Long-term nitrogen addition alters the community and energy channel but not diversity of soil nematodes in a subtropical forest

Summary <ol> <li>Research has indicated that increases in nitrogen (N) deposition can greatly affect ecosystem processes and functions. There is limited information about the effects of long-term N addition on soil nematodes and their functional composition, although nematodes are the most abundant multicellular animals on Earth.</li> <li>We conducted a field experiment in 2004 with four levels of N addition (0, 60, 120, and 240 kg N ha<sup>-1 </sup>yr<sup>-1</sup>) in a subtropical <i>Cunninghamia lanceolata</i> forest. Soil samples with three depths (0-20, 20-40 and 40-60 cm) were collected and the community structure, diversity and trophic groups of soil nematodes were determined in 2014.</li> <li>N addition significantly increased the abundance of bacterial- and fungal-feeding nematodes, but decreased the abundance of plant-feeding nematodes at the 0-20 cm soil layer. Accordingly, the plant parasite index and enrichment index decreased but the basal index and channel index increased, which weaken the importance of the plant-based energy channel, but enhance the importance of the fungal-based energy channel. N addition had no effects on the diversity of soil nematodes in three soil depths. Structural equation modeling analysis indicated that N loading directly changed plant-feeding (total <i>r<sup>2</sup></i>=0.42) nematodes, or indirectly affected bacterial- (<i>r<sup>2</sup></i>=0.43), fungal- (<i>r<sup>2</sup></i>=0.31) and plant-feeding nematodes via change soil nutrients, soil water content and pH.</li> <li>These findings suggest that N addition can change the community structure and energy channels soil nematodes, which would affect soil processes and food web functions in forest soils under future environmental change scenarios.</li> </ol>

Data from: Nonlinearities between inhibition and T-type calcium channel activity bidirectionally regulate thalamic oscillations

<p>Absence seizures result from 3-5 Hz generalized thalamocortical oscillations that depend on highly regulated inhibitory neurotransmission in the thalamus. Efficient reuptake of the inhibitory neurotransmitter GABA is essential, and reuptake failure worsens seizures. Here, we show that blocking GABA transporters (GATs) in acute brain slices containing key parts of the thalamocortical seizure network modulates epileptiform activity. As expected, we found that blocking either GAT1 or GAT3 prolonged oscillations. However, blocking both GATs unexpectedly suppressed oscillations. Integrating experimental observations into single-neuron and network-level computational models shows how a non-linear dependence of T-type calcium channel opening on GABA<sub>B</sub> receptor activity regulates network oscillations. Receptor activity that is either too brief or too protracted fails to sufficiently open T-type channels necessary for sustaining oscillations. Only within a narrow range does prolonging GABA<sub>B</sub> receptor activity promote channel opening and intensify oscillations. These results have implications for therapeutics that modulate GABA transporters.</p>

Channel cross section and floodplain water level data from the Allanmore Floodplain, River Dee, Scotland, UK (2013-2018).

Data from: Conformational dynamics in TRPV1 channels reported by an encoded coumarin amino acid

TRPV1 channels support the detection of noxious and nociceptive input. Currently available functional and structural data suggest that TRPV1 channels have two gates within their permeation pathway: one formed by a ′bundle-crossing′ at the intracellular entrance and a second constriction at the selectivity filter. To describe conformational changes associated with channel gating, the fluorescent non-canonical amino acid coumarin-tyrosine was genetically encoded at Y671, a residue proximal to the selectivity filter. Total internal reflection fluorescence microscopy was performed to image the conformational dynamics of the channels in live cells. Photon counts and optical fluctuations from coumarin encoded within TRPV1 tetramers correlates with channel activation by capsaicin, providing an optical marker of conformational dynamics at the selectivity filter. In agreement with the fluorescence data, molecular dynamics simulations display alternating solvent exposure of Y671 in the closed and open states. Overall, the data point to a dynamic selectivity filter that may serve as a gate for permeation.

An unusual amino acid substitution within hummingbird cytochrome c oxidase alters a key proton-conducting channel

<p>Hummingbirds in flight exhibit the highest metabolic rate of all vertebrates. The bioenergetic requirements associated with sustained hovering flight raise the possibility of unique amino acid substitutions that would enhance aerobic metabolism. Here, we have identified a non-conservative substitution within the mitochondria-encoded cytochrome <i>c</i> oxidase subunit I (COI) that is fixed within hummingbirds, yet exceedingly rare among other vertebrates. This unusual change is also rare among metazoans, but can be identified in several clades with diverse life histories. We performed atomistic molecular dynamics simulations using bovine and hummingbird COI models, thereby bypassing experimental limitations imposed by the inability to modify mtDNA in a site-specific manner. Intriguingly, our findings suggest that COI amino acid position 153 (bovine numbering system) provides control over the hydration and activity of a key proton channel in COX. We discuss potential phenotypic outcomes linked to this intriguing alteration encoded by the hummingbird mitochondrial genome.</p>

Adaptive and non-adaptive convergent evolution in feather reflectance of Channel Islands songbirds

<p>Convergent evolution is widely regarded as a signature of adaptation. But testing the adaptive consequences of convergent phenotypes is challenging, making it difficult to exclude non-adaptive explanations for convergence. Here, we combined feather reflectance spectra and phenotypic trajectory analyses with visual and thermoregulatory modeling to test the adaptive significance of dark plumage in songbirds of the California Channel Islands. By evolving dark dorsal plumage, island birds are generally less conspicuous to visual-hunting raptors in the island environment than mainland birds. Dark dorsal plumage also reduces the energetic demands associated with maintaining homeothermy in the cool island climate. We also found an unexpected pattern of convergence, wherein the most divergent island populations evolved greater reflectance of near-infrared radiation. However, our heat flux models indicate that elevated near-infrared reflectance is not adaptive. Analysis of feather microstructure suggests that island-mainland differences are related to coloration of feather barbs and barbules rather than their structure. Our results indicate that adaptive and non-adaptive mechanisms interact to drive plumage evolution in this system. This study sheds light on the mechanisms driving the association between dark color and wet, cold environments across the tree of life, especially in island birds.</p>

Data from: Blood pressure pulsations modulate central neuronal activity via mechanosensitive ion channels

<p><span>The transmission of heartbeat through the cerebral vascular system is known to cause intracranial pressure pulsations. Here we report that arterial pressure pulsations can directly modulate central neuronal activity. </span><span>In a semi-intact rat brain preparation, vascular pressure pulsations elicit correlated local field oscillations in the olfactory bulb (OB) mitral cell layer. These oscillations do not require synaptic transmission, but reflect baroreceptive transduction in mitral cells. This transduction is mediated by an excitatory mechanosensitive ion channel and modulates neuronal spiking activity. Indeed, in awake animals, the heartbeat entrains the activity of a subset of OB neurons within ~20 ms. Thus, we propose that this fast intrinsic interoceptive mechanism can modulate perception, e.g. during arousal, within the OB and also possibly across various brain areas.</span></p>
