Data from: Numerical simulations of targeted delivery of magnetic drug aerosols in the human upper and central respiratory system: a validation study

In the present study, we investigate the concept of the targeted delivery of pharmaceutical drug aerosols in an anatomically realistic geometry of the human upper and central respiratory system. The geometry considered extends from the mouth inlet to the 8th generation of the bronchial bifurcations and is identical to the phantom model used in the experimental studies of [Banko {em et al.} (2015), Exp. Fluids, {bf 56} (117):1-12]. In our computer simulations, we combine the transitional Reynolds-Averaged Navier-Stokes (RANS) and the wall-resolved Large Eddy Simulation (LES) methods for the air phase with the Lagrangian approach for the particulate (aerosol) phase. We validated simulations against recently obtained magnetic resonance velocimetry (MRV) measurements of [Banko {em et al.} (2015), Exp. Fluids, {bf 56} (117):1-12] that provide full a 3D mean velocity field for steady inspiratory conditions. Both approaches produced good agreement with experiments, and the transitional RANS approach is selected for the multi-phase simulations of aerosols transport, because of significantly lower computational costs. The local and total deposition efficiency are calculated for different classes of pharmaceutical particles (in the $0.1mu$m$le d_{rm p} le 10mu$m range) without and with a paramagnetic core (the shell-core particles). For the latter, an external magnetic field is imposed. The source of the imposed magnetic field was placed in the proximity of the first bronchial bifurcation. We demonstrated that both total- and local-depositions of aerosols at targeted locations can be significantly increased by an applied magnetization force. This finding confirms the possible potential for further advancement of the magnetic drug targeting (MDT) technique for more efficient treatments for respiratory diseases.

Data from: Anthracycline drugs on modified surface of quercetin-loaded polymer nanoparticles: a dual drug delivery model for cancer treatment

Polymer nanoparticles are vehicles used for delivery of hydrophobic anti-cancer drugs, like doxorubicin, paclitaxel or chemopreventors like quercetin (Q). The present study deals with the synthesis and characterisation of nano formulations (NFs) from Q loaded PLGA (poly lactic-co-glycolic acid) nano particles (NPs) by surface modification. The surface of Q-loaded (NPs) is modified by coating with biopolymers like bovine serum albumin (BSA) or histones (His). Conventional chemotherapeutic drugs adriamycin (ADR) and mitoxantrone (MTX) are bound to BSA and His respectively before being coated on Q-loaded NPs to nano formulate NF1 and NF2 respectively. The sizes of these NFs are in the range 400-500 nm as ascertained by SEM and DLS measurements. Encapsulation of Q in polymer NPs is confirmed from shifts in FT-IR, TGA and DSC traces of Q-loaded NPs compared to native PLGA and Q. Surface modification in NFs is evidenced by three distinct regions in their TEM images; the core, polymer capsule and the coated surface. Negative zeta potential of Q-loaded NPs shifted to positive potential on surface modification in NF1 and NF2. In vitro release of Q from the NFs lasted up to twenty days with an early burst release. NF2 is better formulation than NF1 as loading of MTX is 85% compared to 23% loading of ADR. Such NFs are expected to overcome multi-drug resistance (MDR) by reaching and treating the target cancerous cells by virtue of size, charge and retention.

Photodynamic Ocular Drug Delivery System with Optical Coherence Tomography Oriented Microscale Robots

<p>Retinal diseases are the most common reason for irreversible blindness worldwide. Common retinal diseases, such as diabetic retinopathy, macular degeneration, retinal vein occlusion, and non-infectious uveitis, mostly require intravitreal steroid injections in clinical management. Unfortunately, intravitreal steroid injections lead to other important ocular problems, such as elevation of intraocular pressure, intravitreal infections, and the progression of cataracts. Because of these reasons, targeted drug delivery systems are necessary for the medical treatment of retinal diseases. PHOTodynamic Ocular Drug delivery system with Optical Coherence Tomography Oriented microscale Robots (PHOTODOCTOR) project combines three novel technologies to solve this important clinical problem. The first technological advancement for the project is dexamethasone-saturated hyaluronic acid (HA) hydrogels with organo-ruthenium complexes, which enables controllable degradation under visible light exposure. With the help of visible light-controlled degradation, dexamethasone will be released with the light pulses. Then the hydrogels will be supplemented with superparamagnetic iron oxide nanoparticles (SPIONs), and 3D printed as helical micro-swimmers. Insertion of the SPIONs will help the magnetic actuation of the hydrogel-based microrobots in the intraocular space. Lastly, SPION-supplemented HA hydrogels will be observed with high-resolution optical coherence tomography (OCT) system in the intraocular space to guide them in diseased areas of the retina. While HA-based structure increases penetration in the vitreous and helps the attachment on the retina, SPIONs will increase the visibility of the microrobots in OCT imaging. In this way, a magnetic-driven, visible light-triggered intraocular drug release system with OCT guidance will be produced, and it could be used not only for intravitreal steroid delivery but also for the treatment of retinal tumours and other retinal problems.</p>
