VISION Invited lecture - New Perspectives in Clinical Trials

<p>Recording and presentation&nbsp;of the invited lecture that took place online on 15 June 2022 -&nbsp;<strong>Prof. Stefano Bonassi, Ph.D., ERT -&nbsp;New Perspectives in Clinical Trials</strong>.</p> <p>Clinical trials are our best approach to generate clinical evidence. Since the early stages in the first decades of the past century, clinical trial has been in continuous evolution. Innovation has interested the formulation of stopping guidelines for safety, efficacy, and futility; the use of internet, changes in the way the informed consent is requested, the introduction of pragmatic trials, the challenge of personalized medicine, the concept of non-inferiority, and many others. Today, trials range from a single person&nbsp;to hundred-thousand people, from a single lab to hundreds of centers around the world, from simple two-arm randomizations to increasingly complex study designs. The lecture will address issues related to the performance of clinical trials, how to deal with the increasing use of the internet, the dramatic change concerning ethics in clinical studies.</p>

VISION Invited lecture - Objective: Personalised Prevention of Pancreatic Cancer

<p>Recording of the invited lecture that took place online on 16 December 2021 -&nbsp;<strong>N&uacute;ria Malats, MD, MPH, PhD -&nbsp;Objective: Personalised Prevention of Pancreatic Cancer.</strong></p> <p>Identifying the population at high risk of developing non-hereditary pancreatic cancer in order to offer them to enter screening programs and diagnose cancer in early stages, is a must. To this end, we integrate information on the risk factors for this tumor with biomarkers determined at the omics level, including genomics and microbiome. All this information comes from a large study carried out in 28 centres from six European countries. For the integration of this big molecular data with epidemiological and clinical data, we use artificial intelligence tools. In the lecture, I will go through the most recent achievements of these collective efforts.</p>

Genet., 1st Faculty of Medicine, Charles University, Albertov 6, Prague 2, Czech Rep.</p> <p><sup>3</sup>&nbsp;Biomedical Center, Faculty of Medicine in Pilsen, Charles University Prague, Pilsen, 30100, Czech Rep.</p> <p>Colorectal cancer (CRC) continues to be one of the leading malignancies and causes of tumour-related deaths worldwide. Both impaired DNA repair mechanisms and disrupted telomere length homeostasis represent potential culprits in CRC onset, its dissemination in the body and prognosis. Above parameters are becoming critical as prognostic markers, since CRC therapy is based on compounds interacting with DNA. DNA repair capacity in CRC patients have recently been studied in order to address prediction of therapy response. Due to the substantial interindividual variations in DNA repair capacities and relative telomere length, these markers may pose a possible contribution in individualized therapeutical regimen of CRC patients. Telomere attrition, responsible for replicative senescence in healthy cells, may become a hallmark of malignant transformation of the cell due to by-passing cell cycle checkpoints. Telomerase &ndash; a key enzyme keeping homeostasis of telomere - is almost ubiquitous in advanced solid cancers, including CRC, and its expression is fundamental to cell immortalization.<br> Here we present our data based on the investigation of base excision repair capacities and relative telomere length in tumor tissues and adjacent non-malignant mucosa of sporadic CRC patients. The relative gene expression of telomerases is monitored as well. Particular attention will be dedicated to the CRC phenotypes and clinicopathological characteristics. We also addressed telomere homeostasis in peripheral blood lymphocytes of CRC patients in several consecutive samplings (at diagnosis, immediately after treatment and in additional follow-up intervals), which could provide us the insight into the treatment response. This aspect is of particular relevance, since there is currently a persistent effort to develop therapeutics, which are telomerase-specific and gentle to non-malignant tissue. However, in practice, we are at the dawn of clinical trials. Additionally, emerging crosstalks between DNA repair and telomere length homeostasis may cast some lights on a dynamic of genomic instability, a fundamental hallmark (or cause) of cancer.</p> <p>Acknowledgement: GACR 21-04607X, 19-10543S, AZV NV18/03/00199</p>

