Age- and sex-dependent variation in relatedness corresponds to reproductive skew, territory inheritance and workload in cooperatively breeding cichlids
<p>Kin selection plays a major role in the evolution of cooperative systems. However, many social species exhibit complex within-group relatedness structures, where kin selection alone cannot explain the occurrence of cooperative behaviour. Understanding such social structures is crucial to elucidate the evolution and maintenance of multi-layered cooperative societies. In lamprologine cichlids, intragroup relatedness seems to correlate positively with reproductive skew, suggesting that in this clade dominants tend to provide reproductive concessions to unrelated subordinates to secure their participation in brood care. We investigate how patterns of within-group relatedness covary with direct and indirect fitness benefits of cooperation in a highly social vertebrate, the cooperatively breeding, polygynous lamprologine cichlid Neolamprologus savoryi. Behavioural and genetic data from 43 groups containing 578 individuals show that groups are socially and genetically structured into subgroups. About 17% of group members were unrelated immigrants, and average relatedness between breeders and brood care helpers declined with helper age due to group membership dynamics. Hence the relative importance of direct and indirect fitness benefits of cooperation depends on helper age. Our findings highlight how both direct and indirect fitness benefits of cooperation and group membership can select for cooperative behaviour in societies comprising complex social and relatedness structures.</p>

Altruistic bet-hedging and the evolution of cooperation in a Kalahari bird
<p>Analyses of the global biogeography of altruism suggest that unpredictable environments have favoured the evolution of altruistic helping behaviour (helping to rear the offspring of others). It has therefore been hypothesised that selection for altruism may frequently arise because helping reduces variance in the reproductive success of relatives in unpredictable environments (a scenario termed 'altruistic bet-hedging'). Here we show that helping behaviour does reduce environmentally-induced variance in the reproductive success of relatives in a wild cooperative bird, the white-browed sparrow-weaver (<i>Plocepasser mahali</i>). Our decade-long study in the Kalahari desert reveals that non-breeding helpers have no overall effect on the mean reproductive success of related breeders, but instead reduce <i>variance </i>in the reproductive success of related breeders. Moreover, this variance reduction arises in part because helpers specifically reduce unpredictable rainfall-induced variance in reproductive success, just as hypothesised by global comparative analyses. Our novel analytical approach implicates effects of helping <i>per se </i>rather than correlated effects of group size and isolates within-mother effects of helping from potentially confounding among-mother variation in performance. Our findings lend new strength to the leading explanation for the global biogeography of altruism and highlight the wider importance of considering the impacts of altruism on both the mean and variance in performance of recipients.</p>

Extra-pair paternity explains cooperation among neighbours in a bird
<p>In many social animals, females mate with multiple males, but the adaptive role of female extra-pair mating is not fully understood. This data set tests whether male pied flycatchers (<em>Ficedula hypoleuca</em>) engaging in extra-pair copulations with neighbouring females were more likely to assist their neighbours in predator mobbing. It shows that extra-pair sires joined mobs more often, approached predators more closely, and attacked predators more aggressively than males without extra-pair offspring in the neighbouring nest. Our results support theoretical predictions suggesting that extra-pair mating incentivizes males to assist in nest defences. </p>

Predictive modeling reveals that higher-order cooperativity drives transcriptional repression in a synthetic developmental enhancer
<p>A challenge in quantitative biology is to predict output patterns of gene expression from knowledge of input transcription factor patterns and from the arrangement of binding sites for these transcription factors on regulatory DNA. We tested whether widespread thermodynamic models could be used to infer parameters describing simple regulatory architectures that inform parameter-free predictions of more complex enhancers in the context of transcriptional repression by Runt in the early fruit fly embryo. By modulating the number and placement of Runt binding sites within an enhancer, and quantifying the resulting transcriptional activity using live imaging, we discovered that thermodynamic models call for higher-order cooperativity between multiple molecular players. This higher-order cooperativity capture the combinatorial complexity underlying eukaryotic transcriptional regulation and cannot be determined from simpler regulatory architectures, highlighting the challenges in reaching a predictive understanding of transcriptional regulation in eukaryotes and calling for approaches that quantitatively dissect their molecular nature.</p>
