Data from: Molecular field analysis of trophic relationships in soil-dwelling invertebrates to identify mercury, lead and cadmium transmission through forest ecosystems

Contamination pathways in complex food chains in soil ecosystems can be difficult to elucidate. Molecular analysis of predator gut content can, however, rapidly reveal previously unidentified trophic interactions between invertebrates and thereby uncover pathways of pollutant spread. Here we measured concentrations of the toxic metals lead, cadmium and mercury in carabid beetle predators and their prey. Invertebrates were sampled at one control and four heavy metal polluted sites in order to reveal the impact of diet composition and seasonal variation in prey availability on metal burden in carabids and metal transfer pathways through forest ecosystems. This is the first report, to our knowledge, of carabid diet composition based on PCR analysis of gut contents at the forest community level, rather than in cultivated fields. Extensive screening using group- and species-specific primers revealed that carabids ate primarily earthworms and slugs, as well as smaller numbers of woodlice and springtails. Metal concentrations in carabids correlated with seasonal changes in diet. Mercury accumulated in beetle predators more than in their slug prey. Since earthworms, slugs and carabid beetles are the major prey of many birds and mammals, prey-predator transfer and associated toxicity are major risks at mercury-contaminated sites. Carabids may be useful bioindicators for assessing the impact of pollutants on soil ecosystems, as long as species and seasonal factors are taken into account.

Preservation of the soil resources stability is of high importance for ecosystems, particularly in the current era of environmental change, which presents a severe pollution burden (e.g. by heavy metals) to soil fauna. Gut microbiomes are becoming recognized as important players in organism health, with comprehension of their perturbations in the polluted environment offering new insights into the nature and extent of heavy metal effects on the health of soil biota. Our aim was to investigate the effect of environmentally relevant heavy metal concentrations of cadmium (Cd) on the earthworm (Lumbricus terrestris) gut microbiota. Our results revealed that Cd exposure led to perturbations of earthworm gut microbiota with an increase in bacteria previously described as heavy metal resistant or able to bind heavy metals, revealing the potential of the earthworm-gut microbiota system in overcoming human-caused heavy metal pollution. Furthermore, an 'indicator species analysis' linked the bacterial genera Paenibacillus, Flavobacterium and Pseudomonas, with Cd treatment, suggesting these bacterial taxa as biomarkers of exposure in earthworms inhabiting Cd-stressed soils. The results of this study help to understand the impact of anthropogenic disturbance on soil fauna health and will have implications for environmental monitoring and protection of soil resources.

Data from: Effect of cadmium accumulation on the performance of plants and of herbivores that cope differently with organic defenses

Some plants are able to accumulate in their shoots metals at levels that are toxic to most other organisms. This ability may serve as a defence against herbivores. Therefore, both metal-based and organic defences may affect herbivores. However, how metal accumulation affects the interaction between herbivores and organic plant defences remains overlooked. To fill this gap, we studied the interactions between tomato (Solanum lycopersicum), a model plant that accumulates cadmium, and two spidermite species, Tetranychus urticae and Tetranychus evansi that, respectively, induce and suppress organic plant defences, measurable via the activity of trypsin inhibitors. We exposed plants to different concentrations of cadmium and measured its effects on mites and plants. In the plant, despite clear evidence for cadmium accumulation, we did not detect any cadmium effects on traits that reflect the general response of the plant, such as biomass, water content, and carbon/nitrogen ratio. Still, we found effects of cadmium upon the quantity of soluble sugars and on leaf reflectance, where it may indicate structural modifications in the cells. These changes in plant traits affected the performance of spider mites feeding on those plants. Indeed, the oviposition of both spider mite species was higher on plants exposed to low concentrations of cadmium than on control plants, but decreased at concentrations above 0.5 mM. Therefore, herbivores with contrasting responses to organic defences showed a similar hormetic response to metal accumulation by the plants. Additionally, we show that the induction and suppression of plant defences by these spider-mite species was not affected by the amount of cadmium supplied to the plants. Furthermore, the effect of cadmium on the performance of spider mites was not altered by infestation with T. urticae or T. evansi. Together, our results suggest no interaction between cadmium-based and organic plant defences, on our system. This character may be useful for plants living in heterogeneous environments, as they may use one or the other defence mechanism, depending on their relative performance in each environment.

Deciphering evolutionary processes occurring within contaminated populations is important for the ecological risk assessment of toxic chemicals. Whereas increased tolerance to contaminants is well documented in aquatic animal populations, whether such phenotypic changes occur through genetic adaptation is still debated. In that sense, several studies with the freshwater crustacean Gammarus concluded in a weak potential for genetic adaptation to cadmium (Cd), while others reported inheritable increased tolerance in Cd-contaminated populations. Using quantitative genetics and selection experiments, this study sought to further assess the potential of Gammarus populations to genetically adapt to Cd. By combining the control of the reproductive cycle of this species in the laboratory and protocols of individual Cd exposure, we conducted half-sib analyses to establish the genetic and environmental sources of variance in Cd sensitivity of neonates. Prior to experiments, computations allowed optimizing the experimental design in order to increase the power to detect additive genetic variance. The main findings are the existence of strong between-brood variability along with weak heritability of Cd sensitivity within Gammarus populations. This study also revealed a significant maternal effect on individual Cd sensitivity. This sheds new light on the importance of maternal influence in microevolutionary processes occurring in contaminated environments.
