Assessment of tobacco and N. benthamiana as biofactories of irregular monoterpenes for sustainable crop protection

Irregular monoterpenes are important precursors of different compounds employed in pest control such as insecticides and insect sex pheromones. Metabolically engineered plants are appealing as biofactories of such compounds, but specially as potential live biodispensers of related bioactive volatiles, which could be continuously emitted to the environment from different plant tissues. Here we assess the use of cultivated tobacco and Nicotiana benthamiana as biofactories for the irregular monoterpenes chrysanthemol and lavandulol. We evaluate the impact of high levels of constitutive metabolite production on the plant physiology and biomass, and their biosynthetic dynamics for different plant tissues and developmental stages. As an example of an active pheromone compound, we super-transformed the best lavandulol-producing tobacco line with an acetyl transferase gene to obtain a tobacco lavandulyl acetate biodispenser producing up to 0.63 mg of lavandulyl acetate per plant every day. We estimate that with these volatile emission levels, between 200 and 500 plants per hectare would be sufficient to ensure a daily emission of pheromones comparable to commercial lures. This is an important step towards plant-based sustainable solutions for pest control, and it lays the ground for further developing biofactories for other irregular monoterpenoid pheromones, whose biosynthetic genes are yet unknown.

The dataset includes GC-MS data (peak areas quantified for each compound with its Qi, normalized) for all the figures and supplementary figures of the associated manuscript. Peak areas are normalized with an admixture, analyzed on the same day as the samples.

It also includes biomass data and measurements for the analyzed plants (plant height, leaf biomass, days to flowering time).

File S3 shows data for quantitative PCR to determine transgene copy number, and for qRT-PCR to measure transgene expression levels.

For all datasets, statistical analysis is also included to support the conclusions showed in the graphs.

Data from: The effects of ground-irregularity-cancelling prosthesis control on balance over uneven surfaces

Over half of individuals with a lower-limb amputation are unable to walk on uneven terrain. Using a prosthesis emulator system, we developed an irregularity-cancelling controller intended to reduce the effect of disturbances resulting from uneven surfaces. This controller functions by changing the neutral angles of two forefoot digits in response to local terrain heights. To isolate the effects of the controller, we also programmed a spring-like controller that maintained fixed neutral angles. Five participants with transtibial amputation walked on a treadmill with an uneven walking surface. Compared to the spring-like controller, the irregularity-cancelling controller reduced ankle torque variability by 41% in the sagittal plane and 64% in the frontal plane. However, user outcomes associated with balance were mostly unaffected; only trunk movement variability was reduced, whereas metabolic rate, mediolateral center of mass motion, and variabilities in step width, step length, and step time were unchanged. We conclude that reducing ankle torque variability of the affected limb is not sufficient for reducing the overall effect of disturbances due to uneven terrain. It is possible that other factors, such as changes in step height or disturbances to the intact limb, play a larger role in difficulty balancing while walking over uneven surfaces.

Data from: Irregular forest structures originating after fire: an opportunity to promote alternatives to even-aged management in boreal forests

1. Even-aged silviculture based on short-rotation clearcuts had severely altered boreal forests. Silvicultural alternatives (e.g., continuous cover or retention forestry) has the potential to restore and protect the habitats and functions of boreal forests. These alternatives are however often restricted to structurally complex old-growth forest, which are particularly threatened by anthropogenic disturbances. Increasing the use of alternatives to even-aged silviculture in early-successional stands could help recruit more structurally complex forests, with characteristics closer to the old-growth. In this article, we therefore evaluate the potential for silvicultural alternatives to even-aged management in boreal forests that burned less than a century ago.

2. We analyzed 1085 field plots in a 243000 km2 area situated in the boreal forest of eastern Canada. These plots burned 30 to 100 years before the survey and had not been subjected to previous or subsequent anthropogenic disturbance; they hence represent young primary forests. The main patterns of tree diameter distribution variation within the plots were identified using k-means clustering. Stand structure, tree species composition, and environmental variables that most explained the differences among the clusters were identified with a random forest model, and then compared using Kruskal-Wallis and Fisher's exact tests.

3. The majority (>75 %) of the plots presented an irregular structure of stem diameters (i.e., non-normally distributed, with many small diameter trees). The understorey was generally dominated by black spruce (Picea mariana [Mill.] BSP), a shade-tolerant species. Irregular structures were observed in both forests of high and low productivity, implying that different processes (e.g., early regeneration, variable tree growth) can lead to observed early irregular structure. Regular structures were generally characterized by a higher productivity and abundance in hardwood species compared to the irregular structures.

4. Synthesis and application: Many boreal forests of eastern Canada progress towards an irregular structure in the decades following the last stand-replacing fire. A substantial part of these early-successional forests may be suitable for alternatives to even-aged silviculture that better maintains habitats and functions of preindustrial boreal forests.
