N2O and CO2 fluxes and related environmental conditions in response to manure and synthetic fertilization treatments, with and without a urease inhibitor

<p>Agricultural best management practices (BMPs) intended to solve one environmental challenge may have unintended climate impacts. For example, manure injection is often promoted for its potential to reduce runoff and N loss as NH<sub>3</sub>, but the practice has been shown to increase N<sub>2</sub>O, a powerful GHG, compared to surface application. Urease inhibitor application with N fertilizer is another BMP that can enhance N retention by reducing NH<sub>3</sub> emissions, but its impact on N<sub>2</sub>O emissions is mixed. Thus, we measured N<sub>2</sub>O, CO<sub>2</sub>, soil mineral N availability, soil moisture, soil temperature, and yield in a two-year perennial hayfield trial with four fertilization treatments (manure injection, manure broadcast, synthetic urea, and control) applied with or without a urease inhibitor in Alburgh, VT (n=4 treatment replicates; 32 total subplots). This dataset includes the daily N<sub>2</sub>O and CO<sub>2</sub> fluxes in every subplot on each sampling day in our trial, along with cumulative emissions for 2020 and 2021. We also include the other variables that we measured (listed above) and relevant weather data (precipitation and air temperature). Note that during the second treatment application in 2021, the incorrect treatment was applied to one synthetic urea subplot. We therefore excluded this subplot from our analysis of daily fluxes after 30 July 2021 (n=3 synthetic urea replicates). tRIBS Model Scenarios: Forest Treatment Effects on Watershed Responses under Warming in the Beaver Creek (2003-2018)

<p>This dataset contains the model simulation setup and scenario results of the individual and combined effects of forest thinning and warming on the forest hydrologic response in the Beaver Creek watershed of central Arizona. The simulations were conducted with the Triangulated Irregular Network (TIN)-based Real-time Integrated Basin Simulator (tRIBS) model at a variable resolution of about 120 m, hourly resolution from 2003-2018 and aggregated daily values in this dataset. &nbsp;Raw model outputs at hourly resolution are available upon request from the authors, but not included here due to their excessive size.&nbsp;</p> <p>The model setup files are organized into the tar gzipped file: <strong>BCmodel.tar.gz</strong>. This contains the following files: (1) a series of input files (*.in) used for the model simulations, and (2) the ancillary data sets required for model execution (terrain model, soil map, land cover map, initialization file, data descriptor tables).&nbsp;</p> <p>The model rainfall forcing files are organized into the tar gzipped file:&nbsp;<strong>BCrain.tar.gz</strong>. This contains the following directories: (1) rainfall data from the bias-corrected NEXRAD product, and (2) rainfall data from the NLDAS-2 product.</p> <p>The model meteorological forcing files are organized into the tar gzipped file:&nbsp;<strong>BCweather.tar.gz</strong>. This contains the following data from bias-corrected, adjusted NLDAS-2: (1) atmospheric pressure, (2) wind speed, (3) air temperature, (4) incoming solar radiation, and (5) relative humidity.&nbsp;</p> <p>The daily values of the model outputs are stored in the file <strong>tRIBSModelScenarios_DailyOutputs.xlsx</strong>. This includes the scenarios (BC0, BC1, BC2, BC4, BC6, PT0, PT1, PT2, PT4, PT6) and the variables streamflow (Q), total evapotranspiration (ET), snowmelt (M), ground sublimation (Subg), and canopy sublimation (Subc). All values in mm/day over the period 10/01/2002 to 09/30/2018 (16 water years). Non-target woody plant responses to broadcast herbicide treatment for mesquite and pricklypear control

