Magnetic resonance imaging reveals human brown adipose tissue is rapidly activated in response to cold

<p class="MsoNoSpacing"><b>Context.</b> In rodents, cold exposure induces the activation of brown adipose tissue (BAT) and the induction of intracellular triacylglycerol (TAG) lipolysis. However, in humans, the kinetics of supraclavicular (SCV) BAT activation and the potential importance of TAG stores remain poorly defined.</p> <p class="MsoNoSpacing"><b>Objective.</b> To determine the time course of BAT activation and changes in intracellular TAG using magnetic resonance imaging (MRI) assessment of the SCV (i.e. BAT depot) and fat in the posterior neck region (i.e. non BAT).</p> <p class="MsoNoSpacing"><b>Design.</b> Cross-sectional.</p> <p class="MsoNoSpacing"><b>Setting.</b> Clinical research centre.</p> <p class="MsoNoSpacing"><b>Patients or Other Participants.</b> Twelve healthy male volunteers ages 18-29 years [BMI=24.7±2.8kg/m<sup>2</sup> and body fat percentage = 25.0±7.4% (both mean±SD)].</p> <p class="MsoNoSpacing"><b>Intervention(s).</b> Standardized whole-body cold exposure (180 minutes at 18<span>°</span>C) and immediate re-warming (30 minutes at 32°C).</p> <p class="MsoNoSpacing"><b>Main Outcome Measure(s).</b> Proton density fat fraction (PDFF) and T2* of the SCV and posterior neck fat pads. Acquisitions occurred at 5-15 minute intervals during cooling and subsequent warming.</p> <p class="MsoNoSpacing"><b>Results.</b> SCV PDFF declined significantly after only 10 minutes of cold exposure [-1.6% (standard error (SE) 0.44%), <i>p</i>=0.007) and continued to decline until 35 minutes after which time it remained stable until 180 minutes. A similar time course was also observed for SCV T2*. In the posterior neck fat (non-BAT) there were no cold-induced changes in PDFF or T2*. Re-warming did not result in a change in SCV PDFF or T2*.</p> <p class="MsoNoSpacing"><b>Conclusions.</b> The rapid cold-induced decline in SCV PDFF suggests that in humans, BAT is activated quickly in response to cold and that TAG is a primary substrate.</p>

Interrelationship among thigh intermuscular adipose tissue, cross-sectional area, muscle strength and functional mobility in older subjects

<p><strong>Objectives:</strong> The aim of this cross-sectional study was to investigate the association between lower limb strength, muscle mass and composition and balance ability in elders. <strong>Methods:</strong> Thirthy-four older participants (Age: 65.6&plusmn;4.73 years; male=10 and female=24) were assessed for muscle strength (maximal isometric strength of knee extensors and one repetition maximum by leg press, 1RM), balance and gait capacity (Mini-BESTest), body composition by DXA (obtaining Appendicular Skeletal Muscle Mass Index, ASMMI) and magnetic resonance imaging of thigh to evaluate Intermuscular Adipose Tissue (IMAT) and muscle Cross Sectional Area (CSA). <strong>Results:</strong> Positive correlations between 1RM leg press and ASMMI (r<sub>s</sub>=0.64, P&lt;.0001) and thigh CSA (r<sub>s</sub>=0.52, P=.0017), but not with thigh IMAT, were found. In addition, significant correlations between knee extensors strength and ASMMI (r<sub>s</sub>=0.48, P=.004) and thigh CSA (r<sub>s</sub>=0.49, P=.0033) and IMAT (r<sub>s</sub>=-0.35, P=.043) were observed whereas no significant correlations between the Mini-BESTest with ASMMI, thigh CSA and IMAT were observed. <strong>Discussion:</strong> Lower limb strength positively correlated with appendicular muscle mass. Further, the maximal isometric strength of knee extensors negatively correlated with thigh IMAT in elderly patients whereas the dynamic balance ability did not correlate with any of the morphological variables of the muscle (i.e. ASMMI, CSA and IMAT). A reduced muscle size and strength could affect movement and reduce physical function in older patients. Improving the composition and size of muscle in elder subjects could reduce frailty and risk of falls.</p>

<p><span>The volume of epicardial adipose tissue (EAT) is associated with an increased incidence of cardiovascular disease (CVD); however, only a few studies have examined its effect on the myocardial function of endurance in athletes. The association between the EAT and the variation of myocardial function is still unclear in amateur marathoners. IL-17 signalling is critical for controlling subcutaneous adipose tissue dynamics and parasite burden during chronic Trypanosoma brucei infection

<p>In the skin, <em>Trypanosoma brucei </em>colonises the subcutaneous white adipose tissue (WAT) and harbours a pool of parasites competent for forward transmission. The interaction between parasites, adipose tissue, and the local immune system is likely to drive adipose tissue wasting and weight loss observed in cattle and humans infected with <em>T. brucei</em>. &nbsp;However, mechanistically, this process is not fully understood. Here, using several complementary approaches including mass cytometry by time of flight, bulk and single cell transcriptomics, we found that <em>T. brucei</em> infection drives a local expansion of several IL-17A-producing cells in the murine WAT, including T<sub>H</sub>17 and Vg6<sup>+</sup> T cells. We also found that global IL-17 deficiency, or mice lacking IL-17 receptor expression specifically on adipocytes, were protected from infection-induced WAT wasting and weight loss. Unexpectedly, we found that abrogation of IL-17 signalling on adipocytes results in higher burden of extravascular parasites in the WAT. Taken together, our study highlights the central role of IL-17 signalling on adipocytes in controlling WAT responses to infection, suggesting that adipocytes are a critical coordinator of the tissue immune response to <em>T. brucei</em> infection.&nbsp;</p>
