Heat stress responses of the kelp Laminaria digitata (Phaeophyceae) across Northeast Atlantic populations: growth, biochemistry, chlorophyll fluorescence, pigments

<p>To understand the thermal plasticity of a coastal foundation species across its latitudinal distribution, we assess physiological responses to high temperature stress in the kelp <i>Laminaria digitata</i> in combination with population genetic characteristics and relate heat resilience to genetic features and phylogeography. We hypothesize that populations from Arctic and cold-temperate locations are less heat resilient than populations from warm distributional edges. Using meristems of natural <i>L. digitata</i> populations from six locations ranging between Kongsfjorden, Spitsbergen (79°N), and Quiberon, France (47°N), we performed a common-garden heat stress experiment applying 15°C to 23°C over eight days. We assessed growth, photosynthetic quantum yield, carbon and nitrogen storage, and xanthophyll pigment contents as response traits. Population connectivity and genetic diversity were analysed with microsatellite markers. Results from the heat stress experiment suggest that the upper temperature limit of <i>L. digitata</i> is nearly identical across its distribution range, but subtle differences in growth and stress responses were revealed for three populations from the species' ecological range margins. Two populations at the species' warm distribution limit showed higher temperature tolerance compared to other populations in growth at 19°C and recovery from 21°C (Quiberon, France), and photosynthetic quantum yield and xanthophyll pigment responses at 23°C (Helgoland, Germany). In <i>L. digitata</i> from the northernmost population (Spitsbergen, Norway), quantum yield indicated the highest heat sensitivity. Microsatellite genotyping revealed all sampled populations to be genetically distinct, with a strong hierarchical structure between southern and northern clades. Genetic diversity was lowest in the isolated population of the North Sea island of Helgoland and highest in Roscoff in the English Channel. All together, these results support the hypothesis of moderate local differentiation across <i>L. digitata</i>'s European distribution, whereas effects are likely too weak to ameliorate the species' capacity to withstand ocean warming and marine heatwaves at the southern range edge.</p>

Estimates of mixed lyer gross primary productivity and photophysiology based on underway Lagrangian measurements of the dissolved chlorophyll fluorescence of phytoplankton usung Fast Repetition Rate Fluorometry (FRRF)

GPP was estimated on the P1706 cruise based on the photo-physiology of the mixed-layer phytoplankton community measured by FRRF. Shipboard measurements were made using a bench-top FastAct 2+ Fast TRAKA instrument (Chelsea, UK) plumbed into the ship’s running seawater system. Photosynthesis versus irradiance (P vs. E) curves were run continuously on a ~45 min sampling interval.

Data from: Comparison of solar-induced chlorophyll fluorescence, light-use efficiency, and process-based GPP models in maize

Accurately quantifying cropland gross primary production (GPP) is of great importance to monitor cropland status and carbon budgets. Satellite-based light-use efficiency (LUE) models and process-based terrestrial biosphere models (TBMs) have been widely used to quantify cropland GPP at different scales in past decades. However, model estimates of GPP are still subject to large uncertainties, especially for croplands. More recently, space-borne solar-induced chlorophyll fluorescence (SIF) has shown the ability to monitor photosynthesis from space, providing new insights into actual photosynthesis monitoring. In this study, we examined the potential of SIF data to describe maize phenology and evaluated three GPP modeling approaches (space-borne SIF retrievals, a LUE-based Vegetation Photosynthesis Model (VPM), and a process-based Soil Canopy Observation of Photochemistry and Energy flux (SCOPE) model constrained by SIF) at a maize (Zea mays L.) site in Mead, Nebraska, USA. The result shows that SIF captured the seasonal variations (particularly during the early and late growing season) of tower-derived GPP (GPP_EC) much better than did satellite-based vegetation indices (enhanced vegetation index, EVI and land surface water index, LSWI). Consequently, SIF was strongly correlated with GPP_EC than were EVI and LSWI. Evaluation of GPP estimates against GPP_EC during the growing season demonstrated that all three modeling approaches provided reasonable estimates of maize GPP, with Pearson's correlation coefficients (r) of 0.97, 0.94, and 0.93 for the SCOPE, VPM, and SIF models, respectively. The SCOPE model provided the best simulation of maize GPP when SIF observations were incorporated through optimizing the key parameter of maximum carboxylation capacity (Vcmax). Our results illustrate the potential of SIF data to offer an additional way to investigate the seasonality of photosynthetic activity, to constrain process-based models for improving GPP estimates, and to reasonably estimate GPP by integrating SIF and GPP_EC data without dependency on climate inputs and satellite-based vegetation indices.

