Continuous Digital Monitoring of Walking Speed in Frail Elderly Patients: Noninterventional Validation Study and Longitudinal Clinical Trial (Data for independent validation study)
<p>Digital technologies and advanced analytics have drastically improved our ability to capture and interpret health relevant data from patients. However, to date, limited data and results have been published detailing real-world patient compliance, demonstrating accuracy in target indications or examining what novel insights and clinical value can be derived. Here we present novel, digital mobility data from two studies: an independent, non-interventional validation study with elderly, naturally slow walking subjects, and a global, multi-site phase IIb clinical trial involving patients with age-related muscle loss and slow walking speed (sarcopenia). Based on these data, we validate the accuracy of a novel algorithm for capturing in-clinic and real-world gait speed in frail, slow-walking adults. We demonstrate the feasibility of continuous monitoring with a wearable inertial sensor in elderly adults in real-world settings, and propose minimum thresholds for compliance required for robust capture of gait behaviors in this population. We also show how simple, inferred contextual information, describing the length of a given walking bout, can explain some of the variation in real-world gait speed, and use this information to demonstrate for the first time a relationship between in-clinic performance and real-world gait speed behavior. This work lays a foundation for exploration of the clinical relevance and value of such measures and is a first step in building a more complete chain of evidence between standardized physical performance assessment, real-world behavior, and subjective perceptions of mobility, independence and health.</p> <p>This dataset contains data collected during the independent validation study: derived data from raw accelerometry data, and summary performance data.</p> <p>The full dataset, including raw accelerometry data, is available here: <a href="https://mueller-et-al-2019.s3.amazonaws.com/index.html">https://mueller-et-al-2019.s3.amazonaws.com/index.html</a></p>

Weight-loss & metabolite data of cachetic cancer patients over multiple time points
<p>This dataset contains weight-loss data and metabolite measurements from cachetic cancer patients. This data set is part of the publication "Biomarkers related to fatty acid oxidative capacity are predictive for continued weight loss in cachectic cancer patients" by Catanese et al., 2021 (<a href="https://doi.org/10.1002/jcsm.12817">LINK</a>). Data were collected over a extended time period starting from the day of hospital admission. Up to seven time points are available for individual patients. A metabolite profile consisting of amino acids and acylcarnitines measured via tandem MS/MS from both blood plasma and dried blood spots is available for each patient at each time point. Surface electromyography data set of knee osteoarthritis patients undergoing hydrotherapy and physiotherapy rehabilitation routines
<p>Osteoarthritis is one of the most prevalent degenerative diseases in the elderly that affects the structural and functional integrity of the musculoskeletal system. Currently, hydrotherapy is taking relevance for osteoarthritis treatment due to its possible efficacy on pain relief and functionality. This therapy can be evaluated by means of an electromyographic analysis. The objective is to show a database of lower limbs muscles electrical activity in charge of postural stability, comparing the difference between hydrotherapy and conventional therapy through surface electromyography. A signal acquisition methodology was developed measuring electromyographic activity of the Tibialis Anterior, Soleus, Vastus Medialis, Biceps Femoris, Medial Gastrocnemius and Lateral Gastrocnemius muscles, before and after applying a specific exercise routine for knee osteoarthritis. This routine consisted of 12 therapy sessions spread over 4 weeks and could be done in water and on land. </p> <p>The data is organized according to the attendance of 28 patients to the therapy sessions. There are two records per person, before and after each hydrotherapy or physiotherapy session, obtaining a total of 1344 records at the end of the 12 sessions, and 56 data for each session. However, a total of 864 complete records were obtained for both therapies, withdrawing those who missed any of the sessions. There is a personal information database in .CSV format and electromyographic records with a .MAT extension.</p>
Patient Representatives' Perspective on Informed Consent: Nominal Group Data
<p>For Task 1.6 of the i-CONSENT project, a one day workshop was organised with nine attendees from eight European patient groups. During the workshop, Nominal Group Technique (NGT) was used to explore issues that were considered pertinent within each of the four themes (comprehension, patient expectations, gender and assent), and to reach a consensus in terms of their significance. During this exercise, the workshop facilitator presented a short introduction to the theme, before each participant was given sticky notes and instructed to write down as many individual issues as possible using a separate sticky note for each idea. There was no limit to the number of ideas the participants could generate. This stage was conducted in silence and repeated for each of the four themes. The attached documents include the list of all the ideas collected on sticky notes notes during this stage and the ranking of the items done by the participants.</p>

