neutrophil-lymphocyte ratio and platelet-lymphocyte ratio in early stage breast cancer as predictor of disease free survival
<p>The relationship between neutrophil-lymphocyte ratio (NLR) and outcome probably a complex and multifactorial issues. High neutrophil-lymphocyte ratio (NLR) is reflect the role of systemic inflammation in accelerate angiogenesis, tumor grow-up, and metastasis development.</p> <p><strong>Objective:</strong> The study aimed to estimate the relation between NLR, and PLR with disease free survival (DFS), and assess the values of clinico-pathologic factors on the prognosis.</p> <p><strong>Methods: </strong>Retrospectively we reviewed 1167 files and 102 patients with breast cancer included in Babylon Oncology Center from January 2009 to September 2014, and follow up for at least 36 months.</p> <p><strong>Results: </strong>102 patients, with mean age of 50.4 ± 11.7 years and minimum follow up 40 months, the overall median DFS was 62 months with 5 – years DFS of 52.5%, stage N0 show significant better DFS compared to stage N1 (P value 0.004), also patients with positive hormonal status show significant better DFS compared to negative hormonal status. The isolated local recurrence is 15% while combined local recurrences and distant metastasis is 37%.</p> <p><strong>Conclusion: </strong>NLR is a prognostic marker, and its establish the prediction models warrants further investigation. Increase NLR is strongly associated with poor DFS, and it can be as a predictive and prognostic factor.</p>

<p>This record contains raw data related to article "ACKR2 in hematopoietic precursors as a checkpoint of neutrophil release and anti-metastatic activity"</p> <p>Atypical chemokine receptors (ACKRs) are regulators of leukocyte traffic, inflammation, and immunity. ACKR2 is a scavenger for most inflammatory CC chemokines and is a negative regulator of inflammation. Here we report that ACKR2 is expressed in hematopoietic precursors and downregulated during myeloid differentiation. Genetic inactivation of ACKR2 results in increased levels of inflammatory chemokine receptors and release from the bone marrow of neutrophils with increased anti-metastatic activity. In a model of NeuT-driven primary mammary carcinogenesis ACKR2 deficiency is associated with increased primary tumor growth and protection against metastasis. ACKR2 deficiency results in neutrophil-mediated protection against metastasis in mice orthotopically transplanted with 4T1 mammary carcinoma and intravenously injected with B16F10 melanoma cell lines. Thus, ACKR2 is a key regulator (checkpoint) of mouse myeloid differentiation and function and its targeting unleashes the anti-metastatic activity of neutrophils in mice.</p>
Comparison of Fixed Single Cell RNA-seq Methods to Enable Transcriptome Profiling of Neutrophils in Clinical Samples - Time course data
<p>Monitoring neutrophil gene expression is a powerful tool for understanding disease mechanisms, developing new diagnostics, therapies and optimizing clinical trials. Neutrophils are sensitive to the processing, storage and transportation steps that are involved in clinical sample analysis. This study is the first to evaluate the capabilities of technologies from 10X Genomics, PARSE Biosciences, and HIVE (Honeycomb Biotechnologies) to generate high-quality RNA data from human blood-derived neutrophils. Our comparative analysis shows that all methods produced high quality data, importantly capturing the transcriptomes of neutrophils. 10X FLEX cell populations in particular showed a close concordance with the flow cytometry data. Here, we establish a reliable single-cell RNA sequencing workflow for neutrophils in clinical trials: we offer guidelines on sample collection to preserve RNA quality and demonstrate how each method performs in capturing sensitive cell populations in clinical practice.</p> <p><strong>This dataset includes only the 10X Flex time course data and analysis.</strong></p>
Data from: Legacy and emerging per- and polyfluoroalkyl substances suppress the neutrophil respiratory burst
<p>Per- and polyfluoroalkyl substances (PFASs) are used in a multitude of processes and products, including nonstick coatings, food wrappers, and fire-fighting foams. These chemicals are environmentally persistent, ubiquitous, and can be detected in the serum of 98% of Americans. Despite evidence that PFASs alter adaptive immunity, few studies have investigated their effects on innate immunity. We investigated the impact of nine environmentally relevant PFASs [perfluorooctanoic acid (PFOA), perfluorooctane sulfonic acid potassium salt (PFOS-K), perfluorononanoic acid (PFNA), perfluorohexanoic acid (PFHxA), perfluorohexane sulfonic acid (PFHxS), perfluorobutane sulfonic acid (PFBS), ammonium perfluoro(2-methyl-3-oxahexanoate) (GenX), 7H-perfluoro-4-methyl-3,6-dioxaoctane sulfonic acid (Nafion byproduct 2), and perfluoromethoxyacetic acid sodium salt (PFMOAA-Na)] on one component of the innate immune response, the neutrophil respiratory burst. The respiratory burst is a key innate immune process by which microbicidal reactive oxygen species (ROS) are rapidly induced by neutrophils in response to pathogens, and defects in the respiratory burst can increase susceptibility to infection. We utilized larval zebrafish, a human neutrophil-like cell line, and primary human neutrophils to ascertain whether PFAS exposure inhibits ROS production in the respiratory burst. We observed that exposure to PFHxA and GenX suppresses the respiratory burst in zebrafish larvae and a human neutrophil-like cell line. GenX also suppresses the respiratory burst in primary human neutrophils. This report is the first to demonstrate that these PFASs suppress neutrophil function and supports the utility of employing zebrafish larvae and a human cell line as screening tools to identify chemicals that may suppress human immune function. </p>
Development of a prognostic model for early breast cancer integrating neutrophil to lymphocyte ratio and clinical-pathological characteristics
<p><strong>Abstract</strong></p> <p>Breast cancer-related inflammation is critical in tumorigenesis, cancer progression, and patient prognosis. Several inflammatory markers derived from peripheral blood cells count, such as the neutrophil-lymphocyte ratio (NLR), derived neutrophil-lymphocyte ratio (dNLR), platelet-lymphocyte ratio (PLR), monocyte-lymphocyte ratio (MLR) and systemic immune-inflammation index (SII) are considered as prognostic markers in several types of malignancy. Here, we investigate and validate a prognostic model in early breast cancer (eBC) patients to predict disease-free survival (DFS) based on readily available baseline clinicopathological prognostic factors and preoperative peripheral blood-derived indexes. We analyzed a training cohort of 710 BC patients and two external validation cohorts of 980 and 157 eBC patients, respectively, with different demographic origins. An elevated preoperative NLR is a better DFS predictor than PLR, MLR, and SII in patients with eBC. The prognostic model generated in this study was able to classify patients into three groups with different risks of relapse based on ECOG-PS, presence of comorbidities, T and N stage, PgR status, and NLR. Prognostic models derived from the combination of clinicopathological features and peripheral blood indices, such as NLR, represent attractive markers mainly because they are easily detectable and applicable in daily clinical practice. More comprehensive prospective studies are needed to unveil their actual effectiveness.</p>
