Flight training and dietary antioxidants have mixed effects on the oxidative status of multiple tissues in a female migratory songbird

<p>Birds, like other vertebrates, rely on a robust antioxidant system to protect themselves against oxidative imbalance caused by energy-intensive activities such as flying. Such oxidative challenges may be especially acute for females during spring migration, since they must pay the oxidative costs of flight while preparing for reproduction; however, little previous work has examined how the antioxidant system of female spring migrants responds to dietary antioxidants and the oxidative challenges of regular flying. We fed two diets to female European starlings, one supplemented with a dietary antioxidant and one without, and then flew them daily in a windtunnel for two weeks during the fall and spring migration periods. We measured the activity of enzymatic antioxidants (GPx, SOD, CAT), non-enzymatic antioxidant capacity (ORAC), and markers of oxidative damage (protein carbonyls and lipid hydroperoxides) in four tissues: pectoralis, leg, liver, and heart. Dietary antioxidants affected enzymatic antioxidant activity and lipid damage in the heart, and non-enzymatic antioxidant capacity in the pectoralis and protein damage in leg muscle. In general, birds fed less antioxidants appear to incur increased oxidative damage while upregulating non-enzymatic and enzymatic antioxidant activity, though these effects were strongly tissue-specific. We also found a significant diet x training interaction for enzymatic antioxidant activity in the leg, and strong trends for diet x training interactions for enzymatic antioxidant activity in the heart and liver. Flight-training may condition the antioxidant system of females to dynamically respond to oxidative challenges, and females during spring migration may shift antioxidant allocation to reduce oxidative damage.</p>

Season, anthocyanin supplementation, and flight training have mixed effects on the antioxidant system of migratory European starlings (Sturnus vulgaris)

<p>Migratory birds engage in two periods of endurance flight annually as they travel between summer breeding and overwintering grounds, and such endurance flights likely incur oxidative costs. These costs may differ between fall and spring migration, especially for females who must prepare for breeding and egg laying in spring. The objective of this study of a migratory bird was to test proposed hypotheses about how key components of the female's antioxidant system differ in response to flight training in the fall and spring and to dietary antioxidant supplementation. We hand-raised female European starlings (<i>Sturnus vulgaris</i>) and then fed them either a diet supplemented with dietary anthocyanins or a diet without added anthocyanins. <a name="_Hlk53656115">We then flew females in a windtunnel for 15 days during fall and spring migration seasons and measured over time oxidative damage (d-ROMs) and three components of the antioxidant system</a>—non-enzymatic antioxidant capacity (OXY), uric acid, and glutathione peroxidase (GPx) activity. Prior to flight training, non-enzymatic antioxidant capacity and oxidative damage were lower in females during spring compared to fall, and females fed a low-antioxidant diet had consistently higher circulating uric acid. GPx activity decreased more in spring immediately after a long-duration flight. Females fed a high-antioxidant diet had a greater decrease in non-enzymatic antioxidant capacity after the 15-day flight training. <a name="_Hlk53656268">Flight-trained females had higher circulating uric acid than untrained females immediately after the longest-duration flight</a>, and decreased GPx activity after the 15-day flight training. In sum, females upregulated enzymatic and non-enzymatic endogenous antioxidants in spring, and females fed a diet with less antioxidants appear to compensate by increasing circulating uric acid. Our findings emphasize the important role of dietary antioxidants for birds during migration, <a name="_Hlk53656372">and that similar flights in fall and spring likely represent distinct oxidative challenges in the life history of female birds.</a></p>

Can antioxidant responses be induced by habitat fragmentation process?

<p>Animal populations are increasingly forced to live in small residual natural or semi-natural areas due to habitat loss and fragmentation. Here, the viability of populations is often compromised by intrinsic threat factors typical of small and isolated populations, such as inbreeding depression, genetic drift, and environmental and demographic stochasticity. Under these circumstances, organisms may have low fitness due to inadequate physiological responses needed to face environmental challenges. However, few studies have investigated the relationship between habitat fragmentation and stress defences. In this study, we aimed to test whether an increase in the level of individual inbreeding produced an increase in the antioxidant system response. To this purpose, we genotyped 151 individuals of Fire Salamander Salamandra salamandra (Amphibia: Urodela) within 5 sampling populations, located in forest landscapes with different degree of fragmentation in northern Italy. For 113 individuals we also measured the glutathione-S-transferase (GST) and catalase (CAT) enzyme activity. Results showed a significant increase in individual GST activity for increasing levels of inbreeding, whereas no relationship was found for CAT. We also measured acetylcholinesterase to test the possible confounding effects of pesticides that might have occurred in fragmented landscapes with forests interspersed with agricultural areas. However, no difference in this enzyme activity was found among sampling populations. We argue that high levels of GST activity may be symptomatic of oxidative stress derived from inbreeding. An increased frequency of homozygous deleterious alleles due to inbreeding may cause homeostatic alterations and trigger the expression of GST for protection against hydrogen peroxide reactive oxygen species. We suggest using GST as a biomarker for environmental stressors with great caution and not to underestimate that the sources of stress deriving from habitat fragmentation could lead to an unbalance in the oxidative status, possibly increasing population susceptibility to infectious diseases and, potentially, spillover events and zoonoses.</p>

Warm acclimation alters antioxidant defences but not metabolic capacities in the Antarctic fish, Notothenia coriiceps