<p>Aerial spraying of herbicides is an option for treating undesirable woody species on grasslands and rangelands, but few studies have determined the effects of these products on non-target woody plants important to wildlife. A recently introduced herbicide containing a mixture of clopyralid and aminopyralid ("CA") is thought to be specific to honey mesquite (<em>Prosopis glandulosa</em>) control. Our objective was to document the effects of CA alone and mixed with other brush herbicides, including picloram and triclopyr, on two target species, honey mesquite and pricklypear (<em>Opuntia</em> spp.), and two non-target woody plants, lotebush (<em>Zizyphus obtusifolia</em>) and hackberry (<em>Celtis laevigata </em>var <em>reticulata</em>). Treatments were 1) CA, 2) CA + triclopyr (CA+Tr), 3) CA + picloram (CA+Pc), and 4) clopyralid + triclopyr (Cp+Tr). We applied aerial spray treatments on four, 4-ha replicated plots of mature mesquite thickets that also contained pricklypear in each of 3 consecutive years in north-central Texas and evaluated plots at 1 year and 2 years post-treatment (YPT). We developed a tolerance-rating model with 5 levels (highly tolerant, tolerant, moderately tolerant, moderately susceptible, and susceptible) that integrated stand-level percent whole plant mortality (root-kill) and percent canopy reduction of surviving plants. Mesquite was susceptible to all treatments in all spray years. Pricklypear was susceptible to CA+Pc [root-kill more than doubled (33 to 84%) from 1 to 2 YPT], but highly tolerant of the other treatments. Lotebush was highly tolerant or tolerant of all treatments. Hackberry was tolerant of CA and Cp+Tr but susceptible to CA+Pc. The negative effect of CA+Pc on hackberry was greater when hackberry was drought-stressed. We recommend inspection of drought status, foliage condition, and abundance of non-target woody species prior to broadcast spraying for control of targeted woody species or cacti.</p>

Osmia lignaria larval development response to heatwave treatments

<p>Heatwaves are expected to increase in frequency, intensity, and duration due to climate change. For organisms like insects with discrete development, sensitivity may differ among life stages. Thermal sensitivity is of particular concern for species like bees that provide critical ecosystem services. Although social bees moderate nest temperatures through worker behavior, solitary bees do not thermoregulate their nests, making immobile developing offspring especially vulnerable to such extreme events.</p> <p>We studied the effects of heatwaves on larval development in the solitary bee, <em>Osmia lignaria</em>, an important orchard pollinator and model species for solitary bee biology. We used a factorial design to assess the impacts of heatwave temperature and duration on larval mortality and development rate. Larvae were exposed to heatwaves under realistic diel temperature regimes, with daytime maxima of 31˚C or 37˚C for four or seven days at the beginning of development.</p> <p>Heatwave temperature strongly affected larval mortality. Exposure to 37˚C heatwaves increased larval mortality by 130%, but the cooler 31˚C heatwaves did not significantly impact mortality. Heatwave duration did not impact larval mortality.</p> <p>Larval development time also was affected by heatwave exposure. Compared to the no-heatwave-control, bees in the 31˚C heatwave developed faster, and bees in the 37˚C heatwave developed slower.</p> <p>Our study reveals the importance of stage-specific effects of extreme events and suggests that the timing and maximum temperature of projected heatwaves may be more detrimental to populations than heatwave duration.</p>

Do tradeoffs govern plant species responses to different global change treatments?

<p>Plants are subject to tradeoffs among growth strategies such that adaptations for optimal growth in one condition can preclude optimal growth in another. Thus, we hypothesized that the response of plant species abundance to one global change treatment would relate inversely to the response to a second treatment, particularly for treatment combinations that accentuate distinct traits. To address this hypothesis, we examined plant species abundances in 39 global change experiments manipulating CO2, nitrogen, phosphorus, water, temperature, or disturbance. Overall, the directional response of a species to one treatment was 13% more likely than expected to oppose its response to a second. This tendency was detectable across the global dataset but held little predictive power for individual treatment combinations or within individual experiments. While tradeoffs in the ability to respond to different global change drivers exert detectable effects globally, other forces may obscure their influence in local communities.</p>

