross exhibit UV patterns, the size of which varies extensively among genotypes. In RILs, we further observe strong statistical-genetic and QTL correlations within petal morphological traits and within measurements of petal UV-patterning; however, correlations between morphology and UV-patterning are weak or non-significant, suggesting that UV patterning is regulated and may evolve independently of overall petal size. HPLC analyses reveal a high concentration of sinapoyl glucose in UV-absorbing petal regions, which, in concert with physical locations of UV-trait QTLs, suggest a regulatory and structural gene as candidates underlying observed quantitative variation. Finally, insects prefer flowers with UV bulls-eye patterns over those that lack patterns, validating the importance of UV-patterning in pollen-limited populations of B. rapa.

Data from: Evolution of long-term coloration trends with biochemically unstable ingredients

The evolutionarily persistent and widespread use of carotenoid pigments in animal coloration contrasts with their biochemical instability. Consequently, evolution of carotenoid-based displays should include mechanisms to accommodate or limit pigment degradation. In birds, this could involve two strategies: (i) evolution of a moult immediately prior to the mating season, enabling the use of particularly fast-degrading carotenoids and (ii) evolution of the ability to stabilize dietary carotenoids through metabolic modification or association with feather keratins. Here, we examine evolutionary lability and transitions between the two strategies across 126 species of birds. We report that species that express mostly unmodified, fast-degrading, carotenoids have pre-breeding moults, and a particularly short time between carotenoid deposition and the subsequent breeding season. Species that expressed mostly slow-degrading carotenoids in their plumage accomplished this through increased metabolic modification of dietary carotenoids, and the selective expression of these slow-degrading compounds. In these species, the timing of moult was not associated with carotenoid composition of plumage displays. Using repeated samples from individuals of one species, we found that metabolic modification of dietary carotenoids significantly slowed their degradation between moult and breeding season. Thus, the most complex and colourful ornamentation is likely the most biochemically stable in birds, and depends less on ecological factors, such as moult timing and migration tendency. We suggest that coevolution of metabolic modification, selective expression and biochemical stability of plumage carotenoids enables the use of unstable pigments in long-term evolutionary trends in plumage coloration.

Data from: Biochemical and functional analysis of Cyanobacterium Oscillatoria sp. LPS on human monocytes

Cyanobacterial blooms are an increasing source of environmental toxins that affect both human and animals. After ingestion of cyanobacteria, such as Geitlerinema sp., toxins and lipopopolysaccharide (LPS) from this organism induce fever, gastro-intestinal illness, and even death. However, little is known regarding the effects of cyanobacterial LPS on human monocytes after exposure to LPS upon ingestion. Based on our previous data using Geitlerinema sp. LPS (which was previously named Oscillatoria sp., a genus belonging to the same order as Geitlerinema), we hypothesized that Geitlerinema sp. LPS would activate human monocytes to proliferate, phagocytose particles and produce cytokines that are critical for promoting pro-inflammatory responses in the gut. Our data demonstrate that Geitlerinema sp. LPS induces monocyte proliferation and TNF-α, IL-1, and IL-6 production at high concentrations. In contrast, Geitlerinema sp. LPS is equally capable of inducing monocyte-mediated phagocytosis of FITC-Latex beads when compared to E. coli LPS, which was used as a positive control for our experiments. In order to understand the mechanism responsible for the difference in efficacy between Geitlerinema sp. LPS and E. coli LPS, we performed biochemical analysis and identified that Geitlerinema sp. LPS is comprised of significantly different sugars and fatty-acid side chains in comparison to E. coli LPS. The lipid A portion of Geitlerinema sp. LPS contains longer fatty acid side chains, such as C15:0, C16:0, and C18:0, instead of C12:0 found in E. coli LPS which may explain the decreased efficacy and toxicity of Geitlerinema sp. LPS in comparison to E. coli LPS.

Data from: A multiscale vibrational spectroscopic approach for identification and biochemical characterization of pollen

Background: Analysis of pollen grains reveals valuable information on biology, ecology, forensics, climate change, insect migration, food sources and aeroallergens. Vibrational (infrared and Raman) spectroscopies offer chemical characterization of pollen via identifiable spectral features without any sample pretreatment. We have compared the level of chemical information that can be obtained by different multiscale vibrational spectroscopic techniques. Methodology: Pollen from 15 different species of Pinales (conifers) were measured by seven infrared and Raman methodologies. In order to obtain infrared spectra, both reflectance and transmission measurements were performed on ground and intact pollen grains (bulk measurements), in addition, infrared spectra were obtained by microspectroscopy of multigrain and single pollen grain measurements. For Raman microspectroscopy measurements, spectra were obtained from the same pollen grains by focusing two different substructures of pollen grain. The spectral data from the seven methodologies were integrated into one data model by the Consensus Principal Component Analysis, in order to obtain the relations between the molecular signatures traced by different techniques. Results: The vibrational spectroscopy enabled biochemical characterization of pollen and detection of phylogenetic variation. The spectral differences were clearly connected to specific chemical constituents, such as lipids, carbohydrates, carotenoids and sporopollenins. The extensive differences between pollen of Cedrus and the rest of Pinaceae family were unambiguously connected with molecular composition of sporopollenins in pollen grain wall, while pollen of Picea has apparently higher concentration of carotenoids than the rest of the family. It is shown that vibrational methodologies have great potential for systematic collection of data on ecosystems and that the obtained phylogenetic variation can be well explained by the biochemical composition of pollen. Out of the seven tested methodologies, the best taxonomical differentiation of pollen was obtained by infrared measurements on bulk samples, as well as by Raman microspectroscopy measurements of the corpus region of the pollen grain. Raman microspectroscopy measurements indicate that measurement area, as well as the depth of focus, can have crucial influence on the obtained data.

DISTINCTIONS BETWEEN BIOCHEMICAL INDICATORS OF DIFFERENT BLOOD GROUPS

<p>The purpose of this work is to evaluate the differences in some biochemical parameters in blood plasma of blood groups I (0), II (A), III (B) and IV (AB). Biochemical indicators were determined on a Humastar100 (Germany) biochemical analyzer. The obtained results showed that there are certain statistically reliable differences between the amounts of biochemical indicators such as albumin, glucose, cholesterol, triglyceride and urea in blood plasmas of different blood groups.</p>

Berberine protects against sepsis-related acute lung injury in rats via the upregulation of the PPAR - Υ signaling pathway, with amelioration at the cellular level - A functional, immune-histological and biochemical study

<p>Aim: To assess the therapeutic effects of berberine in experimentally induced lung sepsis.  Examining its effects on selected cytokines, gene and protein expression and the histopathological findings.</p> <p>Results: Berberine significantly reduced the wet/dry lung ratio, the BALF protein, cell, and neutrophils percentage &amp; the BALF cytokines levels. In addition, pretreatment with berberine decreases the myeloperoxidase and MDA levels and decreases gene expression of NFKB, MCP -1 and the ICAM - 1 by RT - PCR analysis. Thus, suggesting an antioxidant and anti-inflammatory mode of the action. Western blot analysis revealed increased PPAR – Υ expression in the berberine pretreated group compared to the CLP group. Histopathological examination revealed improved histopathological parameters, in the berberine pretreated group, compared to the CLP group.</p> <p>Conclusion: Berberine improves the outcome in sepsis via antioxidant and anti-inflammatory effects, modulation of cytokine levels, upregulation of PPAR - Υ protein expression, and histopathological amelioration.</p>