Cell size, chlorophyll fluorescence and cadmium bioaccumulation between wild-type and mutant strains of Chlamydomonas reinhardtii upon exposure to cadmium

<p>Heavy metal contamination presents a constant threat to biological systems. Simultaneously, heavy metals have become one of the major contaminants in the aquatic ecosystem. In this regard, the investigation of heavy metal-tolerance genes in algae is relevant. <i>Chlamydomonas reinhardtii</i> is a unicellular green alga, and an excellent model organism used in heavy metal studies. In <i>C. reinhardtii</i>, a novel gene designated as Cia7, was hypothesized to play a role in heavy metal homeostasis due to CIA7's conserved cysteine-residue motif. This study compared two strains of <i>C. reinhardtii</i>, cc4425, the wild-type possessing the functional CIA7 protein and cc5013, the mutant strain possessing the disrupted cia7- gene. The hypothesis was that the expression of Cia7 contributes to an increased cadmium (Cd)-tolerance in <i>C. reinhardtii</i>. The Cd-tolerance would be described by physiological markers of microalgae health, and by intracellular accumulation of the metal. Methods: The objectives of this study were (1) to compare chlorophyll fluorescence and cell size in cc4425 and cc5013 exposed to Cd<sup>2+</sup>, and (2) to compare Cd<sup>2+</sup> bioaccumulation in cc4425 and cc5013 strains in different growth media. Flow cytometry, and inductively coupled plasma optical emission spectrometry (ICP-OES) analysis were performed. Results: There was no significant statistical difference in Cd<sup>2+</sup> bioaccumulation between the two strains, cc4425 and cc5013, regardless of growth media. However, a statistically significant difference in Cd<sup>2+</sup> bioaccumulation (p&lt;0.0001) was determined between the media (with acetate and without acetate). The cia7- mutant, cc5013 was found to be more susceptible to a Cd<sup>2+</sup>-induced decrease in chlorophyll fluorescence and had a reduced cell size compared to cc4425, the wild-type strain. Conclusions: These observed differences between the strains suggest that CIA7's biological activity could play a direct or indirect role in increasing Cd tolerance in <i>C. reinhardtii</i>.</p>

Data for: The photosynthetic response of spectral chlorophyll fluorescence differs across species and light environments in a boreal forest ecosystem

<p>Chlorophyll fluorescence can serve as a proxy of photosynthesis in boreal forests. When sustained non-photochemical quenching (NPQS) relaxes towards summer, leaf chlorophyll fluorescence (ChlF) emission increases along with photosynthesis. Yet, other physical and physiological factors can also leave a measurable imprint on the fluorescence emission spectra, and disrupt this relationship.</p> <p>We measured leaf-level spectral ChlF of Scots pine, Norway spruce and lingonberry exposed to contrasting light environments throughout the spring recovery of photosynthesis, simultaneously with a series of photosynthetic, biochemical and morphological traits. Correlations between traits and ChlF spectral components were analyzed to identify the mechanisms underlying both the spatial variation found between species and light environments, and the temporal variation found across the study period.</p> <p>Spatially, we found evidence of baseline differences in leaf-level ChlF magnitude, which we attribute to species- and light environment-specific changes in leaf morphology. Temporally, ChlF magnitude followed the relaxation of NPQS  towards summer, but only in upper canopy foliage and lingonberry, suggesting a seasonal compensation effect between sustained photochemical quenching (PQS) and NPQS, potentially decoupling the seasonal relationship between ChlF and photosynthesis in shaded foliage. Finally, we show subtle changes in the shape of the ChlF spectra that took place independently of chlorophyll concentration dynamics, pointing to the complexity of NPQs which can involve structural rearrangements in the thylakoids and changes in the relative contribution of PSI to emitted ChlF.</p> <p>We conclude that the diversity of species and light environments found within an ecosystem generates a baseline level of variation in leaf spectral ChlF as well as contrasting seasonal photosynthetic acclimation patterns. These sources of variability should be taken into account when developing quantitative models for the interpretation of ChlF data, in particular for applications involving high resolution SIF imaging systems capable of resolving different plant individuals and their parts.</p>
