Cereal yield response to climate change in key grain producing areas of the Northern Hemisphere

<p>Vulnerability mapping is a field of increasing importance as we search for an ecologically and economically sustainable<br> climate-smart land-use system, which is necessary for the development of green infrastructure and agricultuVulnerability<br> mapping is a growing field with increasing importance in developing an ecologically and economically sustainable climate-smart&nbsp;land-use system, which may serve the advancement of green infrastructure and agricultural production. The<br> transformation of the climatic regime has an undeniable impact on plant production, but we rarely have long enough data<br> series to scrutinize the unfolding effects of the microregional patterns of the global climate-cereal yield relationship. To fill this<br> gap, I will analyze three regions of the Northern Hemisphere under continental climate, which represent key landscapes<br> concerning global food production and security, contributing a multi-way knowledge transfer through training, teaching, and<br> different form of communication activities. The relationship between climate indices, maize, and wheat yields will be analyzed<br> applying high geographical resolution data from Northern China, Hungary, and northern states of the US Midwest at a<br> landscape scale by K&ouml;ppen-Geiger zones using linear, local, and spatial regression and bootstrap resampling tests and<br> developed data visualization techniques. This offers a unique opportunity to comparatively analyze the differences in<br> climate-cereal yield associations by landscape types and regions. The expected results will provide highly supportive factors<br> for planning and managing an ecologically and economically sustainable land-use system. The intended publications,<br> lectures, and collaborative actions will have a positive impact on domestic and EU research. The planned training and the<br> expected results of the action will further strengthen my career opportunities and put me in a position where I will be<br> considered for a tenured position at a research institution in Hungary.</p>

Sugar signal manipulation by T6P for yield on wheat grain - whole grain RNA-seq

<p>Trehalose 6-phosphate (T6P) is a powerful internal sugar signal in plants yet cannot be directly added nor<br>fully genetically controlled. A timed microdose of a plant-permeable T6P signalling precursor, DMNB-T6P, causes substantial yield improvements. This repository contains normalised counts table for a RNA-seq of wheat (Triticum aestivum) whole grains after spray with mock/DMNB-T6P.</p> <p>DESeq2 normalised count table for raw reads deposited in ncbi SRA under Bioproject PRJNA1007614. Related manuscript in press in Nature Biotechnology.</p> <p>&nbsp;</p> <p>Methods extract below:</p> <p>(...) Following RNA integrity analysis and quantitation (Bioanalyser; Agilent, USA), poly-A enriched cDNA libraries were generated and&nbsp;sequenced on an Illumina Novaseq 6000 sequencing platform generating 30&ndash;50 million 150 bp paired-end reads per sample. Low-quality reads and adaptor sequences were removed with Trimmomatic (trimmomatic-0.39.jar PE ILLUMINACLIP:TruSeq3-PE.fa:2:30:10:2:True TRAILING:30 MINLEN:40). The reads were aligned to the wheat reference genome (<em>Triticum aestivum</em> iwgsc_refseqv2.1&nbsp;using HISAT2/2.2.1-foss-2019b with default parameters&nbsp;and converted to Bam format with SAMtools. Gene abundance was quantified using featureCounts&nbsp;with the High Confidence iwgsc_refseqv2.1 annotation (counting only primary alignments of read pairs with a quality cut-off of 10).&nbsp;Raw counts were normalized using the trimmed mean of M-values method by DESeq2.</p>

Data from: Validation of grain yield QTL from soft winter wheat using a CIMMYT spring wheat panel

Validation of quantitative trait loci (QTLs) is an essential step in marker-assisted breeding. The objectives of this study were to validate grain yield (GY) QTLs previously identified in soft red winter wheat (Triticum aestivum L.) through biparental and association mapping using the spring wheat association mapping initiative (WAMI) panel from CIMMYT, Mexico, and to identify allele combinations of the validated QTLs that resulted to the highest GY. Linked single-nucleotide polymorphisms for IWA3560 (3A), IWA1818 (4B), and IWA755 (6B) were significantly associated (P &lt; 0.001) with GY, grain number, and thousand-grain weight in the WAMI. Lines possessing the favorable allele for the QTL at the 3A, 4B, and 6B loci (ACG allele combination) validated on the WAMI had the highest mean GY at 4.55 t ha−1, but three other haplotypes (ACA, GCA, and GCG) differing by one or two alleles in the validated QTL regions were not significantly different. These results validate GY QTLs across winter and spring wheat through genome-wide association analysis and further demonstrate the potential for pyramiding favorable alleles for the genetic improvement of wheat breeding populations.

Datasets generated in the ConvAE-RF modelling of grain yield in the mid-lower Yangtze plains
