Data from: Multi-modal defenses in aphids offer redundant protection and increased costs likely impeding a protective mutualism
1.The pea aphid, Acyrthosiphon pisum, maintains extreme variation in resistance to its most common parasitoid wasp enemy, Aphidius ervi, which is sourced from two known mechanisms: protective bacterial symbionts, most commonly Hamiltonella defensa, or endogenously encoded defenses. We have recently found that individual aphids may employ each defense individually, occasionally both defenses together, or neither. 2.In field populations, Hamiltonella-infected aphids are found at low to moderate frequencies and while less is known about the frequency of resistant genotypes, they show up less often than susceptible genotypes in field collections. To better understand these patterns, we sought to compare the strengths and costs of both types of defense, individually and together, in order to elucidate the selective pressures that maintain multi-modal defense mechanisms or that may favor one over the other. 3.We experimentally infected five aphid genotypes (two lowly and three highly resistant), each with two symbiont strains, Hamiltonella-APSE8 (moderate protection) and Hamiltonella-APSE3 (high protection). This resulted in three sublines per genotype: uninfected, +APSE8, and +APSE3. Each of the fifteen total sublines was first subjected to a parasitism assay to determine its resistance phenotype and in a second experiment a subset were chosen to compare fitness (fecundity and survivorship) in presence and absence of parasitism. 4.In susceptible aphid genotypes, parasitized sublines infected with Hamiltonella generally showed increased protection with direct fitness benefits, but clear infection costs to fitness in the absence of parasitism. In resistant genotypes, Hamiltonella infection rarely conferred additional protection, often further reduced fecundity and survivorship when enemy challenged, and resulted in constitutive fitness costs in the absence of parasitism. We also identified strong aphid-genotype X symbiont-strain interactions, such that the best defensive strategy against parasitoids varied for each aphid genotype; one performed best with no protective symbionts, the others with particular strains of Hamiltonella. 5.This surprising variability in outcomes helps explain why Hamiltonella infection frequencies are often intermediate and do not strongly track parasitism frequencies in field populations. We also find that variation in endogenous traits, such as resistance, among host genotypes may offer redundancy and generally limit the invasion potential of mutualistic microbes in insects.

Data from: Fecundity of diamondback moth females when offered honey with pyridalyl, a selective insecticide
<p class="MsoNormal"><span>We previously reported that the presence of pyridalyl, a selective insecticide, in aqueous honey (50 % v/v) negatively affects the survival of diamondback moth (<em>Plutella xylostella</em>: DBM) adults. However, it remains unclear whether toxicity of pyridalyl in aqueous honey affects the fecundity of adult female DBMs. We analyzed the survival and fecundity of adult DBM under the water, honey, or honey + pyridalyl conditions in glass tubes</span><span> containing Japanese mustard spinach (</span><em><span>Brassica rapa</span></em><span>: Komatsuna) leaves</span><span> for 10 days. The survival of adults under the honey (50 % v/v) and honey + pyridalyl (100-fold dilution) conditions was significantly higher and lower, respectively, than under the water condition during the experimental period. The number of eggs laid by DBM females each day (fecundity) under the water condition was significantly lower than that under the honey condition throughout the experimental period, except on day 1. In contrast, the numbers under the water, and honey + pyridalyl conditions were not significantly different, except on day 2 (the honey + pyridalyl condition was significantly lower). To study the effects of plants grown in pots on the survival and fecundity of DBM females, we conducted experiments using acrylic cages containing potted komatsuna plants. The survival trends under the honey, and honey + pyridalyl conditions in the cages were similar to those in the glass tubes. Fecundity was evaluated based on the total number of subsequent generations (DBM larvae and pupae) on day 10. The numbers were significantly higher in the honey condition than in the water condition. </span><span>In contrast, the number in the honey + pyridalyl condition was not significantly different from that in the water condition. Based on these findings, the possible use of pyridalyl in honey for the biological control of DBM is discussed.</span></p>

