Thereby, the performance of magnesiumion batteries can strongly depend on the morphology of the intercalation cathode. Moreover, high mass loadings are essential for commercialization. In this work the influence of different mass loadings are studied in addition to the impact of the particle size distribution of the active material. Therefore, a detailed continuum model is developed, which is able to describe the complex intercalation of magnesium into a Chevrel phase (CP) cathode. The model considers the thermodynamics, kinetics and interplay of the two energetically different intercalation sites of Mo</span><span>6</span><span>S</span><span>8</span><span>, which results from its unique crystal structure, as well as the impact of the desolvation on the electrochemical reactions and possible ion agglomeration. Ideal combinations of mass loading and electrolyte concentration as well as the desired CP particle size are determined for the state-of-the-art magnesium tetrakis(hexafluoroisopropyloxy)borate Mg[B(hfip)</span><span>4</span><span>]</span><span>2 </span><span>electrolyte.</span> </p>

<p>Cells (5 ×10<sup>5</sup> cells/mL) were incubated with two concentrations of (<strong>a</strong>) mTOR activator MHY1485, (<strong>b</strong>) AMPK inhibitor dorsomorphin, (<strong>c</strong>) ULK1/2 inhibitor MRT68921, (<strong>d</strong>) PI3K class III inhibitor wortmannin, (<strong>e</strong>) autophagosome-lysosome fusion inhibitor chloroquine, and (<strong>f</strong>) late-stage autophagy inhibitor bafilomycin A1 for 24 h. Autophagic flux was determined by measuring the fluorescence of RFP and GFP. The ratio of median fluorescence intensity of RFP to GFP (MFI) was normalized (nMFI) to the vehicle control (CTRL DMSO). Data are means ±SEM of 4 independent experiments, each carried out in duplicate.</p>

Abnormal GGGGCC repeat elongations, and in particular their folding states, are associated with amyotrophic lateral sclerosis and frontotemporal dementia. Due to methodological constraints however, most studies of G quadruplex structures are restricted to <em>in vitro</em> conditions. Evidence of how GGGGCC repeats form into G-quadruplexes <em>in vivo</em> is sparse. We devised a readout strategy, exploiting the sensitivity of <em>trans</em>-<em>cis</em> isomerization of cyanine dyes to local viscosity and sterical constraints. Thereby, folding states of cyanine-labeled RNA, and in particular G-quadruplexes, can be identified in a sensitive manner. The isomerization kinetics, monitored via fluorescence blinking generated upon transitions between a fluorescent <em>trans</em> isomer and a non-fluorescent <em>cis</em> isomer, was first characterized for RNA with GGGGCC repeats in aqueous solution using fluorescence correlation spectroscopy and transient state (TRAST) monitoring. With TRAST, monitoring the isomerization kinetics from how the average fluorescence intensity varies with laser excitation modulation characteristics, we could then detect folding states of fluorescently tagged RNA introduced into live cells.</p>

Blood cell differential count discretization modeling predicts survival in adults reporting to the emergency room: a retrospective cohort study
<p><strong>Objectives</strong>: to assess survival predictivity of baseline blood cell differential count (BCDC), discretized according to two different methods, in adults visiting the Emergency Room (ER) for illness or trauma over one-year. </p> <p><strong>Design</strong>: Retrospective cohort study of hospital records. </p> <p><strong>Setting</strong>: Tertiary care public hospital in northern Italy. </p> <p><strong>Participants</strong>: 11052 patients aged > 18 years, consecutively admitted to the ER in one year, and for whom BCDC collection was indicated by ER medical staff at first presentation.</p> <p><strong>Primary outcome</strong>: Survival was the referral outcome for explorative model development. Automated BCDC analysis at baseline assessed hemoglobin, red cell mean volume (MCV) and distribution-width (RDW), platelet distribution-width (PDW), plateletcrit (PCT), absolute red blood cells, white blood cells, neutrophils, lymphocytes, monocytes, eosinophils, basophils, and platelets. Discretization cutoffs were defined by Benchmark and Tailored methods. Benchmark cutoffs were stated on laboratory reference values (CLSI). Tailored cutoffs for linear, sigmoid-shaped and for U-shaped distributed variables were discretized by Maximally Selected Rank Statistics and by Optimal-Equal Hazard Ratio respectively. Explanatory variables (age, gender, ER admission during SARS-CoV2 surges, in-hospital admission) were analyzed using Cox multivariable regression. ROC curves were drawn by sum of Cox-significant variables for each method.</p> <p><strong>Results</strong>: Of 11052 patients (median age 67 years, IQR 51–81, 48% female), 59% (n=6489) were discharged and 41% (n=4563) were admitted in hospital. After a 306-day median follow up (IQR 208–417 days), 9455 (86%) patients were alive and 1597 (14%) deceased. Increased HRs were associated with age >73-years (HR=4.6 CI=4.0–5.2), in-hospital admission (HR=2.2 CI=1.9–2.4), ER admission during SARS-CoV2 surges (Wave-I HR=1.7 CI=1.5–1.9); Wave-II HR=1.2 CI=1.0–1.3). Gender, hemoglobin, MCV, RDW, PDW, neutrophils, lymphocytes and eosinophils counts were significant in overall. Benchmark-BCDC model included basophils and platelet count (AUROC 0.74). Tailored-BCDC model included monocyte counts and plateletcrit (AUROC 0.79).</p> <p><strong>Conclusions</strong>: baseline discretized BCDC provides meaningful insight regarding Emergency Room patients survival.</p>
