Stepwise construction of dynamic microscale concentration gradients around hydrogel-encapsulated cells in a microfluidic perfusion culture device
<p><span>Inside living organisms, concentration gradients dynamically change as biological processes progress. Therefore, methods to construct dynamic microscale concentration gradients in a spatially controlled manner are needed to provide more realistic research environments. Here, we report a novel method for the construction of dynamic microscale concentration gradients in a stepwise manner around cells in micropatterned hydrogel. In our method, cells are encapsulated in a photodegradable hydrogel formed inside a microfluidic perfusion culture device, and perfusion microchannels are then fabricated in the hydrogel by micropatterned photodegradation. The cells in the micropatterned hydrogel can then be cultured by perfusing culture medium through the fabricated microchannels. By using this method, we demonstrate the simultaneous construction of two dynamic concentration gradients, which allowed us to expose the cells encapsulated in the hydrogel to a dynamic microenvironment.</span></p>
Data from: In silico study of the role of cell growth factors in photosynthesis using a virtual leaf tissue generator coupled to a microscale photosynthesis gas exchange model
Computational tools that allow in silico analysis of the role of cell growth and division on photosynthesis are scarce. We present a freely available tool that combines a virtual leaf tissue generator and a two-dimensional microscale model of gas transport during C3 photosynthesis. A total of 270 mesophyll geometries were generated with varying degree of growth anisotropy, growth extent and extent of schizogenous airspace formation in the palisade mesophyll. The anatomical properties of the virtual leaf tissue and microscopic cross sections of actual leaf tissue of tomato (Solanum lycopersicum L.) were statistically compared. Model equations for transport of CO2 in the liquid phase of the leaf tissue were discretized over the geometries. The virtual leaf tissue generator produced a leaf anatomy of tomato that was statistically similar to real tomato leaf tissue. The response of photosynthesis to intercellular CO2 predicted by a model that used the virtual leaf tissue geometry compared well with measured values. The results indicate that the light-saturated rate of photosynthesis was influenced by interactive effects of extent and directionality of cell growth and degree of airspace formation through the exposed surface of mesophyll per leaf area. The tool could be used further in investigations of improving photosynthesis and gas exchange in relation to cell growth and leaf anatomy.
Data from: Feeding preference as a main determinant of microscale patchiness among terrestrial nematodes
Soil biota are responsible for essential ecosystem services such as carbon storage, nutrient cycling and water retention. However, assessment of the condition of soil biota is hampered by an overwhelming level of diversity. With representatives in all trophic levels of the food web, nematode communities can be used as bio-indicators. Accurate assessment of nematode assemblages requires insight in the distribution of specimens with distinct food preferences. With the availability of taxon-specific quantitative-PCR assays, distribution patterns of multiple nematode groups can be investigated simultaneously. Here, microscale patchiness of 45 nematode taxa was studied on 12 sampling sites (each with four adjacent microplots) located on arable fields or semi-natural grasslands ('system'), and on marine-, river clay or sandy soils ('soil type'). From each microplot five composite samples were collected. Contrary to our expectations, an increase of the number of cores per composite sample did not result in more accurate measurements, and apparently the levels of microscale patchiness of the taxa are low compared to what has been reported for oligophagous plant-parasites. System and soil type did not affect microscale distribution. To investigate the level of patchiness in more detail, detection probability (DP) and variability of abundances were calculated. Common and widespread bacterivorous and fungivorous taxa had DP ≥ 90%, confirming low level of microscale patchiness. With DPs of 40-70%, predators and most omnivores showed degrees of local clustering. An overview of mean variabilities of abundances is presented that offers insight in how feeding preferences impact the microscale distribution both between and within trophic groups.
Data from: A computational model of flow between the microscale respiratory structures of fish gills
The gills of most teleost fishes are covered by plate-like structures, the secondary lamellae, that provide the bulk of the respiratory surface area. Water passing over the secondary lamellae exchanges gases with blood passing through the secondary lamellae, forming a system that has served as a classic model of counter-current exchange. In this study, a computational model of flow around the secondary lamellae is used to examine the hydrodynamic consequences of changes to the lamellar morphology. Consistent with previous studies, the interlamellar distance is found to strongly affect the hydrodynamic resistance of the gills. However, the presence of a small gap between the tips of the secondary lamellae is found to have a similar strong effect on the hydrodynamic resistance and flow patterns within the gills. The results from this model have been generally formulated, allowing the calculation of the hydrodynamic resistance for measured morphometric parameters. These results provide a new basis for comparing theoretical predictions of the gill resistance with measured values, and provide a general model for examining the diversity gill morphologies observed in teleost fishes.

