Cellulose Synthase Tethering Attenuates Mechano-induced Microtubule Organization in pavement cells

<p>Mechanical forces control development in plants and animals, acting as cues in pattern formation and as the driving force of morphogenesis. In mammalian cells, molecular assemblies residing at the interface of the cell membrane and the extracellular matrix play an important role in perceiving and transmitting external mechanical signals to trigger physiological responses. Similar processes occur in plants, but there is little understanding of the molecular mechanisms and their genetic basis. Here, we show that number and movements directions of cellulose synthase complexes (CSCs) at the plasma membrane vary during initial stages of development in the cotyledon epidermis of Arabidopsis, closely mirroring the microtubule organization. Uncoupling microtubules and CSCs resulted in enhanced microtubule co-alignment as caused by mechanical stimuli driven either by cell shape or by tissue-scale physical perturbations. Furthermore, micromechanical perturbation resulted in depletion of CSCs from the plasma membrane suggesting a possible link between cellulose synthase removal from the plasma membrane and microtubule response to mechanical stimuli. Taken together, our results suggest that the interaction of cellulose synthase and cortical microtubules forms a physical continuum between the cell wall, plasma membrane, and the cytoskeleton that modulates the mechano-response of the cytoskeleton.</p>

<p>Training data for training models for doing segmentation of edges of the Kymographs obtained by using the Multi Kymograph tool of Fiji. The images were acquired at the Kyushu university by Professor Daisuke INOUE and annotation of the data and training of the segmentation model was done by Varun Kapoor at Kapoorlabs in Paris.</p>

The movie sequence shows the progression through the tomogram in z with annotations on individual virtual sections. The combination of virtual sections results in the 3D model of the site. After an initial rotation of the model, the nuclear envelope is faded out to provide better visibility of the filaments. Mass spectrometry of natively decorated doublet microtubule from Tetrahymena thermophila WT and mutants

<p>Cilia are thin microtubule-based protrusions of eukaryotic cells. The swimming of ciliated protists and sperm cells is propelled by the beating of cilia. Cilia propagate the flow of mucus in the trachea and protect the human body from viral infections. The main force generators of ciliary beating are the outer dynein arms (ODAs) which attach to the doublet microtubules. The bending of cilia is driven by the ODAs' conformational changes caused by ATP hydrolysis. Here, we report the native ODA complex structure attaching to the doublet microtubule by cryo-electron microscopy and mass spectrometry. The structure reveals how the ODA complex is attached to the doublet microtubule via the docking complex in its native state. Combined with coarse-grained molecular dynamics simulations, we present a model of how the attachment of the ODA to the doublet microtubule induces remodeling and activation of the ODA complex.</p>
