CD8+ T cell differentiation status correlates with the feasibility of sustained unresponsiveness following oral immunotherapy

<p>While food allergy oral immunotherapy (OIT) can provide safe and effective desensitization (DS), the immune mechanisms underlying development of sustained unresponsiveness (SU) following a period of avoidance are largely unknown. We compared high dimensional immunophenotypes of innate and adaptive immune cell subsets of participants in a phase 2 randomized, controlled, peanut OIT trial who achieved SU vs. DS (no vs. with allergic reactions upon food challenge after a withdrawal period; n=21 vs. 30 respectively among total 120 intent-to-treat participants). Lower frequencies of naïve CD8<sup>+</sup> T cells and terminally differentiated CD57+ CD8<sup>+</sup> T cell subsets at baseline (pre-OIT) were highly associated with SU. Frequency of naïve CD8<sup>+</sup> T cells showed a significant positive correlation with peanut- and Ara h 2-specific IgE at baseline.  Higher frequencies of IL-4<sup>+</sup> and IFNg<sup>+</sup> CD8<sup>+</sup> T cells post-OIT were negatively correlated with SU. Our findings provide compelling evidence that an immune signature consisting of certain CD8<sup>+</sup> T cell subset frequencies is predictive of SU following OIT.</p>

Temporal stability of polymorphic Arctic charr parasite communities reflect sustained divergent trophic niches

<p>Polymorphic Arctic charr Salvelinus alpinus populations frequently display distinct differences in habitat use, diet and parasite communities. Changes to the relative species densities and composition of the wider fish community have the potential to alter the habitat-niche of sympatric Arctic charr populations. This study evaluated the temporal stability of the parasite community, diet and stable isotopes (δ13C, δ15N) of three sympatric Arctic charr morphs (piscivore, benthivore and planktivore) from Loch Rannoch, Scotland, in relation to changes to the fish community. All Arctic charr morphs displayed distinct differences parasite communities, diet and stable isotope signatures over time, despite the establishment of four new trophically transmitted parasite taxa, and increased fish and zooplankton consumption by the piscivorous and planktivore morphs respectively. Native parasite prevalence also increased in all Arctic charr morphs. Overall, Loch Rannoch polymorphic Arctic charr morph populations have maintained their distinct trophic niches and parasite communities through time despite changes in the fish community. This result indicates that restocking of a native fish species has the potential to induce shifts in the parasite community and diet of Arctic charr morphs.</p>

Autonomous And Sustained Perching On Smooth Surfaces

<p><strong>In this video the results are shown of the extended work on aerial perching, which was started in the work &ldquo;Mechanism For Perching On Smooth Surfaces Using Aerial Impacts&rdquo;. </strong></p> <p>&nbsp;</p> <p><strong>The experiment in this video shows the capabilities of an automated and self-regulating perching drone. After the user gives the &lsquo;go&rsquo; signal, the drone autonomously impacts the surface and slows down the rotors after successfully attaching. The drone measures the pressure in the vacuum-cup using a pressure-sensor. If the vacuum gets below a threshold level, meaning that the vacuum is about to become too low to remain perched, the drone reactivates and actively presses the vacuum-cup to regain the vacuum. A significant force is required to repress the vacuum-cup, which was done using the knowledge gained from the work &ldquo;Application Of Substantial And Sustained Force To Vertical Surfaces Using A Quadrotor&rdquo;.</strong></p> <p>&nbsp;</p> <p><strong>Using the approach described above and the integrated sensing capabilities, the drone is capable of safely remaining airborne (mostly perched on a smooth surface) for 45 minutes(!) on a single battery and having the drone turned on only for about 3 minutes.</strong></p>

Application Of Substantial And Sustained Force To Vertical Surfaces Using A Quadrotor

<p><strong>One of the challenges in the interaction between aerial manipulators (drones with manipulator arms) and the environment is to maintain stability under high interaction forces. The dynamics of the system change, and due to limitations to the drone-platform-- the drone not being to apply sideways forces and having limited yaw-torque-- stability is not naturally guaranteed. </strong></p> <p>&nbsp;</p> <p><strong>In this video we demonstrate a new control approach to overcome these limitations. The video shows a quadrotor drone with manipulator pushing on the environment. More precisely, the stability is achieved using a state-feedback approach to compensate for the roll and yaw errors. This approach uses the linearized contact dynamics under pseudo-static conditions. The key element is that non-zero roll-states are used to compensate for errors &nbsp;&nbsp;in the yaw-state. At a certain point the drone reaches pitch angles of over 45&ordm;. Given that the drone has a mass of &nbsp;1.5 kg, this means that the drone is pushing with 15N(!) of force on the environment. The video demonstrates that the drone is easily able to maintain stability over a longer time period.</strong></p> <p>&nbsp;</p> <p><strong>These high interaction forces can be extremely useful when operating tools on the environment, as tasks such as grinding and brushing require sufficient contact pressure to function.</strong></p>