Photodynamic Ocular Drug Delivery System with Optical Coherence Tomography Oriented Microscale Robots
<p>Retinal diseases are the most common reason for irreversible blindness worldwide. Common retinal diseases, such as diabetic retinopathy, macular degeneration, retinal vein occlusion, and non-infectious uveitis, mostly require intravitreal steroid injections in clinical management. Unfortunately, intravitreal steroid injections lead to other important ocular problems, such as elevation of intraocular pressure, intravitreal infections, and the progression of cataracts. Because of these reasons, targeted drug delivery systems are necessary for the medical treatment of retinal diseases. PHOTodynamic Ocular Drug delivery system with Optical Coherence Tomography Oriented microscale Robots (PHOTODOCTOR) project combines three novel technologies to solve this important clinical problem. The first technological advancement for the project is dexamethasone-saturated hyaluronic acid (HA) hydrogels with organo-ruthenium complexes, which enables controllable degradation under visible light exposure. With the help of visible light-controlled degradation, dexamethasone will be released with the light pulses. Then the hydrogels will be supplemented with superparamagnetic iron oxide nanoparticles (SPIONs), and 3D printed as helical micro-swimmers. Insertion of the SPIONs will help the magnetic actuation of the hydrogel-based microrobots in the intraocular space. Lastly, SPION-supplemented HA hydrogels will be observed with high-resolution optical coherence tomography (OCT) system in the intraocular space to guide them in diseased areas of the retina. While HA-based structure increases penetration in the vitreous and helps the attachment on the retina, SPIONs will increase the visibility of the microrobots in OCT imaging. In this way, a magnetic-driven, visible light-triggered intraocular drug release system with OCT guidance will be produced, and it could be used not only for intravitreal steroid delivery but also for the treatment of retinal tumours and other retinal problems.</p>

Remote sensing of Electrification, Lightning, And Mesoscale/microscale Processes with Adaptive Ground Observations (RELAMPAGO) Lightning Mapping Array (LMA) V1
The Remote sensing of Electrification, Lightning, And Mesoscale/microscale Processes with Adaptive Ground Observations (RELAMPAGO) Lightning Mapping Array (LMA) was an 11-station, ground-based network located in north-central Argentina from November 2018 to April 2019 in support of the RELAMPAGO field campaign. The RELAMPAGO campaign aimed to characterize the atmospheric conditions and terrain effects that facilitate the initiation and growth of intense weather systems in this region of South America. The LMA maps Very High Frequency (VHF) emissions from lightning in three dimensions. These emissions have also been grouped, temporally and spatially, into individual flashes, and the flash characteristics analyzed to produce gridded products. The dataset was produced by NASA Marshall Space Flight Center (MSFC), via an agreement with the National Oceanic and Atmospheric Administration (NOAA), in order to serve as a validation dataset for the Geostationary Lightning Mapper (GLM). These LMA data are available from November 8, 2018 through April 20, 2019 in ASCII, HDF5, and netCDF-4 format.

Connecting Microscale And Macroscale Damage Models In A Bayesian Framework for Fatigue Damage Prognostics Of CFRP Composites
Composites offer unique advantages for aerospace structures and are increasingly being adopted into newer designs. However, it is also acknowledged that given current understanding of damage mechanisms in composites there is a significant risk with the extensive use of composites materials in aerospace applications. On one hand the uncertainty in damage evolution along lifetime is extremely large, and on the other hand there is a lack of knowledge about the mechanics of the onset, posterior growth, and interactions between several micro-scale damage modes. All these factors lead to the adoption of high safety margins in the design and costly inspection schedules along the service to mitigate the risks. Structural health monitoring for onboard damage diagnosis and prognosis of structural failures has the potential to reduce maintenance costs and improve the safety of the structure through a condition based maintenance scheduling. In this scheme the current damage state of a specific structural element is estimated and further used as the input for a prognostic algorithm that predicts the propagation of damage through time using updated models and based on some knowledge of the future load conditions. A novel damage prognostics framework for composites FRP under fatigue loadings is proposed in this work. The proposed methodology is grounded on physics-based models for evolution of damage at (1) micro-scale, i.e. micro-cracks and delamination, and (2) macro-scale such as stiffness reduction induced by micro-scale damage modes. Through stochastic embedding, these apriori deterministic models are converted to probabilistic models by introducing a modeling error term. This error term is controlled by a probability density function whose parameters are estimated in addition to the rest of "physical" parameters. The probabilistic damage models are then incorporated in a Bayesian filtering algorithm that sequentially updates both, a damage state variable and the set of model parameters, as fresh damage data become available along the fatigue cycling process. Next, these damage models are used to simulate fault propagation with this updated state information to generate a prognostic estimate of the remaining useful life of the structure in a probabilistic sense. The proposed methodology is demonstrated using experimental NDE damage data for micro-crack density, delamination area, and stiffness reduction from an extensive post-impact tension-tension fatigue test performed over several CFRP [0,90]4s laminates.
