Organochemical Characterization of Peat Reveals Decomposition of Specific Hemicellulose Structures as the Main Cause of Organic Matter Loss in the Acrotelm
<p>Peatlands store carbon in the form of dead organic residues. Climate change and human impact impose risks on the sustainability of the peatlands carbon balance due to increased peat decomposition. Here, we investigated molecular changes in the upper peat layers (0–40 cm), inferred from high-resolution vertical depth profiles, from a boreal peatland using two-dimensional <sup>1</sup>H–<sup>13</sup>C nuclear magnetic resonance (NMR) spectroscopy, and comparison to δ<sup>13</sup>C, δ<sup>15</sup>N, and carbon and nitrogen content. Effects of hydrological conditions were investigated at respective sites: natural moist, drainage ditch, and natural dry. The molecular characterization revealed preferential degradation of specific side-chain linkages of xylan-type hemicelluloses within 0–14 cm at all sites, indicating organic matter losses up to 25%. In contrast, the xylan backbone, galactomannan-type hemicelluloses, and cellulose were more resistant to degradation and accumulated at the natural moist and drainage site. δ<sup>13</sup>C, δ<sup>15</sup>N, and carbon and nitrogen content did not correlate with specific hemicellulose structures but reflected changes in total carbohydrates. Our analysis provides novel insights into peat carbohydrate decomposition and indicates substantial organic matter losses in the acrotelm due to the degradation of specific hemicellulose structures. This suggests that variations in hemicellulose content and structure influence peat stability, which may have important implications with respect to climate change.</p>

Advanced Laser Fluorescence (ALF) measurements of chlorophyll-a (Chl), phycoerythrin (PE), chromophoric dissolved organic matter (CDOM), and variable fluorescence (Fv/Fm) from CCE process cruises in the California Current System, 2006 - 2012.
The Advanced Laser Fluorometry (ALF) provides spectral deconvolution (SDC) analysis of the laser-stimulated emission (LSE) excited at 405 or 532/510 nm for assessment of chlorophyll a, phycoerythrin, and chromophoric dissolved organic matter. Three spectral types of phycoerythrin are discriminated for characterization of cyanobacteria and cryptophytes in mixed phototrophic populations. The SDC analysis is integrated with measurements of variable fluorescence, Fv/Fm, corrected for the SDC-retrieved background fluorescence, BNC, for improved photophysiological assessments of phytoplankton photochemical efficiency. Fluorescence measurements are normalized to the water Raman scattering to improve the accuracy of measurements. The ALF measurements of discrete water samples from various depths were made using several ALF instrument modifications (ALF-1, ALF-T, Aquatic Laser Fluorescence Analyzer (ALFA) , or Custom Laser Analytical Spectroscopic System (CLASS)) during CCE process cruises.

This experiment is one implementation of a globally distributed experiment, known as the Nutrient Network. At Cedar Creek, as in over 70 other sites in grasslands around the world, the experiment aims to describe impacts of increased nutrients (nitrogen, phosphorus, potassium, sulfur and other metals) and decreased herbivory (removal of mammals by fencing). Two overarching questions are being explored with these manipulations: 1. To what extent are plant production and diversity co-limited by multiple nutrients in herbaceous-dominated communities? 2. Under what conditions do grazers or fertilization control plant biomass, diversity, and composition? By utilizing identical protocols at diverse grassland sites around the world, NutNet aims to uncover both the generalities in ecosystem functioning, and the contingencies or differences which can obscure those common mechanisms. In addition to the standard NutNet protocol, e247 includes an additional low Nitrogen gradient (1 gram Nitrogen per meter squared per year and 5 grams Nitrogen per meter squared per year in addition to the standard 10 grams Nitrogen per meter squared per year).