Nur so kann, z.B. die globale, interdisziplin&auml;re Forschung &uuml;ber Pandemien erm&ouml;glicht und daraus neue Erkenntnisse gewonnen werden. Das FDM soll uns die Arbeit erleichtern. Wir versuchen unser Bestes, um es so einfach wie m&ouml;glich zu machen. Daf&uuml;r geben wir Ihnen den Zugang zu Tools und Informationen, damit Sie Ihre Forschungsdaten besser organisieren. H&auml;ufig ist es so, dass Daten nur von einem Forscher, einer Forscherin oder einer Arbeitsgruppe bearbeitet und analysiert werden und danach auf einem USB-Stick gespeichert in einer Schublade verschwinden. Unter anderem besch&auml;ftigen wir uns mit FDM wegen einer neuartigen Datennutzung, Nachnutzung / Publikation. Ein h&auml;ufiger Ansatz generischer Infrastruktureinrichtungen ist immer noch, dass Bibliotheken und RZ auf klassische Publikationsparadigma fokussieren und dabei oft den Forschungsprozess ignorieren. Wichtige Fragen sind f&uuml;r mich / uns wie wir Forschungsdaten &uuml;bernehmen k&ouml;nnen? Und gerade: wie k&ouml;nnen wir Forschungsdaten mit den vorhandenen Recherchetools bereitstellen?</p> <p>Das Teilen von Daten erm&ouml;glicht eine weltweite Arbeitsaufteilung. Forscherinnen und Forscher k&ouml;nnen je nach Zeit, Ressourcen und Aufgabengebieten ihren eigenen Blick auf die Daten werfen und sie entsprechend ihrer individuellen Bed&uuml;rfnisse analysieren.</p>

<p>Es liegen Informationsbl&auml;tter vor, die zeitnah auf unserer Universit&auml;tswebseite &uuml;ber ein Formularcenter zum &#39;Download&#39; bereitgestellt werden sollen. Seit Januar 2020 liegen diese Informationen im Justiziariat. Forscherinnen und Forscher verlangen nach Muster-Einwilligungen und so entstand die Idee zum Thema Muster-Einwilligungen eine Coffee Lecture durchzuf&uuml;hren. Gerne w&auml;re mitgeteilt worden, wo universit&auml;tsintern solche Formulare heruntergeladen werden k&ouml;nnen, aber es kann inzwischen gut auf externe Seiten verweisen werden, z.B. auf den Verbund Forschungsdaten Bildung. In meinem Alltag im FDM werde ich oft zum Datenschutz gefragt, z.B. wie lange die Forschungsdaten archiviert werden d&uuml;rfen und ob die Daten w&auml;hrend der Archivierung anonymisiert werden m&uuml;ssen.</p> <p>Forscherinnen und Forscher m&ouml;chten und m&uuml;ssen wissen, was alles ganz genau vor der Datenerhebung beachtet werden muss.</p>

Artificially-generated Lecture Video Fragmentation Dataset and Ground Truth

<p>We provide a large-scale lecture video dataset consisting of artificially-generated lectures, and the corresponding ground-truth fragmentation, for the purpose of evaluating lecture video fragmentation techniques.</p> <p>For creating this dataset, 1498 speech transcript files (generated automatically by ASR software) were used from the world&#39;s biggest academic online video repository, the VideoLectures.NET. These transcripts correspond to lectures from various fields of science, such as Computer science, Mathematics, Medicine, Politics etc. In order to create the synthetic video lectures, all transcripts were randomly split in fragments, the duration of which ranges between 4 and 8 minutes. Each synthetic lecture was then assembled by combining (stitching) exactly 20 randomly selected fragments. 300 such artificially-generated lectures are included in the released dataset. Each such lecture file has a mean duration of about 120 minutes, thus the dataset contains altogether about 600 hours of artificially-generated lectures. Every pair of consecutive fragments in these lectures originally comes from different videos, consequently the point in time where such two fragments are joined is a known ground-truth fragment boundary. All these boundaries form the dataset&#39;s ground truth. We should stress that we do not generate the corresponding video files for the artificially-generated lectures (only the transcripts), and one should not try to reverse-engineer the dataset creation process so as to use in some way the visual modality for detecting the fragments in this dataset.</p> <p><strong>File format</strong></p> <p>After you download the provided .zip and unpack it, the extracted folder will contain two sub-folders:</p> <pre><code>1. ALV_srt 2. ALV_srt_GT </code></pre> <p>Each of them contains 300 files.</p> <p>The <strong>ALV_srt</strong> folder contains the transcripts of every artificially-generated lecture, in the standard SRT format:</p> <pre><code>1. A numeric counter identifying each sequential subtitle 2. The time that the subtitle should appear on the screen, followed by --&gt; and the time it should disappear 3. Subtitle's text itself on one or more lines 4. A blank line containing no text </code></pre> <p>The <strong>ALV_srt_GT</strong> folder contains the ground truth (GT) fragments corresponding to the lectures (transcripts) of the <strong>ALV_srt</strong> folder. This dataset seeks to help those researchers who want to perform data analytics of student behavior in a classroom.</p> <p>This dataset has the peculiarity that it has been built inside a classroom in an authentic learning situation wherein a face-to-face model a teacher teaches his class in front of a traditional group.</p> <p>The data set achieved seeks to provide support for the analysis of learning activities to identify the good and bad practices and the availability of students to appropriate new thoughts. In addition, these results also serve as support to generate new research through other learning scenarios, whether traditional or distance learning, where the results of the physiological activity of the brain can be used to compare the didactics that produce the best results in the students and the areas of opportunity in the teaching processes.</p>

