Coda Calibration for Small Earthquakes near Rock Valley, Nevada

This dataset was produced using the Coda Calibration Tool (Barno, 2017). It estimates moment magnitude (Mw) for small earthquakes near Rock Valley in southern Nevada. Included in the dataset are the calibration parameter file and curated waveform envelopes in SAC format for calibration and test events. The calibration parameter file (Calibration_Parameters.json) includes shape parameters and site terms for 20 stations, as well as independent Mw for 8 calibration events. Waveform envelopes for only the calibration events are found in the calibration_env folder. Waveform envelopes for test events are found in the all_env folder. A calibration should be run using only the envelopes in calibration_env since there is not validation Mw for the test events (i.e., running a calibration including the test events will affect the site terms).

Bottle-calibrated dissolved oxygen profiles from yearly turn-around cruises for the Ocean Observations Initiative (OOI) Irminger Sea Array 2014-2022

This dataset contains bottle-calibrated dissolved oxygen (DO) profiles collected from Conductivity Temperature Depth (CTD) casts on turn-around cruises performed yearly to maintain the Ocean Observations Initiative (OOI) Global Irminger Sea Array (60.46°N, 38.44°W). DO profiles were used in conjunction with oxygen bottle measurements (Winklers) to produce a post-cruise oxygen-calibrated CTD product for scientific use. Bottle-calibrated CTD salinity products were used to produce post-cruise oxygen-calibrated CTD profiles starting in 2018 (Year 5). This document contains overviews of CTD data collection and processing and post-processing oxygen sensor calibration method. Reports for each cruise include a summary of relevant cruise events, oxygen sensor calibration results, and issues/problems associated with oxygen data collected on each cruise. This dataset has been created for end-users that require field-calibrated oxygen data products that are currently not provided by OOI through its standard data dissemination.

Miniature iron-carbon eutectic point crucible for the calibration of thermometers

The article presents the design, construction and evaluation of a miniature graphite iron-carbon eutectic point crucible for the calibration of thermometers. These are either contact such as thermocouples or non-contact such as infrared thermometers. The double wall of the crucible protects its structure in case of cracks in graphite that can appear due to thermal expansion or eutectic adhesion. Such design proved robust – even after almost 100 melting realizations, the crucible does not show significant signs of damage. Performance of the crucible was confirmed with several realized plateaus at three different furnace setpoints. Results of these measurements agree with previously published findings. The measured melting point temperature was 1153.81 °C and the achieved expanded calibration uncertainty of a type R thermocouple was 0.2 °C.

Patch-clamp dataset (SyncroPatch384PE) for control variants used in a calibrated functional patch clamp assay to enhance clinical variant interpretation in KCNH2-related long QT syndrome

Modern sequencing technologies have revolutionised our detection of gene variants. However, in most genes, including KCNH2, the majority of missense variants are currently classified as variants of uncertain significance (VUS). The aim of this study was to investigate the utility of an automated patch-clamp assay for aiding clinical variant classification in the KCNH2 gene. The assay was designed according to recommendations proposed by the ClinGen Sequence Variant Interpretation Working Group. Thirty-one variants (17 pathogenic/likely pathogenic, 14 benign/likely benign) were classified internally as variant controls. They were heterozygously expressed in Flp-In HEK293 cells for assessing the effects of variant on current density and channel gating in order to determine the sensitive and specificity of the assay. All 17 pathogenic variant controls had reduced current density and 13/14 benign variant controls had normal current density, which enabled determination of normal and abnormal ranges for applying moderate or supporting evidence strength for VUS reclassification. Inclusion of KCNH2 functional assay evidence enabled us to reclassify 6 out of 44 VUS as likely pathogenic. The high-throughput patch clamp assay can provide moderate strength evidence for clinical interpretation of clinical KCNH2 variants and demonstrates the value of developing automated patch clamp assays for functional characterisation of ion channel gene variants.
