Data from: Bacterial biophotons as non-local information carriers: species-specific spectral characteristics of a stress response

Studies by Alexander Gurwitsch in the 1920's with onion root cells revealed the phenomenon of mitogenetic radiation. Subsequent works by Popp, Van Wijk, Quickenden, Tillbury and Trushin have demonstrated a link between Gurwitsch's mitogenetic radiation and the biophoton, emissions of light correlated with biological processes. The present study seeks to expand upon these works and explore whether biophoton emissions of bacterial cultures is used as an information carrier of environmental stress. Bacterial cultures (Escherichia coli and Serratia marcescens) were incubated for 24 h in 5 mL of Nutrient Broth to stationary phase and cell densities of ~107 cells/mL. Cultures of E. coli were placed upon a photomultiplier tube housed within a dark box. A second bacterial culture, either E. coli or S. marcescens, was placed in an identical dark box at a distance of 5 m and received injections of hydrogen peroxide. Spectral analyses revealed significant differences in peak frequencies of 7.2, 10.1, and 24.9 Hz in the amplitude modulation of the emitted biophoton signal with respect to whether a peroxide injection occurred or not, and whether the species receiving the injection was E. coli or S. marcescens. These and the subsequent results of discriminant functions suggest that bacteria may release biophotons as a non-local communication system in response to stress, and that these biophotons are species specific.

Data from: Scaling up effects of measures mitigating pollinator loss from local- to landscape-level population responses

1. Declining pollinator populations have caused concern about consequences for food production, and have initiated an increasing number of initiatives that aim to mitigate pollinator loss through enhancement of floral resources. Studies evaluating effects of mitigation measures generally demonstrate positive responses of pollinators to floral resource enhancement. However, it remains unclear whether this represents landscape-level population effects or results from a spatial redistribution of individuals from otherwise unaffected populations. 2. Here we present a method for estimating landscape-level population effects using data from commonly used standardized pollinator transect surveys. The approach links local density responses of pollinators in both mitigation sites and surrounding landscape elements to the area these habitats occupy in mitigation landscapes as well as control landscapes to obtain landscape-level population estimates. 3. We demonstrate the method using data from a two-year study examining the effects of experimental wildflower enhancements on wild bumble bees and solitary bees in Dutch agricultural landscapes. The results show that conclusions based on local responses may differ significantly from those based on landscape-level responses. 4. Wildflower enhancements significantly enhanced landscape-level abundance of both bumble bees and solitary bees. Bumble bees showed a pronounced positive local density response in mitigation sites and the surrounding landscape that was in line with significant landscape-level increases in abundance. However, solitary bees showed no local response to mitigation sites, and the landscape-level increases in abundance only became apparent when the area of bee habitat was taken into account. 5. Incorporating the area of both newly created and pre-existing pollinator habitats into effect estimates accounts for density-dependent processes such as dilution, spill-over and local concentration of individuals. It therefore results in more reliable estimates of the response to mitigation measures of pollinators, as well as other mobile arthropod groups that are often being surveyed using transect surveys.

Data from: A cross-seasonal perspective on local adaptation: metabolic plasticity mediates responses to winter in a thermal-generalist moth

The physiological and ecological impact of the thermal environment across life-stages can result in trade-offs that determine fitness and population dynamics. Understanding mechanisms and consequences of local adaptation for any organism that overwinters requires taking a cross-seasonal perspective. We used a trait-based approach to distinguish variation among ecotypes in ecological and physiological responses to overwintering conditions. We used fall webworms (Hyphantria cunea; Lepidoptera: Arctiidae) from Ottawa, Ontario and Columbus Ohio, representing the centre and periphery of the native range. We hypothesised that populations would be locally adapted to their overwintering environments, with fitness maximised under natal overwintering conditions. We predicted that this local adaptation would result from modulation of rates of energy use, growth and development. Each ecotype had higher overwinter survival in their natal compared to non-natal winter environment, and this was associated with larger pupal mass, size and carbohydrate reserves at the end of winter. This suggests that the ecotypes are locally adapted to winter conditions. Larger adults laid more eggs, but there was no effect of ecotype or environment on fecundity. Pupae overwintering at warm, energetically demanding southern temperatures suppressed metabolic rates in autumn, and developed more quickly in the spring, compensating for energetic demands of warmer winters. Northern ecotypes had lower thermal sensitivity of metabolism, leading to higher metabolic rates at cool temperatures that correlated with faster post-winter development. Local adaptation to winter conditions suggests performance of peripheral populations may not be enhanced by warming winters. Decoupling of winter and growing season temperatures may negatively impact ectotherms.

