Laccases from Pleurotus ostreatus Applied to the Oxidation of Furfuryl Alcohol for the Synthesis of Key Compounds for Polymer Industry
<p>Laccases are oxidative enzymes with high synthetic potential. In this work, their value in biocatalysis is shown through the green and selective oxidation of furfuryl alcohol into furfural with the aid of mediators. The influence of different parameters, such as pH, enzyme/mediator composition, buffer type, cosolvent tolerance, and reaction times, is investigated. Under the optimal conditions, 20 mol % of TEMPO as mediator and 5.8 U mL<sup>−1</sup> of laccases POXC and POXA1b from <em>Pleurotus ostreatus</em>, quantitative production of furfural is attained after 16 h. POXC laccase stands out for its ability to catalyze the reaction at pH 6.5, whereas POXA1b is notable for its high stability. Furfural conversions reach excellent values (95 %) after 72 h using only 5 mol % of TEMPO at 100 mM. Furthermore, furfuryl alcohol bioamination is achieved by employing the amine transaminase from <em>Chromobacterium violaceum</em>, providing furfuryl amine, a key compound for the polymer industry, through a one-pot sequential approach.</p>
GDNF gene therapy for alcohol use disorder in male non-human primates
<p>These the minimal data sets that were utilized in analysis and interpretation for the manuscript "GDNF gene therapy treatment for alcohol use disorder" by Matthew Ford et al. These data sets were collected during a study to evaluate the utility of viral-based GDNF expression in the ventral tegmental area to reduce alcohol intake and prevent relapse in a primate model of alcohol use disorder, and ultimately the feasibility of GDNF as a gene therapy for alcohol use disorder. There are data sets on behavior, biochemical analyses, and cyclic voltammetry.</p>

<p><strong>Abstract</strong></p> <p>Fatty liver disease is a serious complication of childhood obesity. Calorie-restricted regimen (RCR) is one of the e ective therapy for this condition. Aim of the study was to evaluate the effect of lycopene-rich tomato sauce with oregano and basil extracts in obese children with fatty liver on RCR. 61 obese children with fatty liver were enrolled, 52 completed the study. A randomized cross over clinical trial was performed. Participants were assigned to RCR alone or with a supplement of lycopene-rich tomato juice for 60 days; subsequently, the groups were switched to the alternative regimen for the next 60 days. Reduction in BMI, HOMA-IR, cholesterol, triglycerides, liver size, and steatosis was more profound in tomato-supplemented group. Leptin decreased in both groups whereas adiponectin raised only after tomato supplementation. RCR is associated with the impaired engagement of T-cells glycolysis and proliferation, tomato-supplementation resulted in glycolytic metabolic activation of T-cells. Tomato juice ameliorates glucose and lipid metabolism in obese children, improve oxidative and inflammatory state and modulates the mitochondrial metabolism of T-cells contributing to a maintenance of a proper immune surveillance in children, impaired by RCR. Computational Studies of Human Class V Alcohol Dehydrogenase - The Odd Sibling
<p>A number of class I-VI ADH protein sequences were obtained from the UniProt, NCBI protein database, and Ensembl databases in October 2015. The sequences were aligned using the L-INS-i approach of MAFFT 7.266.</p> <p>The tree was generated using Phyml release 20151210. In the published figure, the tree was visualised as a radial phylogram in Dendroscope 3.</p>
Genetic architecture of alcohol consumption identified by a genotype-stratified GWAS, and impact on esophageal cancer risk in Japanese
<p><span>An East Asian-specific variant on <em>aldehyde</em> <em>dehydrogenase</em> <em>2</em> (<em>ALDH2</em> rs671, G>A) is the major genetic determinant of alcohol consumption. We performed an rs671 genotype-stratified genome-wide association study (GWAS) meta-analysis in up to 175,672 Japanese individuals to uncover additional loci associated with alcohol consumption in an rs671-dependent manner. Three loci (<em>GCKR</em>, <em>KLB</em>, and <em>ADH1B</em>)</span> <span>satisfied the genome-wide significance threshold in wild-type homozygotes (GG), whereas six loci (<em>GCKR</em>, <em>ADH1B</em>, <em>ALDH1B1</em>, <em>ALDH1A1</em>, <em>ALDH2</em>, and <em>GOT2</em>) did so in heterozygotes (GA). Of these, five loci showed genome-wide significant interaction with rs671. Genetic correlation analyses revealed ancestry-specific genetic architecture in heterozygotes. Subsequent polygenic risk scoring depicted interactions highlighted by stratified GWAS. Further, most discovered loci showed significant effects on risk of esophageal cancer, a representative alcohol-related disease, and multiple other phenotypes. Our results identify the genotype-specific genetic architecture of alcohol consumption and reveal its potential impact on alcohol-related disease risk.</span></p>
Mental health and alcohol use among patients attending a post-COVID-19 follow-up clinic: A cohort study.
<p>Study dataset</p> <p>Abstract</p> <p><strong>Background:</strong> Ongoing mental health problems following COVID-19 infection warrant greater examination. This study aimed to investigate psychiatric symptoms and problematic alcohol use among Long COVID patients.<br> <br> <strong>Methods: </strong>The study was conducted at the Mater Misericordiae University Hospital’s post-COVID-19 follow-up clinic in Dublin, Ireland. A prospective cohort study design was used encompassing assessment of patients’ outcomes at 2-4 months following an initial clinic visit (Time 1), and 7–14-month follow-up (Time 2). Outcomes regarding participants’ demographics, acute COVID-19 healthcare use, mental health, and alcohol use were examined.<br> <br> <strong>Results: </strong>The baseline sample’s (n = 153) median age = 43.5yrs (females = 105 (68.6%)). Sixty-seven of 153 patients (43.8%) were admitted to hospital with COVID-19, 9/67 (13.4%) were admitted to ICU, and 17/67 (25.4%) were readmitted to hospital following an initial COVID-19 stay. Sixteen of 67 (23.9%) visited a GP within seven days of hospital discharge, and 26/67 (38.8%) did so within 30 days. Seventeen of 153 participants (11.1%) had a pre-existing affective disorder. The prevalence of clinical range depression, anxiety, and PTSD scores at Time 1 and Time 2 (n = 93) ranged from 12.9% (Time 1 anxiety) to 22.6% (Time 1 PTSD). No statistically significant differences were observed between Time 1 and Time 2 depression, anxiety, and PTSD scores. Problematic alcohol use was common at Time 1 (45.5%) and significantly more so at Time 2 (71.8%). Clinical range depression, anxiety, and PTSD scores were significantly more frequent among acute COVID-19 hospital admission and GP attendance (30 days) participants, as well as among participants with lengthy ICU stays, and those with a previous affective disorder diagnosis.<br> <br> <strong>Conclusions: </strong>Ongoing psychiatric symptoms and problematic alcohol use in Long COVID populations are a concern and these issues may be more common among individuals with severe acute COVID-19 infection and /or pre-existing mental illness.</p>
