PAML data set for integrins, tetraspanins, and ADAMs in the coral Acropora

Data set for gamete composing integrins, tetraspanins, and ADAMs in the coral Acropora digitifera. The LC-MS/MS analyses of eggs or sperm of the coral Acropora digitifera were conducted. Identified integrins, tetraspanins, and ADAMs which are reported in their involvement in the fertilization of the coral Acropora. The data sets were used for analyses of molecular evolutionary analyses with codeml to select which genes encoding the proteins showed accelerated evolution during acquisition of species-specific fertilization. cDNA sequences of the identified proteins were used for collecting the orthologues of the Acropora spp using Orthoscope.

Synergistic phase separation of two pathways promotes integrin clustering and nascent adhesion formation

Integrin adhesion complexes (IACs) are integrin-based plasma membrane-associated compartments where cells sense environmental cues. The physical mechanisms and molecular interactions that mediate initial IAC formation are unclear. We found that both p130Cas ("Cas") and Focal adhesion kinase ("FAK") undergo liquid-liquid phase separation in vitrounder physiologic conditions. Cas- and FAK- driven phase separation is sufficient to reconstitute kindlin-dependent integrin clustering in vitro with recombinant mammalian proteins. In vitro condensates and IACs in mouse embryonic fibroblasts (MEFs) exhibit similar sensitivities to environmental perturbations including changes in temperature and pH. Furthermore, mutations that inhibit or enhance phase separation in vitro reduce or increase the number of IACs in MEFs, respectively. Finally, we find that the Cas and FAK pathways act synergistically to promote phase separation, integrin clustering, IAC formation and partitioning of key components in vitro and in cells. We propose that Cas- and FAK- driven phase separation provides an intracellular trigger for integrin clustering and nascent IAC formation.

Data from: MFAP5 contributes to inflammation, synovial hyperplasia and joint damage in rheumatoid arthritis via integrin-ERK signaling

Synovial hyperplasia is the primary pathological changes of RA and FLS is the principal etiological cellular component that drives and constitutes the pathological lesions. To elucidate the molecular mechanism of how FLS may contribute to RA, we performed transcriptome profiling to determine the DEGs between normal and RA FLS. We compared the primary FLS isolated from healthy control (HFLS) to that isolated from RA patients (RA FLS), and also to an immortalized RA FLS cell line MH7A immortalized by stably transfecting primary RA FLS cells with SV40 T antigen gene.
