OPTICS: an interactive online platform for photosensory and bio-functional proteins in optogenetic systems

The cutting-edge technology of optogenetics opens to new ideas for control of cellular bio-functional proteins (CPs) using optogenetic tools (OTs) in spatial and temporal. Over the past decade, hundreds of optogenetic systems (OSs) have been constructed for various applications from living cells to freely moving organisms. In this work, a new database named OPTICS (an interactive online platform for photosensory and bio-functional proteins in optogenetic systems) was thus introduced. Our OPTICS is unique in (i) systematically describing diverse OSs from the perspective of photoreceptor-based classification and mechanism of action; (ii) featuring the detailed biophysical properties and functional data of OSs; (iii) providing the interaction between OT and CP of each OS refers to distinct applications in research, diagnosis, and therapy; and (iv) enabling light response property-based search against all OSs in the database. Since the information of OSs is essential for design of optogenetic controls, the comprehensive data provided in OPTICS lay a solid foundation for the future development of novel OSs. The OPTICS is freely accessible at https://idrblab.org/optics/.

Photodynamic Ocular Drug Delivery System with Optical Coherence Tomography Oriented Microscale Robots

Retinal diseases are the most common reason for irreversible blindness worldwide. Common retinal diseases, such as diabetic retinopathy, macular degeneration, retinal vein occlusion, and non-infectious uveitis, mostly require intravitreal steroid injections in clinical management. Unfortunately, intravitreal steroid injections lead to other important ocular problems, such as elevation of intraocular pressure, intravitreal infections, and the progression of cataracts. Because of these reasons, targeted drug delivery systems are necessary for the medical treatment of retinal diseases. PHOTodynamic Ocular Drug delivery system with Optical Coherence Tomography Oriented microscale Robots (PHOTODOCTOR) project combines three novel technologies to solve this important clinical problem. The first technological advancement for the project is dexamethasone-saturated hyaluronic acid (HA) hydrogels with organo-ruthenium complexes, which enables controllable degradation under visible light exposure. With the help of visible light-controlled degradation, dexamethasone will be released with the light pulses. Then the hydrogels will be supplemented with superparamagnetic iron oxide nanoparticles (SPIONs), and 3D printed as helical micro-swimmers. Insertion of the SPIONs will help the magnetic actuation of the hydrogel-based microrobots in the intraocular space. Lastly, SPION-supplemented HA hydrogels will be observed with high-resolution optical coherence tomography (OCT) system in the intraocular space to guide them in diseased areas of the retina. While HA-based structure increases penetration in the vitreous and helps the attachment on the retina, SPIONs will increase the visibility of the microrobots in OCT imaging. In this way, a magnetic-driven, visible light-triggered intraocular drug release system with OCT guidance will be produced, and it could be used not only for intravitreal steroid delivery but also for the treatment of retinal tumours and other retinal problems.

GHRSST Level 2P Sea Surface Temperature version 1.0 from the Electro-Optical Infrared Weather System Geostationary (EWSG1) produced by NAVO
A Group for High Resolution Sea Surface Temperature (GHRSST) Level 2P sea surface temperature produced by The Naval Oceanographic Office (NAVO) from the GOES Imager sensor on the Electro-Optical Infrared Weather System – Geostationary satellite (EWS-G1). The EWS-G1, formerly GOES-13, is the first Department of Defense owned geostationary weather satellite, which has been repositioned over Indian Ocean (IO) region at 60.0° West longitude in January 2018 and fully operational since September 8, 2020, providing timely cloud characterization and theater weather imagery to DoD. The EWS-G1 L2P SST product is calculated based on the 4-micron (band 2) and 11-micron (band 4) channels, providing nighttime and daytime SST. However, daytime SSTs are not produced in areas where the 4-micron channel is strongly affected by Solar radiation, which is defined by solar reflection angle > 50 degree. The L2P data are packaged according to the GHRSST Data Specification version 2 (GDS2) in netCDF4 format at 0.04-degree spatial resolution and stored in 48 half-hour granules per day. The data will be continually updated with 24 hours latency.

Data from: Intra- and intersession repeatability of an optical quality and intraocular scattering measurement system in children
Purpose: To evaluate intra- and intersession repeatability of objective optical quality and intraocular scattering measurements with a double-pass system in children. Methods: Forty-two eyes of 42 children were included in the study. An optical quality analysis system (OQAS) was used to measure optical quality parameters, including modulation transfer function cutoff frequency (MTFcutoff), Strehl ratio (SR), OQAS values (OV) at 3 different contrasts and objective scatter index (OSI). Three measurement sessions with 10-min intervals were operated by the same technician, and in each session four consecutive measurements were obtained. Results: Mean values for MTFcutoff, SR and OSI were 46.85 ± 7.45cpd, 0.27 ± 0.06 and 0.34 ± 0.22 respectively. 1) The intraclass correlation coefficients were ranged from 0.89 to 0.97 and coefficients of variation from 0.06 to 0.16 for all the parameters in the first session; the relative repeatability were 11.1% (MTFcutoff), 22.5% (SR), 10.9% (OV100%), 16.6% (OV2%), 22.4% (OV9%) and 56.3% (OSI). Similar results were found in the second and third sessions. 2) Bland-Altman analysis showed that narrow 95% confidence intervals (compared between the first and second sessions) ranged from -5.42 to 5.28 (MTFcutoff), -0.05 to 0.07 (SR), -0.18 to 0.18 (OV100%), -0.26 to 0.29 (OV20%), -0.33 to 0.39 (OV9%) and -0.11 to 0.09 (OSI); the comparison between any two of the three sessions showed similar results. Conclusion: Measurements of optical quality and intraocular scattering in children by the double-pass system showed good intra- and intersession repeatability. Retinal image quality is high and intraocular scattering is low in children.
