Digital image correlation (DIC) measurement of contact stiffness

Measurements with digital image correlation of normal and tangential contact stiffness for ground Ti-6Al-4V interfaces suggest a linear relationship between normal contact stiffness and normal load and a linear relationship between tangential contact stiffness and tangential load. The normal contact stiffness for these surfaces is observed approximately to be inversely proportional to an equivalent surface roughness parameter, defined for two surfaces in contact. The ratio of the tangential contact stiffness to the normal contact stiffness at beginning of a load step is seen to be given approximately by the Mindlin ratio. A simple empirical model is proposed to estimate both normal and tangential contact stiffness at different loads for ground Ti-6Al-4V surfaces of known surface roughness and coefficient of friction.

The grid contains a diagonal, 300 nm wide channel, which restricts free diffusion. The file was produced using the Globals for Images, SimFCS software. The array shape is (32000, 64, 64), the data type is uint16.

The Simulation_Channel.ipcf.bin file contains the expected results of an ipCF analysis with 32 points at radius 6, and 32 bins. The array shape is (52, 52, 32, 30), the data type is float32.

The nih3t3-egfp_2.zip dataset contains a time series of SPIM images of a biological cell. It consists 35,000 TIFF files of 1024x512 16-bit grayscale samples each, 34.5 GB total.

Data from: Single-molecule imaging correlates decreasing nuclear volume with increasing TF-chromatin associations during zebrafish development
Zygotic genome activation (ZGA), the onset of transcription after initial quiescence, is a major developmental step in many species, which occurs after ten cell divisions in zebrafish embryos. How transcription factor (TF)-chromatin interactions evolve during early development to support ZGA is largely unknown. We establish single molecule tracking in live developing zebrafish embryos using reflected light-sheet microscopy to visualize two fluorescently labeled TF species, mEos2-TBP and mEos2-Sox19b. We further develop a data acquisition and analysis scheme to extract quantitative information on binding kinetics and bound fractions during fast cell cycles. The chromatin-bound fraction of both TFs increases during early development, as expected from a physical model of TF-chromatin interactions including a decreasing nuclear volume and increasing DNA accessibility. For Sox19b, data suggests the increase is mainly due to the shrinking nucleus. Our single molecule approach provides quantitative insight into changes of TF-chromatin associations during the developmental period embracing ZGA.