Ultra-short response-guided Hepatitis C treatment with sofosbuvir and daclatasvir: the SEARCH study HCV sequence data

<p><strong>Background</strong></p> <p>WHO has called for research into predictive factors for selecting persons who could be successfully treated with shorter durations of antiviral therapy for Hepatitis C. We evaluated early virological response as a means of shortening treatment and explored host, viral and pharmacokinetic contributors to treatment outcome.</p> <p class="MsoNormal"><strong><span>Methods</span></strong></p> <p class="MsoNormal"><span>Duration of sofosbuvir and daclatasvir (SOF/DCV)</span> was determined according to <span>day 2 (D2) virologic response</span> for <span>HCV genotype (gt) 1- or 6-infected adults in Vietnam with mild liver disease. Participants received 4 or 8 weeks of treatment according to whether D2 HCV RNA was above or below 500 IU/ml (standard duration is 12 weeks). Primary endpoint was sustained virological response (SVR12). Those failing therapy were retreated with 12 weeks SOF/DCV. Host IFNL4 genotype and viral sequencing was performed at baseline, with repeat viral sequencing if virological rebound was observed. Levels of SOF, its inactive metabolite</span> <span>GS-331007 and DCV were measured on day 0 and 28. </span></p> <p class="MsoNormal"><strong>Findings</strong></p> <p class="MsoNormal"><span>Of 52 adults enrolled, 34 received 4 weeks SOF/DCV, 17 got 8 weeks and one withdrew. SVR12 was achieved in 21/34 (62%) treated for 4 weeks, and 17/17 (100%) treated for 8 weeks. Overall, 38/51 (75%) were cured with first-line treatment </span><span>(mean duration of 37 days). Despite a high prevalence of putative </span>NS5A-inhibitor <span>resistance-associated substitutions (RAS), all first-line treatment failures were cured after retreatment (13/13). We found no evidence treatment failure was associated with host </span><span>IFNL4 genotype, viral</span><span> subtype, baseline RAS or DCV levels. SOF metabolite levels were higher in those failing 4-week therapy.</span></p> <p class="MsoNormal"><strong>Interpretation</strong></p> <p class="MsoNormal">Shortened SOF/DCV therapy with retreatment if needed, reduces DAA use while maintaining high cure rates. D2 virologic response alone does not adequately predict SVR12 with 4 weeks of treatment.</p>

Data from: Serum metabolomics predicts treatment response in myasthenia gravis

<p>High-dose prednisone is the primary initial therapy for myasthenia gravis (MG) but upwards of a third of patients do not respond to treatment. No biomarkers can predict clinical responsiveness to corticosteroid treatment. We conducted a discovery-based study to identify treatment responsive biomarkers in MG using sera obtained at study entry to the thymectomy clinical trial (MGTX), an NIH-sponsored randomized, controlled study of thymectomy plus prednisone versus prednisone alone. We applied ultra-performance liquid chromatography coupled with electro-spray quadrupole time of flight mass spectrometry to obtain comparative serum metabolomic and lipidomic profiles at study entry to correlate with treatment response at 6 months. Treatment response was assessed using validated outcome measures of minimal manifestation status (MMS), MG-Activities of Daily Living (MG-ADL), Quantitative MG (QMG) score, or a strictly defined composite measure of response. Increased levels of phospholipids were associated with treatment response as assessed by QMG, MMS, and the Responders classification, but all measures showed limited overlap in metabolomic profiles, in particular the MG-ADL. A panel including histidine, free fatty acid (13:0), <span>γ-</span>cholestenol and guanosine was highly predictive of the strictly defined treatment response measure. The AUC in Responders' prediction for these markers was 0.90. Pathway analysis suggests that xenobiotic metabolism could play a major role in treatment resistance. We have defined a metabolomic and lipidomic profile that can now undergo validation as treatment predictive markers for MG patients undergoing corticosteroid therapy. Distinct metabolomic profiles were appreciated for each outcome measure.</p>