Scientific Webinar on Sustainable Public Food Procurement (SPFP) in the European Union

<p>On 23 April 2024, <a href="https://sapiensnetwork.eu/">SAPIENS Network</a> <a href="https://sapiensnetwork.eu/research/early-stage-researcher-projects/sustainability-to-collective-table/">Early Stage Researcher Chiara Falvo</a> held a scientific webinar focusing on Sustainable Public Food Procurement (SPFP) in the European Union. The event took place in hybrid form and was hosted within the Master&rsquo;s Course in Food Systems Law at the Department of Law of the University of Turin, also in collaboration with the Department of Agricultural, Forestry and Food Sciences (DISAFA). After a brief introduction on public procurement law and practice, Chiara delved into the legal strategies and mechanisms for integrating social and environmental considerations into the procurement of food and catering services. She also highlighted some national and local experiences that are leading the way in the field. During the event, <a href="https://www.giurisprudenza.unito.it/do/docenti.pl/Alias?silvia.mirate#tab-profilo">Professor Silvia Mirate</a>, who also acted as a discussant, provided an overview on the new EU Deforestation Regulation (EUDR), followed by Chiara's exploration of its relevance for public procurement. Contributing to bridging the gap between the scientific domains of law and agricultural and forestry sciences, this webinar may be relevant for students and newcomers to public procurement, especially in the food sector, as well as for anyone interested in understanding deforestation issues and the latest legal mechanisms to combat them.&nbsp;</p>

Measuring social dimensions of sustainability at the community level: An illustrative but cautionary tale

<p>Many communities are working to enhance the sustainability of their physical, economic, and social systems. While economic and physical systems are routinely measured (e.g., money, energy, greenhouse gas emissions), important psychological and behavioral elements of social systems (norms, attitudes, individual behavior) are seldom tracked. This research evaluated a potentially scalable approach to measuring the impact of local sustainability initiatives on these variables in a community engaged in a holistic effort to promote sustainability. Survey data were collected at two timepoints measuring pro-environmental thought and behavior in two small towns in Ohio: Oberlin, a community engaged in holistic efforts to enhance environmentally sustainable behavior; and Berea, a similar community used as a control. Despite verifiable changes in Oberlin due to sustainability programs and awareness of these programs, our survey results did not provide strong evidence that program efforts resulted in the desired changes in attitudes, norms and behaviors. Pro-environmental attitudes about recycling and installing LED bulbs were two exceptions. Conclusions: Assessing the psychological and behavioral dimensions of sustainability poses particular challenges. In our study, we encountered ceiling effects and inadequate statistical power. Possibly norms and attitudes are not easily influenced even by a holistic community-wide effort.</p>

Supplementing a grain diet with insects instead of fruits sustains the body condition of an omnivorous bird

<p>Omnivores utilize dietary sources that differ in nutrients, hence dietary limitations due to environmental change or habitat alteration could cause nutrient limitations, and thus deterioration of body condition if omnivory is obligate. We investigated how the body condition of the omnivorous Village Weaver <em>Ploceus cucullatus</em>, which forages predominantly on grains, responds to the supplementation of its grain diet with insects instead of fruits. Forty wild‐caught weavers held in aviaries were fed a combination of grains and fruits, or grains and insects ad libitum for 8 weeks. We determined diet preference by recording the number of birds on each diet option per minute for 1 h and the amount of food left over after 3 hours of foraging. Fortnightly, we assessed indices of body condition including body mass, pectoral muscle, and fat scores, packed cell volume (PCV), and hemoglobin concentration (HBC). We modeled the number of foragers, food left over, and body condition indices as functions of diet while accounting for time (weeks) and sex effects. Grains were the preferred diet, but males ate more fruits and insects than females. Weavers fed on grains and fruits lost body and pectoral muscle mass and accumulated less fat than those fed on grains and insects. This effect was sex‐dependent: females supplemented with fruits lost more pectoral muscle mass than males of the same group and males but not females, supplemented with insects accumulated more fat reserve than those supplemented with fruits. PCV and HBC did not differ between diets but increased over the 8 weeks. Village Weavers are likely obligate rather than facultative omnivores, with insects being a more nutritive supplement than fruits. Nutrient limitations arising from environmental change or habitat alteration could impair body condition and affect physiological function to environmental seasonality in obligate omnivores like the weavers.</p>
