Metagenomics data of the bacterial community in Bemisia tabaci from Burkina Faso

Microbial symbionts are widespread in insects and some of them have been associated to adaptive changes. Primary symbionts (P-symbionts) have a nutritional role that allows their hosts to feed on unbalanced diets (plant sap, wood, blood). Most of them have undergone genome reduction, but their genomes still retain genes involved in pathways that are necessary to synthesize the nutrients that their hosts need. However, in some P-symbionts, essential pathways are incomplete and secondary symbionts (S-symbionts) are required to complete parts of their degenerated functions. The P-symbiont of the phloem sap-feeder Bemisia tabaci, Portiera aleyrodidarium, lacks genes involved in the synthesis of vitamins, cofactors, and also of some essential amino-acids. Seven S-symbionts have been detected in the B. tabaci species complex. Phenotypic and genomic analyses have revealed various effects, from reproductive manipulation to fitness benefits, notably some of them have complementary metabolic capabilities to Portiera, suggesting that their presence may be obligatory. In order to get the full picture of the symbiotic community of this pest, we investigated, through metabarcoding approaches, the symbiont content of individuals from Burkina Faso, a West African country where B. tabaci induces severe crop damage. While no new putative B. tabaci S-symbiont was identified, Hemipteriphilus, a symbiont only described in B. tabaci populations from Asia, was detected for the first time on this continent. Phylogenetic analyses however reveal that it is a different strain than the reference found in Asia. Specific diagnostic PCRs showed a high prevalence of these S-symbionts and especially of Hemipteriphilus in different genetic groups. These results suggest that Hemipteriphilus may affect the biology of B. tabaci and provide fitness advantage in some B. tabaci populations.

Dataset related to the responses of eggplant plants to Bemisia Tabaci whitefly in presence of one of its natural enemies, the Macrolophus pygmaeus plant bug

This dataset assesses the response of eggplant plants to 3 conditions: noninfested control plants, plants infested by Bemisia tabaci whitefly, and plants infested by Bemisia tabaci where Macrolophus pygmaeus plant bug was also released.

The data of the various parameters relating to the responses of the different eggplant plants to the three tested conditions were analyzed using a one-way ANOVA. Where significant differences were detected, the means were separated by Tukey’s HSD test (p < 0.05). Statistical analysis was carried out using the program Statistica (StatSoft, TIBCO Software Inc., Tulsa, OK, USA).

The provided dataset was used in the investigation of whether the Macrolophus pygmaeus (Hemiptera: Miridae) plant bug can mitigate the damage caused to plants by the Bemisia tabaci whitefly (Hemiptera: Aleyrodidae). A discussion and interpretation of the data, as well as subsequent results, is provided in the scientific publication "Can Macrolophus pygmaeus (Hemiptera: Miridae) Mitigate the Damage Caused to Plants by Bemisia tabaci (Hemiptera: Aleyrodidae)?", published in the Insects journal.

This research was conducted within the VIRTIGATION project, which is part of the EU Open Research Data pilot. The genome assemble of Bemisia tabaci MED

Aim: The sweet potato whitefly, Bemisia tabaci MED is a globally invasive species that causes serious economic damage to agroecosystems. Despite the significant threat it poses to agricultural and economic crops worldwide, the global perspective of the invasion patterns and genetic mechanism contributing to the success of this notorious pest is still poorly understood. The objective of this research was to enhance genome and population genetic analyses to better understand the intricate invasion patterns of B. tabaci MED.

Location: Samples were collected in native (Spain, Croatia, Bosnia and Herzegovina, Cyprus, and Israel) and invaded regions (China, South Korea and North America).

Methods: We first assembled a chromosome-scale reference genome of B. tabaci MED, and then employed the restriction site‐associated 2b‐RAD method to genotype over 20, 000 high‐quality single nucleotide polymorphisms from 29 geographical populations.

Results: A reference genome of B. tabaci MED, with a size of 637.47 Mb, was available. The majority of the assembled sequences (99%) were anchored onto ten linkage groups, with an N50 size of 58.76 Mb, representing a significant improvement over previous whitefly genome assemblies. We identified rapidly expanded gene families and positively selected genes, probably contributing to successful invasion and rapid adaptation to the new environment. Population genomics analysis showed that three highly differentiated genetic groups were formed, and complex and extensive gene flow occurred across the Mediterranean populations. The genetic admixture patterns in East Asia populations were distinct from those in North America, indicating that they had different source populations.

Conclusions: The high-quality, chromosome-scale genome of B. tabaci MED offered opportunities for more comprehensive genome-wide studies, and provided a solid foundation for the complex introduction events and the differential invasiveness of B. tabaci MED worldwide.

Evolved transcriptional responses and their trade-offs after long-term adaptation of Bemisia tabaci to a marginally-suitable host

Scripts and data used in the work "Evolved transcriptional responses and their trade-offs after long-term adaptation of Bemisia tabaci to a marginally-suitable host".

Abstract: Although generalist insect herbivores can migrate and rapidly adapt to a broad range of host plants, they can face significant difficulties when accidentally migrating to novel and marginally-suitable hosts. What happens, at both the performance and transcriptional levels, if these marginally-suitable hosts must be used for multiple generations before migration to a suitable host can take place, largely remains unknown. In this study, we established multigenerational colonies of the whitefly Bemisia tabaci, a generalist phloem-feeding species, adapted to a marginally-suitable host (habanero pepper) or an optimal host (cotton). We used reciprocal host tests to estimate the differences in performance of the populations on both hosts under optimal (30 oC) and mild-stressful (24 oC) temperature conditions, and documented the associated transcriptomic changes. The habanero pepper-adapted population greatly improved its performance on habanero pepper but did not reach its performance level on cotton, the original host. It also showed reduced performance on cotton, relative to the non-adapted population, and an antagonistic effect of the lower-temperature stressor. The transcriptomic data revealed that most of the expression changes, associated with long-term adaptation to habanero pepper, can be categorized as “evolved” with no initial plastic response. Three molecular functions dominated: enhanced formation of cuticle structural constituents, enhanced activity of oxidation-reduction processes involved in neutralization of phytotoxins and reduced production of proteins from the cathepsin B family. Taken together, these findings indicate that generalist insects can adapt to novel host plants by modifying the expression of a relatively small set of specific molecular functions.

Laboratory bioassay of insecticides mixtures (neonicotinoids and ketoenols) against Bemisia tabaci Asia I

Cotton leaves were dipped in serially diluted solutions of formulated insecticides for 10 s with slight agitation. The leaves with second instar nymphs that were dipped in double-distilled water containing 0.1 g L-1 Triton X-100 only, served as control. Each bioassay including control used 3-4 replicates at a minimum of eight different concentrations and were maintained at the controlled growth condition. All the insecticides concentrations were selected to give a range of 0-100% mortality of B. tabaci nymphs. Two weeks later final mortality was assessed when the last nymphal instar had been reached on control plants. It was computed by comparing the number of second instar nymphs present at the time of treatment with the number remaining dead or unhatched on the day of mortality assessment. For bioassays with synergists (PBO and DEF), the cotton leaves containing the second instar nymphs of B. tabaci were dipped into synergist solutions (100 mg L-1) for 10 s at least 2 h before the imposition of insecticide treatments. Other procedures were same to those with insecticides only.