Diagnostic of <em>Plasmodium</em> species is important for administering the appropriate treatment. The gold-standard diagnosis from accurate species identification remains the thin blood smear. Nevertheless, this method is time-consuming and requires highly skilled and trained microscopists. To overcome these issues, new diagnostic tools based on deep learning are emerging. This study aimed to evaluate the performances of a RT-DETR (Real-Time Detection Transformer)object detection algorithm to discriminate <em>Plasmodium</em> species on thin blood smears images. The algorithm was trained and validated on a dataset consisting in 24,720 images from 475 thin blood smears corresponding to 2,002,597 labels. Performances were calculated with a test dataset of 4,508 images from 170 smears corresponding to 358,825labels coming from six French university hospital. At the patient level, the RT-DETR algorithm exhibited an overall accuracy of 79.4% (135/170) with a recall of 74% (40/54) and 81.9% (95/116) for negative and positive smears, respectively. Among <em>Plasmodium </em>positive smears, the global sensitivity was 82.7% (91/110) with a sensitivity of 90% (38/42), 81.8% (18/22) and 76.1% (35/46) for <em>P. falciparum</em>, <em>P. malariae </em>and <em>P. ovale/vivax,</em> respectively. The YOLOv5 model achieved a World Health Organization (WHO) competence level 2 for species identification. Besides, the RT-DETR algorithm may be run in real-time on low-cost devices such as a smartphone and could be suitable for deployment in low-resource setting areas where microscopy experts are lacking.</p> <p><strong>Data collection:</strong></p> <p>The training and validation dataset included 24,720 pictures taken from 475 manually May Grunwald-Giemsa (MGG)-stained thin blood smears from the Montpellier University Hospital collection and for a smaller part from the Toulouse University Hospital collection. In Montpellier, the pictures were taken with a Flexcam C1 microscope camera (Leica) attached to a Leica DM 2000 microscope and Leica DF450C microscope camera adapted with a Leica DM2500 microscope at X1000 magnification. Labelling of pictures was performed manually, and then automatically with manual correction with a Computer Visual Annotation Tools (CVAT) free software. Nine categories of labels were used: white blood cells (n=3,338), red blood cells (n=1,887,781), platelets (n=48,520), <em>Trypanosoma brucei </em>(n=2,773), and red blood cells infected by <em>P. falciparum </em>(n=43,545), <em>P. ovale </em>(n=4,651), <em>P. vivax </em>(n=4,115), <em>P. malariae </em>(n=2,849) and <em>Babesia divergens</em> (n=5,142).</p> <p>The test dataset included 4,508 pictures taken from 170 thin blood smears from the same number of patients from the Parasitology laboratories of University Hospitals of Montpellier, Toulouse, Rouen, Lille, Nantes and Saint-Louis in Paris (Table 1). Among these 170 patients, 54 were not infected, including two patients with Howell-Jolly bodies, and 116 were infected with hematozoa. For each patient, between 20 and 30 photos were taken from one thin blood smear with at least one hematozoan parasite per picture for infected patients.</p> <p>Accurate species diagnostic was made by a senior parasitologist, and for recent smears, it was confirmed by specific PCR, either performed locally (Toulouse) or at the Malaria French National Reference Center (Montpellier, Saint Louis, Rouen, Lille, Nantes).</p>

Raw data to "Specialized pro-resolving lipid mediators are differentially altered in peripheral blood of patients with multiple sclerosis and attenuate monocyte and blood-brain barrier dysfunction"
<p>Background: Lack of resolution of inflammation may be considered a critical player for the onset and progression of multiple sclerosis. To demonstrate this we extracted lipids from plasma samples of healthy donors and MS patients and we quantified over 65 lipid mediators (LMs) through LC-MS-MS using signature diagnostic ions via multiple reaction monitoring.</p> <p>Results: Out of the 65 lipid mediators analyzed, only 42 were detected and out of those only 27 were finally revealed to show differences between healthy subjects and MS patients. These 27 LMs belonged to the arachidonic (AA), docosahexaenoic (DHA) or eicosapentaenoic (EPA) acid metabolomes and we could clusterize each form of MS into a specific profile by means of principal component analysis. Altogether, compared to healthy subjects, MS patients showed a strong production of several AA-derived eicosanoids (i.e. PGE2, PGD2 and PGF2a) (Fig.1D) and a little production of two DHA-derived pro-resolving mediators (SPMs), i.e. Protectin D1 (D1) and protectin DX (PDX) (Fig.1A). However, no production of DHA-derived resolvins and maresins (Fig. 1B) as well as EPA-derived resolvins (Fig.1 C) was observed.</p> <p>When stratifying MS patients according to disease form, both relapsing MS patients showed production of only two pro-resolving mediators (SPMs), i.e. Resolvin D1 (RvD1) and Protectin D1 (D1) compared to healthy subjects, whereas remitting MS patients showed a production of only few AA- and DHA-derived metabolic pathway markers and progressive MS patients a strong production of several eicosanoids as well as other metabolic pathway markers.</p> <p>Conclusions: These data suggest that along disease progression, there is a lack of production of anti-inflammatory and pro-resolving lipid mediators associated to a higher production of pro-inflammatory ones.</p>
