HR-EBSD Measurements near Twins in Zicaloy-2

HR-EBSD measurements made on a Zircaloy-2 sample deformed to 2.7% in tension to generate multiple deformation twins.
HR-EBSD measurements were made using A Bruker HReFlash detector mounted on a Zeiss Merlin SEM. Sample preparation used Ar ion beam milling on a Gatan PECS II system as a final step.
The strain, stress and dislocation density variations available here were then calculated using in-house Matlab code (XEBSD). The work and further analysis of the results is described in

Assessment of residual stress fields at deformation twin tips and the surrounding environments, Abdolvand & Wilkinson, Acta Materialia (2016)
dx.doi.org/10.1016/j.actamat.2015.11.036

The analysis methods implement in XEBSD are described in the following publications:

High resolution electron backscatter diffraction measurements of elastic strain variations in the presence of larger lattice rotations
TB Britton and AJ Wilkinson
Ultramicroscopy, (2012), vol. 114, 82-95
doi:10.1016/j.ultramic.2012.01.004

Measurement of residual elastic strain and lattice rotations with high resolution electron backscatter diffraction
TB Britton and AJ Wilkinson
Ultramicroscopy, (2011) vol. 111, 1395-1404
doi:10.1016/j.ultramic.2011.05.007

Determination of elastic strain fields and geometrically necessary dislocation distributions near nanoindents using electron back scatter diffraction
AJ Wilkinson and D Randman
Philosophical Magazine, (2010), vol. 90, 1159-1177
doi:10.1080/14786430903304145

Crystal Plasticity Analysis of Micro-Deformation, Lattice Rotation and Geometrically Necessary Dislocation Density
FPE Dunne, R Kiwanuka, AJ Wilkinson
Proc. Complete Data and Results for "Four-of-a-kind? Comprehensive atmospheric characterisation of the HR~8799 planets with VLTI/GRAVITY"

Four-of-a-kind? Comprehensive atmospheric characterisation of the HR~8799 planets with VLTI/GRAVITY

We explored the atmospheres of the HR 8799 system of directly imaged planets using petitRADTRANS atmospheric retrievals. This required reprocessing existing data, preparing new observations from VLTI/GRAVITY and combining additional archival spectroscopic and photometric datasets. We ran 95 retrievals across the four planets, and include here the full set of outputs from the sampling process, as well as synthesised results for each retrieval. Finally, we also include templates for running these retrievals.

This data, together with the results of the retrievals and templates for running similar emission spectra retrievals are included in this Zenodo archive. Additional README files are included throughout the archive in order to explain file structures, units, and to provide original references where necessary. An archived repository of the petitRADTRANS code can be found at https://doi.org/10.5281/zenodo.10892021, while the up-to-date repository is available at https://gitlab.com/mauricemolli/petitRADTRANS.

Time series transcriptomes resolve metabolic pathways underlying crocin's anti-cancer activity | Dataset: sequencing reads (2,6,12,24 hr crocin treatments)

Natural products like saffron show promise in treating hepatocellular carcinoma (HCC), but their mechanisms remain unclear. Here, we used time-series transcriptomics to elucidate crocin's anti-cancer mechanisms in HCC cells. We treated HepG2 cells with 1 and 2 mM crocin for 2, 6, 12, and 24 hours and analyzed transcriptomic profiles at each timepoint. The strongest transcriptional response occurred at 2 hours with 1 mM crocin, with diminishing effects at later timepoints. We observed upregulation of metabolic-, adhesion-, and endocytosis-related genes across all timepoints. Pathway analysis revealed activation of DNA damage checkpoints and senescence while proliferation pathways were suppressed. Notably, 52 genes involved in non-alcoholic fatty liver disease were downregulated at 24 hours (FDR p = 8 × 10⁻⁸), suggesting reversal of carcinogenic pathways. Strikingly, crocin consistently downregulated spliceosomal machinery genes across all timepoints while upregulating senescence and autophagy pathways. This spliceosome targeting represents a clinically relevant mechanism, as aberrant splicing drives oncogenesis in more than 90% of cancers. The transcription factor PAX5 was significantly upregulated while oncogenic ELK1 targets were downregulated. Our findings show that crocin treatment is accompanied by HCC cell senescence induction through coordinated spliceosome disruption and metabolic reprogramming, providing novel therapeutic targets for hepatocellular carcinoma.

Keywords: Hepatocellular carcinoma (HCC), crocin, transcriptomics, spliceosome, senescence, natural anti-cancer compounds
