The Efficacy of Daily versus Weekly Ferrous Sulfate in Maintaining Normal Serum Ferritin Levels During Pregnancy: A Randomized Controlled Trial
<p>Serum ferritin is the most reliable indication of stored iron in pregnancy, offering a<br>noninvasive way to detect iron deficiency anemia before it occurs. Therefore, this study aimed to<br>determine serum ferritin levels among women receiving daily versus weekly iron supplementation,<br>with a secondary focus on comparing the proportion developing iron deficiency anemia and compli-<br>ance rates between the two groups.<br>Methods: This non-blinded randomized control trial involved non-anaemic pregnant women attend-<br>ing antenatal clinics at two Teaching Hospitals in Osun State. One hundred twenty-five subjects were<br>recruited to receive 65mg in the control group, while another 125 subjects in the active group re-<br>ceived three tablets (195mg) of ferrous sulfate (Fesulf) once weekly for 17 weeks from the 20th to<br>37th weeks of gestation. The primary outcome measure was comparing mean serum ferritin levels in<br>both groups at 37 weeks.<br>Results: Among the 240 subjects analyzed, the 37-week serum ferritin level was higher in the daily<br>group (73.26±26.67µg/L) compared to the weekly group (63.04±30.71 µg/L), p value=0.006. Four<br>(3.36%) and 10 (8.26%) of our subjects had Iron deficiency anaemia. Nine subjects (3.75%) reported<br>dyspepsia as a side effect. Daily 65 mg of Felsulf proved more effective than weekly 195mg in main-<br>taining normal blood ferritin levels during pregnancy.<br>Conclusions: Daily iron supplementation with 65mg ferrous sulfate was more effective at main-<br>taining adequate maternal iron concentration in this group of non-anaemic pregnant women. This<br>dosage is recommended for routine iron supplementation in our environment.</p>

Targeting Plasmodium falciparum chondroitin sulfate a ligand: A highly conserved malaria antigen with potential for pregnancy-associated malaria vaccine development
<p>The dataset represents the details of sera samples collected from participants recruited for a longitudinal study. The sera was used for ELISA <em>in vitro</em> assays. The immunoreactivity of PfCSA-L antigen was assessed with the utilization of the gravidity status of the study participants. MacLeod, kirstyjmacleod@gmail.com.</p> <p>Abstract: Reproduction is a critical part of an animal’s life history, but one which incurs significant costs to survival and future reproductive potential. These physiological consequences are likely to be influenced by context – for example, if an individual is subject to environmental stressors, physiological and behavioral changes associated with reproduction may be altered. Glucocorticoids, hormones produced as part of the physiological response to stressors, may alter how reproduction affects female physiology and behavior, and therefore the outcomes of reproductive trade-offs. Glucocorticoids prioritize immediate survival over reproduction, for example through changes in immune function, metabolic rate, and foraging, which may reduce energy expenditure or increase energy gain. However, we previously found that female eastern fence lizards (<em>Sceloporus undulatus</em>) experiencing elevated glucocorticoid levels during gestation were nevertheless able to maintain reproductive output and body condition. Here we investigate compensatory mechanisms by which eastern fence lizard females may maintain reproduction under experimental increases in a glucocorticoid, corticosterone (CORT). We found that, although CORT-treated females had similar immune function and behavior, they had reduced metabolic rates 3-5 days post-parturition compared to control females. Given that CORT-treated females spent a similar time basking and had equal food intake compared to control females, we suggest that the reduced metabolic rate is a mechanism by which CORT-treated females maintain their energy balance and reduce the energetic costs of gestation during periods of stress. This study suggests that physiological responses to reproduction may be context-dependent and could act to minimize costs of reproduction in situations where CORT is elevated (such as during periods of environmental stress). </p>
A meta-analysis of pre-pregnancy maternal body mass index and placental DNA methylation identifies 27 CpG sites with implications for mother-child health
<p>Higher maternal pre-pregnancy body mass index (ppBMI) is associated with increased neonatal morbidity, as well as with pregnancy complications and metabolic outcomes in offspring later in life. Placenta is a key organ in fetal development and has been proposed to act as a mediator between the mother and different health outcomes in children. The overall aim of the present work is to investigate the association of ppBMI with epigenome-wide placental DNA methylation (DNAm) in 10 studies from the PACE consortium, amounting to 2,631 mother-child pairs. We identify 27 CpG sites at which we observe placental DNAm variations of up to 2.0% per 10 ppBMI-unit. The CpGs that are differentially methylated in placenta do not overlap with CpGs identified in previous studies in cord blood DNAm related to ppBMI. Many of the identified CpGs are located in open sea regions, are often close to obesity-related genes such as GPX1 and LGR4 and altogether, are enriched in cancer and oxidative stress pathways. Data from: Placental pathology findings in unexplained pregnancy losses