<p><span></span></p> <p>Like many fern lineages comprising reticulate species complexes, <em>Polypodium</em> s.s. (Polypodiacaeae) has a history shaped by rapid diversification, hybridization, and polyploidy that poses substantial challenges for phylogenetic inference with plastid and single-locus nuclear loci. Using target capture probes for 408 nuclear loci developed by the GOFlag project and a custom bioinformatic pipeline, SORTER, we constructed multi-locus nuclear datasets for diploid temperate and Mesoamerican species of <em>Polypodium</em> and five allotetraploid species belonging to the well-studied <em>Polypodium</em> <em>vulgare</em> complex. SORTER employs a clustering approach to separate putatively paralogous copies of targeted loci into orthologous matrices and haplotype phasing to infer allopolyploid haplotypes across loci, resulting in datasets amenable to both concatenated maximum likelihood and multi-species coalescent phylogenetic analyses. By comparing phylogenies derived from maximum likelihood and multi- species coalescent analyses of unphased and phased datasets, as well as evaluating discordance among gene trees and species trees, we recover support for incomplete lineage sorting within <em>Polypodium</em> s.s., novel relationships among diploid taxa of the <em>Polypodium</em> <em>vulgare</em> complex and its Mesoamerican sister clade, and the placement of several <em>Polypodium</em> species within other genera. Additionally, we were able to infer well-supported phylogenies that identified the hypothesized progenitors of the allotetraploid species, indicating that SORTER is an effective and accurate tool for reconstructing homeolog haplotypes of allopolyploids in fern taxa and other non-model organisms from target capture data.</p>

The specific location is at a latitude of 34°59'10", longitude of 100°46'42", and an elevation of 3,766 meters above sea level.</span></p> <h3><span>OVERVIEW</span></h3> <p><span>On the morning of August 3, 2024, I drove with my nephew Bsod nams bkra shis (b. 2014), niece A+yon chos sgron (b. 2018), and sister-in-law Nyi ma lha mo (b. 1996) to visit the family of my maternal uncle, Ngag dbang smin grol (b. 1965). We traveled to our family pasture, Yen dar, in Rka rgan Valley, which my maternal uncle's family was using for summer grazing.</span></p> <p><span>The journey from the county town to the pasture was approximately forty kilometers and took about half an hour due to the rough road through our community pasture.</span></p> <p><span>Recently, my family relocated to </span><span>'Ba' rdzong </span><span>County Town, and my maternal uncle’s family now uses our land, paying us 40,000 RMB annually. My visit was to assist with a water offering and document it on video.</span></p> <h3><span>WATER OFFERING</span></h3> <p><span>Water offerings are an important and growing social trend in our community. Traditionally, families have offered water daily to Buddhist images and statues in their shrine rooms. With increased access to information, many now emulate this practice, motivated by a Buddhist belief that offering water increases virtue. The eleventh-century Buddhist master, Atisha (982-1054), advised Tibetans that offering clear water helped accumulate merit. This practice is now widespread in Tibetan monasteries and by individual families.</span></p> <p><span>I interviewed my aunt, Skal bzang sgrol ma (b. 1970), who explained that the ritual began locally in about 1994 with offerings for the healthy birth of her family’s eldest son, Yum skyabs rgya (b. 2016), and to increase family merit. Initially, we aimed to offer one million water offerings, and for that aim we <a>offer around 100,000 each summer</a></span><span>. We have since made o­fferings for the second son, Tshe dpal mkhar (b. 2018), the second daughter-in-law Choe bzang sgrol ma (b. 1996), and their sons, Sras mchog rgya (b. 2022) and Phur ba skyabs (b. 2024). </span><span>Unfortunately, we couldn’t complete the 100,000 water offerings in 2022 because my third nephew was born that year and was ill for several years. Only recently have we been able to relax and see him in better health. </span><span>We need to complete more than 100,000 offerings to reach our goal by the summer of 2025. Although we hoped to finish in the summer of 2024, the task is not easy. so we expect to accomplish it next summer.</span></p> <h3><span>Ritual Process</span></h3> <p><span>The offering involves scattering a <em>bzang drug</em> (six precious substances: amomum tsao-ko, bamboo manna, clove, saffron, nutmeg, and <em>Elettaria cardamom</em>) powder in water in a large plastic container. On the first day, this water is offered in front of an image of Buddha Amitabha near the water offering location. The following day, the local incarnation of Buddha’s image replaces Buddha’s picture</span></p> <p><span>During the ritual, a statue of Dzam b+ha la ‘God of Wealth’ is placed in a bowl on a metal folding bed at the end of a line of seven bowls. T</span><span>he </span><span>God of Wealth and the line of seven bowls remain stationary until time to take a break. Extra bowls are used repeatedly during the offering. The single string of beads with 108 beads is also placed on the bed. After completing one round, a bead is moved to the side to keep count of the numbers.