Monoclonal antibodies from humans with Mycobacterium tuberculosis exposure or latent infection recognize distinct arabinomannan epitopes

The surface polysacharide arabinomannan (AM) and related glycolipid lipoarabinomannan (LAM) play critical roles in tuberculosis pathogenesis. Human antibody responses to AM/LAM are heterogenous and knowledge of reactivity to specific glycan epitopes at the monoclonal level is limited, especially in individuals who can control M. tuberculosis infection. We generated human IgG mAbs to AM/LAM from B cells of two asymptomatic individuals exposed to or latently infected with M. tuberculosis. We here show that two of these mAbs have high affinity to AM/LAM, are non-competing, and recognize different glycan epitopes distinct from other anti-AM/LAM mAbs reported. Both mAbs recognize virulent M. tuberculosis and nontuberculous mycobacteria with marked differences, can be used for the detection of urinary LAM, and can detect M. tuberculosis and LAM in infected lungs. These mAbs enhance our understanding of the spectrum of antibodies to AM/LAM epitopes in humans and are valuable for tuberculosis diagnostic and research applications.

Transcriptome profiling of human monocytes from accute phase and convalescent patients infected with Plasmodium vivax malaria

RNA sequencing raw data of monocytes obtained from five patients in acute phase of Plasmodium vivax malaria and 45 days after treatment. Individuals from Rondônia (Brazil) were between 18 and 60 years and attended to the exclusion criteria: chronic inflammatory or infectious diseases, pregnancy, and breastfeeding. Diagnostic of malaria by clinical symptoms and thick blood smear was also confirmed by qPCR. According to the recommended protocol of the Brazilian Ministry of Health patients were treated with chloroquine associated to primaquine. Protocols used were approved by the Ethical Committees on Human Experimentation from Instituto Rene ́ Rachou, Fiocruz, and National Ethical Council (CAAE: 59902816.7.0000.5091).

Sequencing was performed in a NextSeq 500 system using the NextSeq 500/550 High Output Kit v2.5 for 75 cycles (both from Illumina). Before sequencing total mRNA samples were prepared using TruSeq Stranded mRNA Kit (Illumina) according to the manufacturer’s protocol. Files are in FASTQ format, just as they were downloaded from Illumina's cloud environment (BaseSpace) right after sequencing. Files are identified by patient number (1 to 5) and health condition: acute phase of the disease or convalescent (45 days after treatment).

Data for: The impact of wildlife and environmental factors on hantavirus infection in host and its translation into human risk

Identifying factors that drive infection dynamics in reservoir host populations is essential in understanding human risk from wildlife-originated zoonoses. We studied zoonotic Puumala orthohantavirus (PUUV) in the host, the bank vole (Myodes glareolus), populations in relation to the host population, rodent and predator community and environment-related factors and whether these processes are translated into human infection incidence. We used 5-year rodent trapping and bank vole PUUV serology data collected from 30 sites located in 24 municipalities in Finland. We found that PUUV seroprevalence was negatively associated with the abundance of red foxes, but this process did not translate into human disease incidence, which showed no association with PUUV seroprevalence. The abundance of weasels, the proportion of juvenile bank voles in the host populations and rodent species diversity were negatively associated with the abundance index of PUUV-positive bank voles, which, in turn, showed a positive association with human disease incidence. Our results suggest certain predators, high proportion of young bank vole individuals and a diverse rodent community, may reduce PUUV risk for humans through their negative impacts on the abundance of infected bank voles.