<p class="MsoNormal">There are approximately 5 million pregnancies per year in the United States, with 1 million ending in miscarriage (a loss occurring prior to 20 weeks of gestation) and over 20,000 ending in stillbirth at or beyond 20 weeks of gestation. As many as 50% of these losses are unexplained. Our objective was to evaluate the efficacy of expanding the placental pathology diagnostic categories to include the explicit categories of 1) dysmorphic chorionic villi and 2) small placenta to decrease the unexplained fraction. Using a clinical database of 1,256 previously unexplained losses at 6–43 weeks of gestation, the most prevalent abnormality associated with each loss was determined through examination of its placental pathology slides. Of 1,256 cases analyzed from 922 patients, there were 878 (69.9%) miscarriages and 378 (30.1%) antepartum stillbirths. We determined the pathologic diagnoses for 1,150/1,256 (91.6%) of the entire series, 777/878 (88.5%) of the miscarriages (<20 weeks' gestation), and 373/378 (98.7%) of the stillbirths (≥20 weeks' gestation). The most common pathologic feature observed in unexplained miscarriages was dysmorphic chorionic villi (757 cases; 86.2%), a marker associated with genetic abnormalities. The most common pathologic feature observed in unexplained stillbirths was a small placenta (128 cases; 33.9%).<strong> </strong>Our classification system reinforced the utility of placental examination for elucidating potential mechanisms behind pregnancy loss. The improved rate of diagnosis appeared to be the result of filling a gap in previous pregnancy loss classification systems via inclusion of the categories of dysmorphic chorionic villi and small placenta.</p>
Calcium fortification of water during pregnancy for the prevention of preeclampsia in a low-income setting: a cost-effectiveness analysis
<p>This database contains the model and the parameters of the study. It's is important to highlight that this is the first version of the model, and thus modifications are forthcoming. With the current database, the abstract is the following:</p> <p>Fortification of water with calcium during pregnancy is proposed as a suitable and promising intervention to increase calcium intake and prevent preeclampsia and eclampsia (PE/E). However, the evidence of the cost-effectiveness of this type of intervention is scarce. We conducted a model-based cost-effectiveness analysis to estimate the incremental cost-effectiveness ratio (ICER) of calcium pills and bottled water fortified with calcium, compared to the standard of care (defined as the use of MgSO4 as the standard treatment for PE/E), in the context of Nepal. Outcome measures were years of life gained (YLG) by mothers and newborns and healthcare costs. We considered a lifetime time horizon and health benefits were discounted at 3%. The cost-effectiveness threshold was set at the 2019 Nepal gross domestic product per capita (USD 1071). For calcium pills versus standard of care, ICER was USD 63 per YLG. By switching from calcium pills to bottled water fortified with calcium, the resulting ICER was USD 1702 per YLG. In conclusion, the bottle of water fortified with calcium was found not cost-effective at the defined threshold for Nepal. Complementary economic evidence adapted to other low-and middle income contexts is required to guide novel preventive interventions to improve maternal and child health based on the efficiency principle</p>
Calorie restriction and pravastatin administration during pregnancy in obese rhesus macaques modulates maternal and infant metabolism and infant brain and behavioral development
<p>Maternal obesity has been associated with a higher risk of pregnancy-related complications in mothers and offspring; however, effective interventions have not yet been developed. We tested two common interventions, calorie restriction and pravastatin administration, during pregnancy in a rhesus macaque model with the hypothesis that these interventions would normalize metabolic dysregulation in pregnant mothers leading to an improvement in infant metabolic and cognitive/social development. A total of 19 obese mothers were assigned to either one of the two intervention groups (n=5 for calorie restriction; n=7 for pravastatin) or an obese control group (n=7) with no intervention, and maternal gestational samples and postnatal infant samples were compared with lean control mothers (n=6). Gestational calorie restriction normalized one-carbon metabolism dysregulation in obese mothers but altered energy metabolism in their offspring. Although administration of pravastatin during pregnancy tended to normalize blood cholesterol in the mothers, it potentially impacted the gut microbiome and kidney function of their offspring. In the offspring, both calorie restriction and pravastatin administration during pregnancy tended to normalize the activity of AMPK in the brain at 6 months, and while results of the Visual Paired-Comparison test, which measures infant recognition memory, were not significantly impacted by either of the interventions, gestational pravastatin administration, but not calorie restriction, tended to normalize anxiety assessed by the Human Intruder test. Although the two interventions tested in a non-human primate model led to some improvements in metabolism and/or infant brain development, negative impacts were also found in both mothers and infants. Our study emphasizes the importance of assessing gestational interventions for maternal obesity on both maternal and offspring long-term outcomes.</p>