Zur Politik der &Uuml;berl&auml;uferkom&ouml;die&quot; gefolgt von einer Podiumsdiskussion mit Tatiana Astafeva, Rasmus Greiner, Julia Schumacher und Thomas Weber am 24.01.2023.</p> <p>Published in: TraMeTraMi 5-2023,&nbsp;<a href="https://trametrami.avinus.org/publikationen/5-2023 ">https://trametrami.avinus.org/publikationen/5-2023&nbsp;</a></p> <p>Als &Uuml;berl&auml;uferfilme werden Spielfilme bezeichnet, die in den letzten Jahren des NS-Regimes produziert wurden, aber erst nach dem Krieg in West- oder Ostdeutschland ihre Premiere hatten. Mehr als die H&auml;lfte dieser Filme waren Kom&ouml;dien, viele mit Publikumslieblingen wie Johannes Heesters, Heinz R&uuml;hmann und Theo Lingen. In der &Uuml;berl&auml;uferkom&ouml;die treffen normative Auspr&auml;gungen der NS-Ideologie auf transformative Kr&auml;fte. Diese komplexe Kombination aus Kontinuit&auml;ten und Br&uuml;chen, Reminiszenzen und Differenzen macht die Filme zu einem ebenso vielschichtigen wie auch prek&auml;ren Forschungsgegenstand, der sowohl NS-konforme als auch subversive Potenziale enthalten. Das Videoessay TANZ DER GESPENSTER zeigt am Beispiel zahlreicher Produktionen, wie vielf&auml;ltig aber auch versteckt sich Politik und &Auml;sthetik &uuml;berschneiden k&ouml;nnen. Die teils jahrelange Produktionszeit und die mitunter erst nach dem Krieg erfolgte Fertigstellung der Filme resultierte in h&ouml;chst differenten und inhomogenen Konstellationen. Diesen umrisshaft aufscheinenden und wieder verschwindenden Spuren der &Uuml;berl&auml;uferkom&ouml;dien forscht das Videoessay nach.</p>

Ein Film von Timo Hempel (Interview), Louise Schmidt (Kamera) und Galina Ponomareva (Schnitt).</p> <p>Das Gespr&auml;ch mit Klaus Wildenhahn wurde am 22.02.2013 in einem Eiscaf&eacute; in Hamburg-Eppendorf aufgezeichnet. Eigentlich hatte sich das Team nur zu einem Vorgespr&auml;ch mit Klaus Wildenhahn dort mit ihm verabredet. Als Klaus Wildenhahn sah, dass das Team die Kameraausr&uuml;stung mit dabei hatte, bestand er darauf, dass die Aufnahmen gleich vor Ort durchgef&uuml;hrt wurden. Im Nachhinein stellte sich heraus, dass es die letzten Filmaufnahmen mit Klaus Wildenhahn wurden, der am 9. August 2018 in Hamburg verstarb.
