For further information on the instruments used please refer to the manuscript.</p> <p>The data was collected between April 2016 and March 2018 in Switzerland by the C-SURF study (<a href="http://www.c-surf.ch">www.c-surf.ch</a>). Participants were on average 25 years old when they answered the questionnaires. The final sample size used in the manuscript is 5516. Please note that the dataset contains 25 datasets created with multiple imputation, therefore there are no missing values in the dataset.</p> <p>The research protocol for this study was approved by the Human Research Ethics Committee of the Canton Vaud (Protocol No. 15/07). For further information on the instruments used please refer to the paper.</p> <p>The dataset contains data for three waves that was collected between September 2010 and March 2018 in Switzerland by the C-SURF study (<a href="http://www.c-surf.ch/">www.c-surf.ch</a>). Participants were on average 20 years old at wave 1, 21 at wave 2 and 25 at wave 3 when they answered the questionnaires. The final sample size used in the paper is 4746 after excluding those that did not reply to a questionnaire or to a variable of interest for the main analysis.</p> <p>The research protocol for this study was approved by the Human Research Ethics Committee of the Canton Vaud (Protocol No. 15/07). Defective HNF4alpha-dependent gene expression as a driver of hepatocellular failure in alcoholic hepatitis [Suppl Data]
<p>Alcoholic hepatitis (AH) is a life-threatening condition characterized by profound hepatocellular dysfunction for which targeted treatments are urgently needed. Identification of molecular drivers is hampered by the lack of suitable animal models. By performing RNA sequencing in livers from patients with different phenotypes of alcohol-related liver disease (ALD), we show that the development of AH is characterized by the defective activity of liver-enriched transcription factors (LETFs). TGFb1is a key upstream transcriptome regulator in AH and induces the use of HNF4aP2 promoter in hepatocytes, which results in defective metabolic and synthetic functions. Gene polymorphisms in LETFs including HNF4aare not associated with the development of AH. In contrast, epigenetic studies show that AH livers have profound changes in DNA methylation state and chromatin remodeling, affecting HNF4a-dependent gene expression. We conclude that targeting TGFb1and epigenetic drivers that modulate HNF4a-dependent gene expression could be beneficial to improve hepatocellular function in patients with AH.</p>

Alcohol and Drug Abuse Research Program (ADARP) Dataset
<p>The Alcohol and Drug Abuse Research Program (ADARP) dataset was collected as a part of a pilot study that aimed to discover how the daily experiences of patients diagnosed with alcohol use disorder (AUD) correspond with physiological biomarkers of stress. Each participant completed three components: 1) a daily diary study using ecological momentary assessment (EMA) of self-reported emotions, cravings, and stress via a web-based survey, prompted 4 times daily for up to 14 days; 2) continuous monitoring of stress with an Empatica E4 wristband that captured, in real-time, continuous physiological markers of stress, including heart rate (HR), skin conductance or electrodermal activity (EDA), skin temperature, and bodily movements; and 3) structured qualitative interviews to assess daily alcohol use, using a timeline follow-back calendar, and to validate self-reported and physiological markers of stress. With the proposed study, we aimed to address three research objectives. Aim 1 examines the fluctuations and associations among the self-reported emotions, alcohol-related cravings, and stress assessed in the EMA component. Analysis for Aim 1 centered on concurrent and lagged associations among negative affect, cravings, and stress. Aim 2 uses the continuous monitoring data acquired via the wearable device to visualize and describe fluctuations among the physiological measures of stress. Analysis for this aim focused on determining the degree of synchrony among the different physiological indicators. Aim 3 combines the EMA and continuous monitoring components in order to determine if self-reported changes in alcohol-related cravings can be predicted from physiological measures of stress. </p> <p>ADARP dataset was collected from participants suffering from alcohol use disorder (AUD) and receiving treatment at an outpatient treatment agency during 2019 - 2020, at Washington State University (WSU), Pullman, WA, USA. Funding for the original study was provided by the Alcohol and Drug Research Program (ADARP) of Washington State University. This investigation was supported in part by funds provided for medical and biological research by the State of Washington Initiative Measure No. 171.</p> <p>The sensor and EMA survey data are made public to facilitate further research into this topic. Heterogeneous Biocatalytic Reduction of 5-(Hydroxy)methyl Furfural Using two Co-immobilised Alcohol Dehydrogenases
<p>Biocatalyst heterogenisation may enable robust processes that can be applied in biorefineries to selectively valorise highly functionalised platform chemicals. In this work, we co-immobilise two dehydrogenases and successfully apply them in the selective reduction of 5-hydroxymethylfurfural (HMF) to 2,5-bis(hydroxymethyl) furan (BHMF) with efficient <em>in situ</em> cofactor regeneration. First, we select the best enzyme candidates (an alcohol dehydrogenase from <em>Escherichia coli</em> together with a thermostable glucose dehydrogenase from <em>Bacillus subtilis</em>) and then screen a variety of carriers and chemistries to find the optimal individual immobilisation protocols for each dehydrogenase. As a result, methacrylate carriers (Purolite™) functionalised with either aldehydes or with epoxy and cobalt-chelate groups co-immobilise both enzymes in high yields with a sufficient activity recovery (>20%). These optimal heterogeneous biocatalysts enable the quantitative bio-reduction of HMF to BHMF with >99% selectivity in only fifteen minutes, exhibiting an outstanding reusability of >15 batch cycles with a total volumetric productivity of ∼5 g L<sup>−1</sup> h<sup>−1</sup> of BHMF. Preliminary experiments on a semipreparative scale with HMF loadings of 40 mM also reach high product conversions (86%). Overall, the judicious selection of enzymes, carriers and reaction conditions enables the design of robust biocatalysts that may contribute to paving the way to the valorisation of highly functionalised chemicals in biorefineries.</p>

Unveiling the role of miRNAs from MSC-EVs in neuroinflammation and behavioral impairments induced by chronic alcohol consumption
<p><strong>Part of the project: "Unveiling the role of miRNAs from MSC-EVs in neuroinflammation and behavioral impairments induced by chronic alcohol consumption."</strong></p> <p>This dataset contains small RNA sequencing data derived from extracellular vesicles (EVs) isolated from human adipose tissue-derived mesenchymal stem cells (MSC-EVs). The data was generated to study the potential therapeutic effects of repeated intravenous administration of MSC-EVs on neuroinflammation, cognitive dysfunction, and addictive-like behaviors induced by chronic alcohol consumption.</p> <p>Total RNA from MSC-EVs was extracted using the Total Exosome RNA Isolation Kit (Invitrogen). Small RNA libraries were constructed with the Small RNA-Seq Library Prep Kit (Lexogen) and sequenced on the Illumina NextSeq 550 platform with a 1 x 150 bp read length. Additionally to excess weight loss, significant improvement in comorbidities is an established benefit after bariatric operations. In recent years, non-alcoholic fatty liver disease (NAFLD) has emerged as one of these comorbidities. Herein we will review the data supporting the positive effect of bariatric surgery in patients with NAFLD.</p>
