Gene conversion facilitates the adaptive evolution of self-resistance in highly toxic newts
<p>Reconstructing the histories of complex adaptations and identifying the evolutionary mechanisms underlying their origins are two of the primary goals of evolutionary biology. <i>Taricha </i>newts, which contain high concentrations of the deadly toxin tetrodotoxin (TTX) as an antipredator defense, have evolved resistance to self-intoxication, which is a complex adaptation requiring changes in six paralogs of the voltage-gated sodium channel (Na<sub>v</sub>) gene family, the physiological target of TTX. Here, we reconstruct the origins of TTX self-resistance by sequencing the entire Na<sub>v</sub> gene family in newts and related salamanders. We show that moderate TTX resistance evolved early in the salamander lineage in three of the six Na<sub>v</sub><i> </i>paralogs, preceding the proposed appearance of tetrodotoxic newts by ~100 million years. TTX-bearing newts possess additional unique substitutions across the entire Na<sub>v</sub> gene family that provide physiological TTX resistance. These substitutions coincide with signatures of positive selection and relaxed purifying selection, as well as gene conversion events, that together likely facilitated their evolution. We also identify a novel exon duplication within Na<sub>v</sub>1.4<i> </i>encoding an expressed TTX-binding site. Two resistance-conferring changes within newts appear to have spread via nonallelic gene conversion: in one case, one codon was copied between paralogs, and in the second, multiple substitutions were homogenized between the duplicate exons of Na<sub>v</sub>1.4. Our results demonstrate that gene conversion can accelerate the coordinated evolution of gene families in response to a common selection pressure.</p>

Effect of six- versus eight-month regimen on treatment success and smear conversion among retreatment patients
<p class="MsoNormal"><strong>Background</strong>: In 2017, the regimen for patients with retreatment tuberculosis (TB) changed from an eight-month treatment regimen that consisted of Isoniazid, Rifampicin, Pyrazinamide, Ethambutol, and Streptomycin (2RHZES/1RHZE/5RHE) to a six-month treatment regimen (2RHZE/4RH). Currently, there is uncertainty regarding the effect of the six-month treatment regimen on treatment outcomes across TB Control Programs. We evaluated the effect of the six-month treatment regimen on treatment success rate (TSR) and sputum smear conversion (SSC) among patients with retreatment pulmonary bacteriologically confirmed TB (PBC-TB) in Kampala, Uganda.</p> <p class="MsoNormal"><strong>Methods:</strong> Between November 2021 and February 2022, we abstracted data for patients with retreatment PBC-TB who had received treatment between January 2012 and July 2021 across six large TB clinics in Kampala, Uganda. The primary outcome was TSR defined as cure or treatment completed. The secondary outcome was SSC defined as a change in sputum smear test result from positive to negative. We applied doubly-robust regression estimation for causal effect analysis, with TSR as the primary outcome and SSC at 2/3, 5, and 6/8 months as secondary outcomes. We reported average causal effects using marginal odds ratios (MOR) and 95% confidence intervals (CI).</p> <p class="MsoNormal"><strong>Results</strong>: We analyzed data for 427 participants with an overall mean age of 34.6±10.8 years. Over half (53.9%) of the participants were treated with a six-month treatment regimen. TSR was 49.6% overall, and higher in the six-month regimen group (52.2%) than in the eight-month regimen group (46.7%). The six-month treatment regimen significantly improved the TSR compared to the eight-month treatment regimen (MOR, 1.27; 95% CI, 1.06-1.52). However, the effect of the six-month treatment regimen on SSC at (MOR, 1.01; 95% CI, 0.91-1.12), 5 months (MOR, 0.96; 95% CI, 0.88-1.04), and 6/8 months (MOR, 0.83; 95% CI, 0.19-3.51) is similar to the eight-month treatment regimen.</p> <p class="MsoNormal"><strong>Conclusion:</strong> The six-month treatment regimen results in a better TSR among patients with retreatment PBC-TB compared to the eight-month treatment regimen. Therefore, patients with retreatment PBC- TB who do not have rifampicin resistance should continue to receive the six-month treatment regimen.</p>

A Computer Graphics Approach to Creating New Method for Generating 3D Gesture Animations in Bangla Sign Language via HamNoSys to SiGML Conversion
<p>To prepare the system, we employed 94 classes of data. In this dataset, there are 13 Bangla numerical data classes, 36 Bangla alphabet data classes, and 41 Bangla word data classes. Every class of data contains different data types like Hand Shape, Hand Orientation, Hand Movement, Notations, etc. These data were created in SiGML tags. Every class of data is unique and different from others. The system was prepared using BdSL. And BdSL is an uncommon and unique sign language, among others. That's why every class of data is unique and created by us. We search HamNoSys notations for Bangla alphabets, words, and numbers in English HamNoSys datasets (almost 6,000 data). However, we find only 20% of the data, which is quite similar to BdSL. We create 80% HamNoSys notation for BdSL and we modify the matching 20% notations. Then, we converted them into SiGML and made the data classes. This was a big challenge in our research. </p>

Dietary availability determines metabolic conversion of long-chain polyunsaturated fatty acids in spiders: a dual compound-specific stable isotope approach
<p>The following dataset contains fatty acid contents (in mg.g<sup>-1</sup> and in %), stable isotopes (‰), and compound specific isotopic analyses of carbon and hydrogen (‰) data for insects (terrestrial and semi-aquatic) and spiders collected around Lake Lunz (47°85'N, 15°05'E), Austria from June to September 2020. Measurements were done after storage at -80°C for 2 hours, and freeze drying (24h), followed by taxonomic identification. Room-Temperature Anode-Less All-Solid-State Batteries via the Conversion Reaction of Metal Fluorides
<p>All-solid-state batteries (ASSBs) that employ anode-less electrodes have drawn attention from across the battery community because they offer competitive energy densities and a markedly improved cycle life. Nevertheless, the composite matrices of anode-less electrodes impose a substantial barrier for lithium-ion diffusion and inhibit operation at room temperature. To overcome this drawback, here, the conversion reaction of metal fluorides is exploited because metallic nanodomains formed during this reaction induce an alloying reaction with lithium ions for uniform and sustainable lithium (de)plating. Lithium fluoride (LiF), another product of the conversion reaction, prevents the agglomeration of the metallic nanodomains and also protects the electrode from fatal lithium dendrite growth. A systematic analysis identifies silver (I) fluoride (AgF) as the most suitable metal fluoride because the silver nanodomains can accommodate the solid-solution mechanism with a low nucleation overpotential. AgF-based full cells attain reliable cycling at 25 °C even with an exceptionally high areal capacity of 9.7 mAh cm<sup>−2</sup> (areal loading of LiNi<sub>0.8</sub>Co<sub>0.1</sub>Mn<sub>0.1</sub>O<sub>2</sub> = 50 mg cm<sup>–2</sup>). These results offer useful insights into designing materials for anode-less electrodes for sulfide-based ASSBs.</p>