Dataset for: Estimating pregnancy rate from blubber progesterone levels of a blindly biopsied beluga population poses methodological, analytical and statistical challenges
<p class="MsoBodyText"><span>Beluga (<em>Delphinapterus leucas</em>) from the St. Lawrence Estuary, Canada, have been declining since the early 2000s, suggesting recruitment issues as a result of low fecundity, abnormal abortion rates or poor calf or juvenile survival. Pregnancy is difficult to observe in cetaceans, making the ground-truthing of pregnancy estimates in wild individuals challenging. Blubber progesterone concentrations were contrasted among 62 SLE beluga with a known reproductive state (i.e., pregnant, resting, parturient, and lactating females), that were found dead in 1997–2019. The suitability of a threshold obtained from decaying carcasses to assess reproductive state and pregnancy rate of freshly-dead or free-ranging and blindly-sampled beluga was examined using three statistical approaches and two datasets (135 freshly-harvested carcasses in Nunavik, and 65 biopsy-sampled SLE beluga). Progesterone concentrations in decaying carcasses were considerably higher in known-pregnant (mean </span><span>±</span><span> sd: </span><span>365 </span><span>±</span><span> 244 ng g<sup>-1</sup> of tissue) </span><span>than resting (</span><span>3.1 </span><span>±</span><span> 4.5 ng g<sup>-1</sup> of tissue</span><span>) or lactating (</span><span>38.4 </span><span>±</span><span> 100 ng g<sup>-1</sup> of tissue</span><span>) females. An approach based on statistical mixtures of distributions and a logistic regression was compared to the commonly-used, fixed threshold approach (here, 100 ng g<sup>-1</sup>) for discriminating pregnant from non-pregnant females. The error rate for classifying individuals of known reproductive status was the lowest for the fixed threshold and logistic regression approaches, but the mixture approach required limited <em>a priori</em> knowledge for clustering individuals of unknown pregnancy status. Mismatches in assignations occurred at lipid content <10% of sample weight. Our results emphasize the importance of reporting lipid contents and progesterone concentrations in both units (ng g<sup>-1 </sup>of tissue and ng g<sup>-1</sup> of lipid) when sample mass is low. By highlighting ways to circumvent potential biases in field sampling associated with capturability of different segments of a population, this study also enhances the usefulness of the technique for estimating pregnancy rate of free-ranging population. </span></p>
Identification of quantitative trait loci and associated candidate genes for pregnancy success in Angus – Brahman crossbred heifers
<p>Development of genomic tools to identify females with high genetic merit for reproductive function could increase the profitability and sustainability of beef production. Here, genome-wide association studies (GWAS) were performed on pregnancy outcome traits from a population of Angus – Brahman crossbred heifers. Furthermore, a validation GWAS was performed using data from another location. Heifers were genotyped with the Bovine GGP F250 array that contains ~250,000 SNPs. In the discovery population, heifers were bred in winter breeding seasons involving a single round of timed artificial insemination (AI) followed by natural mating for three months. Three phenotypes were analyzed: pregnancy outcome to first-service AI (PAI; n = 1481), pregnancy status at the end of the breeding season (PEBS; n = 1725), and pregnancy score (Pregscore where 1 = pregnant to first-service AI, 2 = pregnant to bull, 3 = not pregnant; n =1481). The heritability for PAI was estimated as 0.149. One large quantitative trait locus (QTL) that explained ~3% of the genetic variation for PAI was found on BTA7, in a region containing a cluster of γ-protocadherin genes and SLC25A2. Other QTLs explaining between 0.5-1% of the genetic variation were found on BTA12 and 25. The heritability of PEBS was estimated at 0.122. A large QTL on BTA7 was synonymous with the QTL for PAI, with minor QTL located on BTA5, 9, 10, 11, 19, and 20. Estimated heritability for Pregscore was 0.189. There was a large QTL on BTA7 synonymous with the other two traits as well as smaller QTLs on BTA1, 10, 15, 18, 19, and 20. The validation population for pregnancy status at the end of the breeding season were Angus-Brahman crossbred heifers bred by natural mating. In concordance with the discovery population, the large QTL on BTA7 and QTL on BTA10, 12 and 18 were identified. In summary, QTL and candidate SNPs associated with pregnancy outcomes in beef heifers were identified, including a large QTL associated with a group of protocadherin genes. Confirmation of these associations with larger populations could lead to the development of genomic estimates of reproductive function in beef cattle.</p>
