Data from: Diversification by host switching and dispersal shaped the diversity and distribution of avian malaria parasites in Amazonia

Understanding how pathogens and parasites diversify through time and space is fundamental to predicting emerging infectious diseases. Here, we use biogeographic, coevolutionary and phylogenetic analyses to describe the origin, diversity, and distribution of avian malaria parasites in the most diverse avifauna on Earth. We first performed phylogenetic analyses using the mitochondrial cytochrome b (cyt b) gene to determine relationships among parasite lineages. Then, we estimated divergence times and reconstructed ancestral areas to uncover how landscape evolution has shaped the diversification of Parahaemoproteus and Plasmodium in Amazonia. Finally, we assessed the coevolutionary patterns of diversification in this host–parasite system to determine how coevolution may have influenced the contemporary diversity of avian malaria parasites and their distribution among Amazonian birds. Biogeographic analysis of 324 haemosporidian parasite lineages recovered from 4178 individual birds provided strong evidence that these pathogens readily disperse across major Amazonian rivers and this has occurred with increasing frequency over the last five million years. We also recovered many duplication events within areas of endemism in Amazonia. Cophylogenetic analyses of these blood parasites and their avian hosts support a diversification history dominated by host switching. The ability of avian malaria parasites to disperse geographically and shift among avian hosts has played a major role in their radiation and has shaped the current distribution and diversity of these parasites across Amazonia.

West Nile Virus evolves at the pace of its avian hosts' life-history

<p>Disease resistant hosts are central in the spread, persistence, adaptation and diversification of infectious pathogens. In theory, resistant hosts must compromise on reproduction and invest more energy in self maintenance and immune defence. Here we investigated these hypotheses on the emergence of West Nile Virus (WNV) from 1999 to 2012 in the United States, when the virus caused mass mortality in some bird species. Our study comprehends two steps: First, we used regression tree meta-analysis on 233 reports of dead birds from 90 species, to test correlations between WNV-linked mortality and the birds' life history and ecological traits. Second, we tested links between diversification of WNV and the reproductive investment and self-maintenance of its avian hosts, by analysing the phylogeography of 520 genome sequences of WNV from 1999 to 2012. Statistical analyses revealed less WNV-linked mortality in both, species that incubate their eggs for few days (&gt;17) and large species (&gt;235g). The evolutionary trajectory of the virus suggests that there was no special viral strain infecting the most and the less vulnerable species. However, by 2012 the WNV strains found in large birds like American crows (<i>Corvus brachyrhynchos</i>) diversified and evolved separately from the rest. Together, our findings support that hosts' reproductive investment impairs disease resistance, and hosts investing in self-maintenance may lead WNV diversification.</p>

Data from: Host interaction analysis of PA-N155 and PA-N182 in chicken cells reveals an essential role of UBA52 for replication of H5N1 avian influenza virus

PA-N155 and PA-N182 proteins were translated from the 11th and 13th start codon AUG of the RNA polymerase acidic protein (PA) mRNA of H5N1 influenza A virus (IAV), which plays an important role in viral replication. Little is known about the interactions between PA-N155 and PA-N182 and the host proteins. This study investigated the interaction landscape of PA-N155 and PA-N182 of H5N1 IAV in chicken cells while their interacting complexes were captured by immunoprecipitation and analyzed by mass spectrometry. A total of 491 (PA-N155) and 302 (PA-N182) interacting proteins were identified. Gene ontology and pathway enrichment analyses showed that proteins of the two interactomes were enriched in RNA processing, viral processing and protein transport, and proteins related to signaling pathways of proteasome, ribosome, and aminoacy1-tRNA biosynthesis were significantly enriched, suggesting their potential roles in H5N1 IAV infection. Comparative analysis of the interactome of PA, PA-N155, and PA-N182 identified UBA52 as a conserved host factor that interacted with all three viral proteins. UBA52 is a fusion protein consisting of ubiquitin at the N terminus and ribosomal protein L40 at the C terminus. Knockdown of UBA52 significantly decreased the titer of H5N1 IAV in chicken cells and was accompanied with attenuated production of proinflammatory cytokines. Our analyses of the influenza–host protein interactomes identified UBA52 as a PA interaction protein for virus replication.
