eDNA metabarcoding in lakes to quantify influences of landscape features and human activity on aquatic invasive species prevalence and fish community diversity
<p>Aim: Our goal was to use eDNA metabarcoding to characterize fish community diversity, detect aquatic invasive species (AIS), and assess how measures of community (or AIS) diversity are influenced by lake physical and environmental covariates, measures of hydrological connectivity, and human accessibility.<br> Location: Michigan, USA.<br> Methods: eDNA samples collected from 22 lakes were sequenced using two mitochondrial gene regions (12S and 16S rRNA). Metabarcoding data were compared to traditional fisheries survey data for a subset of lakes, and data from all 22 lakes were combined with environmental information to identify significant associations with community diversity and AIS relative abundance.<br> Results: Occupancy modeling indicated that detection probabilities were generally higher with eDNA than traditional fisheries gear. Measures of connectivity with upstream aquatic habitats were positively associated with both AIS relative abundance and fish species diversity. We also demonstrate the use of spatial interpolation methods to map distributions of species diversity and AIS relative abundance within lakes.<br> Conclusions: eDNA metabarcoding methods provided information on the composition and diversity of fish assemblages and the presence of AIS in freshwater lakes that varied greatly in drainage connectivity and anthropogenic development. Our case study identified associations between environmental covariates and fish diversity or AIS relative abundance across lakes. This information is of particular importance given increasing anthropogenic disturbance, invasive species spread, and associated declines in aquatic biodiversity. Incorporating eDNA metabarcoding as a supplement to traditional fisheries surveys will permit managers to identify greater numbers of taxa, including early detection of AIS, with less field effort and fish mortality. Further, eDNA methods may more accurately identify physical and biological features that correlate with diversity and abundance, and allow agencies to more effectively direct AIS management activities. </p>
Data from: Active and reactive behaviour in human mobility: the influence of attraction points on pedestrians
Human mobility is becoming an accessible field of study thanks to the progress and availability of tracking technologies as a common feature of smart phones. We describe an example of a scalable experiment exploiting these circumstances at a public, outdoor fair in Barcelona (Spain). Participants were tracked while wandering through an open space with activity stands attracting their attention. We develop a general modeling framework based on Langevin Dynamics, which allows us to test the influence of two distinct types of ingredients on mobility: reactive or context-dependent factors, modelled by means of a force field generated by attraction points in a given spatial configuration, and active or inherent factors, modelled from intrinsic movement patterns of the subjects. The additive and constructive framework model accounts for some observed features. Starting with the simplest model (purely random walkers) as a reference, we progressively introduce different ingredients such as persistence, memory, and perceptual landscape, aiming to untangle active and reactive contributions and quantify their respective relevance. The proposed approach may help in anticipating the spatial distribution of citizens in alternative scenarios and in improving the design of public events based on a facts-based approach.
Data from: Patterns and limitations of urban human mobility resilience under the influence of multiple types of natural disaster
Natural disasters pose serious threats to large urban areas, therefore understanding and predicting human movements is critical for evaluating a population's vulnerability and resilience and developing plans for disaster evacuation, response and relief. However, only limited research has been conducted into the effect of natural disasters on human mobility. This study examines how natural disasters influence human mobility patterns in urban populations using individuals' movement data collected from Twitter. We selected fifteen destructive cases across five types of natural disaster and analyzed the human movement data before, during, and after each event, comparing the perturbed and steady state movement data. The results suggest that the power-law can describe human mobility in most cases and that human mobility patterns observed in steady states are often correlated with those in perturbed states, highlighting their inherent resilience. However, the quantitative analysis shows that this resilience has its limits and can fail in more powerful natural disasters. The findings from this study will deepen our understanding of the interaction between urban dwellers and civil infrastructure, improve our ability to predict human movement patterns during natural disasters, and facilitate contingency planning by policymakers.
Data from: Habitat differences influence genetic impacts of human land use on the American beech (Fagus grandifolia)
Natural reforestation after regional forest clearance is a globally common land-use sequence. The genetic recovery of tree populations in these recolonized forests may depend on the biogeographic setting of the landscape, for instance whether they are in the core or in the marginal part of the species' range. Using data from 501 individuals genotyped across 7 microsatellites, we investigated whether regional differences in habitat quality affected the recovery of genetic variation in a wind-pollinated tree species, American beech (Fagus grandifolia) in Massachusetts. We compared populations in forests that were recolonized following agricultural abandonment to those in remnant forests that have only been logged in both central inland and marginal coastal regions. Across all populations in our entire study region, recolonized forests showed limited reduction of genetic diversity as only observed heterozygosity was significantly reduced in these forests (H O = 0.520 and 0.590, respectively). Within inland region, this pattern was observed, whereas in the coast, recolonized populations exhibited no reduction in all genetic diversity estimates. However, genetic differentiation among recolonized populations in marginal coastal habitat increased (F st logged = 0.072; F st secondary = 0.249), with populations showing strong genetic structure, in contrast to inland region. These results indicate that the magnitude of recovery of genetic variation in recolonized populations can vary at different habitats.
Data from: Human activities influence the occupancy probability of mammalian carnivores in the Brazilian Caatinga
The Caatinga is a semi-arid domain, characterized by reduced humidity and high rates of anthropogenic impact. In addition to the low availability of water, carnivorous mammals are still exposed to a number of threats related to landscape modifications. We used data from camera traps and occupancy models to investigate the habitat use by carnivores in an area of Caatinga in northeastern Brazil. We found a negative correlation between the presence of wind farms and the occupancy probability of the jaguar, and a positive relationship with the presence of the jaguarundi. Puma and jaguarundi occupied primarily sites near watercourses, whereas the occupancy of the crab-eating fox was correlated positively with the presence of poachers. The ocelot was detected more frequently at sites distant from human settlements, whereas the jaguar was detected more often in areas far from wind farms. We found a negative correlation between the distance of water and the detection of the ocelot. The detection of the crab-eating fox was influenced positively by the detection of cattle. In addition to the negative influence of some anthropic activities, our results indicate that water is a very important resource for species, and the few permanent sources of this resource available in the area must be preserved. The replication of our research in other systems, worldwide, that are experiencing similar pressures, should permit a systematic evaluation of the management and conservation strategies needed to rebuild or maintain populations, restore ecosystems, and support conservation policies in human-altered landscapes.
