Linking behavioural thermoregulation, boldness and individual state in male Carpetan rock lizards

Mechanisms affecting consistent inter-individual behavioural variation (i.e. animal personality) are of wide scientific interest. In poikilotherms, ambient temperature is one of the most important environmental factors with a direct link to a variety of fitness-related traits. Recent empirical evidence suggests that individual differences in boldness are linked to behavioural thermoregulation strategy in heliothermic species, as individuals are regularly exposed to predators during basking. Here, we tested for links between behavioural thermoregulation strategy, boldness and individual state in adult males of the high mountain Carpetan rock lizard (Iberolacerta cyreni). Principal Component Analysis revealed the following latent links in our data: (i) a positive relationship of activity with relative limb length and colour brightness (PC1, 23% variation explained), (ii) a negative relationship of thermoregulatory precision with parasite load and risk-taking (PC2, 20.98% variation explained) and (iii) a negative relationship between preferred body temperature and relative limb length (PC3, 19.23% variation explained). We conclude that differences in boldness and behavioural thermoregulatory strategy could be explained by both stable and labile state variables. The moderate link between behavioural thermoregulatory strategy and risk-taking personality in our system is plausibly the result of differences in reproductive state of individuals or variation in ecological conditions during the breeding season.

Mouse spontaneous behavior reflects individual variation rather than estrous state

Repository containing datasets obtained for Levy et al. 2023, available online here: https://doi.org/10.1016/j.cub.2023.02.035

Github link to related analysis code: https://github.com/dattalab/spontaneous-behavior-reflects-individuality-not-estrous

Abstract

Behavior is shaped by both the internal state of an animal and its individual behavioral biases. Rhythmic variation in gonadal hormones during the estrous cycle is a defining feature of female internal state, one that regulates many aspects of sociosexual behavior. However, it remains unclear whether estrous state influences spontaneous behavior, and if so how these effects might relate to individual behavioral variation. Here we address this question by longitudinally characterizing the open field behavior of female mice across different phases of the estrous cycle, using unsupervised machine learning to decompose spontaneous behavior into its constituent elements1-4. We find that each female mouse exhibits a characteristic pattern of exploration that uniquely identifies it as an individual across many experimental sessions; in contrast, estrous state only negligibly impacts behavior, despite its known effects on neural circuits that regulate action selection and movement. Like female mice, male mice exhibit individual specific patterns of behavior in the open field; however, the exploratory behavior of males is significantly more variable than that expressed by females both within and across individuals. These findings suggest an underlying functional stability to the circuits that support exploration in female mice, reveal a surprising degree of specificity in individual behavior, and provide empirical support for the inclusion of both sexes in experiments querying spontaneous behaviors.

Behavioral DataFrames: holds the behavioral syllable data used for the analysis described in the manuscript:

mean_df_female / scalar_df_female - main female dataset

mean_df_male / scalar_df_male - main male dataset

mean_df_female_control / mean_df_male_control - datasets for male/female control experiment presented in Fig S3.

Model files:

Additional MoSeq model files used for generating FigS2A and FigS2H.

This dataset contains preprocessed resting state fMRI data (.nii.gz) with accompanying confound files (.tsv) from the Single Individual volunteer for Multiple Observations across Networks (SIMON; http://fcon_1000.projects.nitrc.org/indi/retro/SIMON.html) dataset that has been minimally preprocessed using the NeuroImaging Analysis Kit (NIAK; http://niak.simexp-lab.org/build/html/PREPROCESSING.html). Preprocessing steps included: (1) Slice timing correction; (2) Estimation of rigid-body motion in fMRI runs, both within- and between sessions; (3) Linear or non-linear coregistration of the structural scan in stereotaxic space; (4) Individual coregistration between structural and functional scans; (5) Resampling of functional scans in stereotaxic space. Confound files (.tsv) have been included for purposes of scrubbing and regression of confounds using NIAK or other analysis software, allowing for further processing without conflicts.