<p>The Southern Ocean surrounding the Western Antarctic Peninsula region is rapidly warming. Survival of members of the dominant suborder of Antarctic fishes, the Notothenioidei, will likely require thermal plasticity and adaptive capacity in key traits delimiting thermal tolerance. Herein, we have assessed the thermal plasticity of several cellular and biochemical pathways, many of which are known to be associated with thermal tolerance in notothenioids, including mitochondrial function, activities of aerobic and anaerobic enzymes, antioxidant defences,protein ubiquitination and degradation in cardiac, oxidative skeletal muscles and gill of <em>Notothenia coriiceps</em> warm acclimated to 4◦C for 22 days or 5◦C for 42 days. Levels of triacylglycerol (TAG) were measured in liver and oxidative and glycolytic skeletal muscles, and glycogen in liver and glycolytic muscle to assess changes in energy stores. Metabolic pathways displayed minimal thermal plasticity, yet antioxidant defences were lower in heart and oxidative skeletal muscles of warm-acclimated animals compared with animals held at ambient temperature. Despite higher metabolic rates at elevated temperature, energy storage depots of TAG and glycogen increase in liver and remain unchanged in muscle with warm acclimation. Overall, our studies reveal that <em>N. coriiceps</em> displays thermal plasticity in some key traits thatmay contribute to their survival as the Southern Ocean continues to warm.</p>

<p>Article, Dataset for article</p> <p>&nbsp;</p> <h1>Hop Flower Supercritical Carbon Dioxide Extracts Coupled with Carriers with Solubilizing Properties&mdash;Antioxidant Activity and Neuroprotective Potential</h1> <p>&nbsp;</p> <h2>Abstract</h2> <div><span>Lupuli flos</span>&nbsp;shows many biological activities like antioxidant potential, extended by a targeted effect on selected enzymes, the expression of which is characteristic for neurodegenerative changes within the nervous system.&nbsp;<span>Lupuli flos</span>&nbsp;extracts (LFE) were prepared by supercritical carbon dioxide (scCO<sub>2</sub>) extraction with various pressure and temperature parameters. The antioxidant, chelating activity, and inhibition of acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and tyrosinase by extracts were studied. The extracts containing ethanol were used as references. The most beneficial neuroprotective effects were shown by the extract obtained under 5000 PSI and 50 &deg;C. The neuroprotective effect of active compounds is limited by poor solubility; therefore, carriers with solubilizing properties were used for scCO<sub>2</sub>&nbsp;extracts, combined with post-scCO<sub>2</sub>&nbsp;ethanol extract. Determining Antioxidant Activity of Cannabis Leaves Extracts from Different Varieties—Unveiling Nature's Treasure Trove

<p>Article, Dataset for article</p> <p>&nbsp;</p> <h1>Determining Antioxidant Activity of Cannabis Leaves Extracts from Different Varieties&mdash;Unveiling Nature&rsquo;s Treasure Trove</h1> <p>&nbsp;</p> <h2>Abstract</h2> <div>Cannabis leaves contain a diverse range of antioxidants, including cannabinoids, flavonoids, and phenolic compounds, which offer significant health benefits. Utilising cannabis leaves as a source of antioxidants presents a cost-effective approach because they are typically discarded during the cultivation of cannabis plants for their seeds or fibres. Therefore, this presented study aimed to assess the antioxidant activity of the leaves of selected hemp cultivars, such as Białobrzeska, Tygra, and Henola, based on the results obtained with the 2,2&prime;-Azino-bis(3-ethylbenzthiazoline-6-sulfonic acid, ferric reducing antioxidant power, cupric reducing antioxidant capacity, and 2,2-Diphenyl-1-picrylhydrazyl assays. The cannabinoid profile was analysed for the antioxidant activity to the contents of cannabidiol (CBD), cannabigerol (CBG), &Delta;<sup>9</sup>-tetrahydrocannabinol (&Delta;<sup>9</sup>-THC), and cannabichromene (CBC), determined based on chromatographic assays. The following variables were tested: the impact of various extractants (methanol, ethanol, and isopropanol), and their mixtures (50:50,&nbsp;<span>v/v</span>, as well as extraction methods (maceration and ultra-sound-assisted extraction) significant in obtaining hemp extracts characterised by different cannabinoid profiles. The results revealed that the selection of extractant and extraction conditions significantly influenced the active compounds&rsquo; extraction efficiency and antioxidant activity. The Antioxidant and Neuroprotective Potential of Leaves and Inflorescences Extracts of Selected Hemp Varieties Obtained with scCO2

<p>Article, Dataset for the article</p> <p>&nbsp;</p> <h2>Abstract</h2> <div><span>Cannabis sativa</span>, a versatile plant with numerous varieties, holds promising potential for a wide range of biological activity. As raw materials for research, we chose leaves and inflorescences of hemp varieties such as Białobrzeskie, Henola, and Tygra, which are cultivated mainly for their fibers or seeds. The choice of extraction is a key step in obtaining the selected compositions of active compounds from plant material. Bearing in mind the lipophilic nature of cannabinoids, we performed supercritical carbon dioxide (scCO<sub>2</sub>) extraction at 50 &deg;C under 2000 (a) and 6000 PSI (b). The cannabinoid contents were determined with the use of the HPLC-DAD method. The antioxidant capabilities were assessed through a series of procedures, including the DPPH, ABTS, CUPRAC, and FRAP methods. The capacity to inhibit enzymes that play a role in the progression of neurodegenerative diseases, such as acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and tyrosinase was also studied. The dominant cannabinoids in the extracts were cannabidiol (CBD) and cannabidiolic acid (CBDA). The highest concentration of eight cannabinoids was detected in the Tygra inflorescences extract (b). The most notable antioxidant properties were provided by the Tygra inflorescences extract (b). Nonetheless, it was the Henola inflorescences extract (b) that demonstrated the most efficient inhibition of AChE and BChE, and tyrosinase was inhibited the most significantly by the Białobrzeskie inflorescences extract (b).
