CNO-hM4D mediated perturbation of noradrenergic DBH-Cre neurons whole animal plethysmography recordings
<p>Plethysmography on conscious, unrestrained mice was carried out as described on 6-12 week old adult animals with at least 12 animals in both the experimental and control group for a given experiment. Mice were subjected to a five-day habituation protocol with each day consisting of several minutes of handling, temperature taken by rectal probe, intraperitoneal saline injection, and 30 minutes in the plethysmography chamber. Mice were then tested within one week of the last day of conditioning.</p> <p>On the day of testing, mice were taken from their home cage, weighed, and rectal temperature was taken. Animals were then placed into an airtight, temperature controlled (~32<strong>°</strong>C) plethysmography chamber and allowed to acclimate for at least 20 minutes in room air (21% O2/79% N2) conditions. After acclimation and measurement under room air, the chamber gas was switched to a hypercapnic mixture of 5% CO2/21% O2/74% N2 for 20 minutes. Chamber gas was then switched back to room air for 20 minutes. The mice were briefly removed for rectal temperature measurement and intra-peritoneal injection of clozapine-N-oxide (CNO, National Institute of Mental Health Chemical Synthesis and Drug Supply Program) dissolved in saline (0.1 mg/mL) for an effective concentration 1 mg/kg. The animal was returned to the chamber for another 20 minutes of room air, 20 minutes of hypercapnia, and 20 minutes of room air. The animal was then removed from the chamber and rectal temperature was taken immediately afterwards and again 30 minutes after the termination of the experiment. The animal was placed in its own cage during these 30 minutes at the ambient room temperature (~23<strong>°</strong>C).</p> <p>Individual recordings for all animals expressing hM4D receptors in NA neurons are in this repository. </p>

Neuronal activity in sensory cortex predicts the specificity of learning
<p>Learning to avoid dangerous signals while preserving normal responses to safe stimuli is essential for everyday behavior and survival. Following identical experiences, subjects exhibit fear specificity ranging from high (specializing fear to only the dangerous stimulus) to low (generalizing fear to safe stimuli), yet the neuronal basis of fear specificity remains unknown. Here, we identified the neuronal code that underlies inter-subject variability in fear specificity using longitudinal imaging of neuronal activity before and after differential fear conditioning in the auditory cortex of mice. Neuronal activity prior to, but not after learning predicted the level of specificity following fear conditioning across subjects. Stimulus representation in auditory cortex was reorganized following conditioning. However, the reorganized neuronal activity did not relate to the specificity of learning. These results present a novel neuronal code that determines individual patterns in learning. PINNup: Robust neural network wavefield solutions using frequency upscaling and neuron splitting
<p>Solving for the frequency-domain scattered wavefield via physics-informed neural network (PINN) has great potential in increasing the flexibility and reducing the computational cost of seismic modeling and inversion. We propose a novel implementation of PINN using frequency upscaling and neuron splitting, which allows the neural network model to grow in size as we increase the frequency while leveraging the information from the pre-trained model for lower-frequency wavefields, resulting in fast convergence to high-accuracy wavefield solutions. In this letter, we present the relevant dataset to the paper. </p>
Neuronal data sample from PMd of non-human primates (Macaca Mulatta) performing a countermanding reaching task
<p>Brief presentation of the subjects and task performed</p> <p>Two adult male rhesus macaque monkeys (Macaca mulatta; P, C) performed a countermanding reaching task. All experimental procedures, animal care, housing, and surgical procedures conformed with European (Directive 86/609/ECC and 2010/63/UE) and Italian (D.L. 116/92 and D.L. 26/2014) laws on the use of nonhuman primates in scientific research and were approved by the Italian Ministry of Health. In both monkeys a 96 channels Utah arrays (BlackRock Microsystem, USA) was employed to record neuronal activity from the dorsal premotor cortex (PMd) contralateral to the arm employed during the task.</p> <p>Behavioral task</p> <p>Monkeys performed a countermanding reaching task interacting with visual stimuli presented on a touch-screen. Each trial started with the presentation of a central stimulus (red dot) that they had touch with their finger and hold (400-900ms). Thereafter the central stimulus disappeared and, simultaneously, a target appeared (go signal) randomly at one of two opposite positions (left or right). In no-stop trials to perform a correct trial, monkeys had to move their hand form the central target, touch peripheral target and to maintain their fingers on it for a random time (400-800 ms). In stop trials at a variable delay (Stop signal delay, SSD) after the go signal was presented, the central stimulus reappeared (Stop signal) instructing the monkey to keep the hand on the starting position (additional holding time; 400-1000 ms) to perform a correct-stop trial. If the monkey moved the hand during stop trials, the trial was considered a wrong-stop trial. No-stop and stop trials were randomly intermingled in such a way that the no-stop trials were more frequent.</p> <p>Recordings</p> <p>The neuronal activity from each electrode was recorded by employing a Tucker Davies Technologies RZ2 system (sampling rate at 24.4 Khz). Single unit activity was then isolated by using Kilosort (Pachitariu, et al. in Advances in Neural information processing systems (2016) 4448-4456).</p> <p>Data and code reproduce and are linked the main figures and results of the paper "Neuronal population dynamics during motor plan cancellation in non-human primates" , <em>Proceedings of the National Academy of Sciences</em> (<em>PNAS</em>)</p> <p>For further inquiry please contact: stefano.ferraina@uniroma1.it or pierpaolo.pani@uniroma1.it</p> <p> </p>