</span></p> <p><span>Four people are involved in the process. Two pour water into the offering bowls, one transfers water from the bowls into another container, and a fourth person adds a small amount of water to ensure the bowls are thoroughly rinsed out.</span></p> <p><span>On August 3, 2024, 5,000 bowls of water were offered in the morning, and 2,000 more after lunch. On August 4, 2024, 3,000 offerings were made, totaling 10,000 over these two days. My aunt’s younger sister’s two daughters joined us until noon because that family is my uncle’s neighbor. Data from: Reduced fire severity offers near-term buffer to climate-driven declines in conifer resilience across the western United States
<p>The combination of increasing fire-caused tree mortality and warmer, drier post-fire conditions is making forests in the western United States (West) vulnerable to ecological transformation. Yet, the relative importance of and interactions between these drivers of forest change remain unresolved, particularly over upcoming decades. Here we assess how the interactive impacts of changing climate and wildfire activity influenced conifer regeneration after 334 wildfires, using a novel dataset of post-fire conifer regeneration from 10,230 field plots. Our findings highlight declining regeneration capacity across the West over the past four decades for the eight dominant conifer species studied. Post-fire regeneration is sensitive to high-severity fire, which limits seed availability, and post-fire climate, which influences seedling establishment and survival. In the near-term, projected differences in recruitment probability between low- and high-severity fire scenarios were larger than projected impacts of climate change for most species, suggesting that reductions in fire severity, and resultant impacts on seed availability, could partially offset expected climate-driven declines in post-fire regeneration. Across 40–42% of the study area, we project post-fire conifer regeneration to be likely following low-severity but not high-severity fire under future climate scenarios (2031–2050). However, increasingly warm, dry climate conditions are projected to eventually outweigh the influence of fire severity and seed availability. The percent of the study area considered unlikely to experience conifer regeneration, regardless of fire severity, increased from 5% in 1981–2000 to 26–31% by mid-century, highlighting a limited time window over which management actions that reduce fire severity may effectively support post-fire conifer regeneration.</p>
Data from: Ultraconserved elements improve resolution of marmot phylogeny and offer insights into biogeographic history
<p class="MsoNormal"><span>Marmots (<em>Marmota</em> spp.) comprise a lineage of large-bodied ground squirrels that diversified rapidly in the Pleistocene, when the planet quickly transitioned to a drier, colder, and highly seasonal climate—particularly at high latitudes. Fossil evidence indicates the genus spread from North America, across Beringia, and into the European Alps over the course of only a few million years, beginning in the late Pliocene. Marmots are highly adapted to survive long and severely cold winters, and this likely favored their expansion and diversification over this time period. Previous phylogenetic studies have identified two major subgenera of marmots, but the timing of important speciation events and some species relationships have been difficult to resolve. Here we use ultraconserved elements and mitogenomes, with samples from all 15 extant species, to more precisely retrace how and when marmots came to inhabit a vast Holarctic range. Our results indicate marmots arose in North America in the mid Miocene (~16.3 Mya) and dispersed across the Bering Land Bridge in the late Pliocene (~3-4 Mya); in addition, our fossil-calibrated timeline is suggestive of the rise and spread of open grasslands as being particularly important to marmot diversification. The woodchuck (<em>M. monax</em>) and the Alaska marmot (<em>M. broweri</em>) are found to be more closely related to the Eurasian species than to the other North American species. Paraphyly is evident in the bobak marmot (<em>M. bobak</em>) and the hoary marmot (<em>M. caligata</em>), and in the case of the latter the data are highly suggestive of a second, cryptic species in the Cascade Mountains of Washington.</span></p>

A calf's head, a leg of beef and a trussed bird (right center) are also present. Water pours from two tilted bottles into a small basin.