Data from: Responses of aerial insectivorous bats to local and landscape-level features of coffee agroforestry systems in Western Ghats, India

Shade coffee has shown great promise in providing crucial habitats for biodiversity outside formal protected areas. Insectivorous bats have been understudied in coffee, although they may provide pest control services. We investigated the influence of local and landscape-level features of coffee farms on aerial insectivorous bats in Chikmagalur district in the Western Ghats biodiversity hotspot, India. Bats were monitored in 20 farm sites using ultrasound detectors, and the response of bat species richness and activity to changes in tree density, proportion of built-up area in the neighborhood, and distance of farm from forest areas quantified. We examined if models built to explain the species richness and activity could also predict them in nine additional sites. We detected nine phonic types/species in the study area. The quantified predictors had no effect on assemblage-level species richness and activity of bats. Responses of edge-space and cluttered-space forager guilds mirrored those of the overall assemblage, but some species vulnerable to forest conversion like *Rhinolophus beddomei* were detected rarely. Best models explained up to 20% and 15% variation in assemblage-level species richness and activity respectively, and were poor predictors of both response variables. We conclude that coffee farms in our study area offer an important commuting space for insectivorous bats across a gradient of shade management. Further research should include species-specific responses to management decisions for at-risk species and quantification of ecosystem services like natural pest control to inform biodiversity conservation initiatives in the Western Ghats coffee landscapes.

Data from: Modeling intraspecific adaptation of Abies sachalinensis to local altitude and responses to global warming, based on a 36-year reciprocal transplant experiment

Intraspecific adaptation in Abies sachalinensis was examined using models based on long-term monitoring data gathered during a reciprocal transplant experiment with eight seed source populations and six transplantation sites along an altitudinal gradient. The consequence of local adaptation was evaluated by testing the home-site advantage for upslope and downslope transplants at five ages. The populations' fitness-linked trait was set as their productivity (tree height ×survival rate) at each age. The effects of global warming were evaluated on the basis of the 36-year performance of downslope transplants. Evidence was found for adaptive genetic variation affecting both height and survival from an early age. Increasing the distance between seed source and planting site significantly reduced productivity for both upslope and downslope transplantation, demonstrating the existence of a significant home-site advantage. The decrease in productivity was most distinct for upslope transplantations, indicating strong local adaptation to high altitudes. Global warming is predicted to increase the productivity of high-altitude populations. However, due to their existing local adaptation, all tested populations exhibited lower productivity under warming than demes that were optimal for the new climate. These negative predictions should be considered when planning the management of locally-adapted plant species such as A. sachalinensis.

Data from: Growth responses to soil water potential indirectly shape local species distributions of tropical forest seedlings

1. Local tree species distributions in tropical forests correlate strongly with soil water availability. However, it is unclear how species distributions are shaped by demographic responses to soil water availability. Specifically, it remains unknown how growth affects species distributions along water availability gradients relative to mortality. 2. We quantified spatial variation in dry season soil water potential (SWP) in the moist tropical forest on Barro Colorado Island, Panama, and used a hierarchical Bayesian approach to evaluate relationships between demographic responses of naturally regenerating seedlings to SWP (relative growth rates and first-year mortality) and species distributions along the SWP gradient for 62 species. We also tested whether species that were more abundant at the wet or dry end of the gradient performed better (1) at their 'home end' of the gradient ('best at home' hypothesis) and (2) 'at home' compared to co-occurring species ('home advantage' hypothesis). 3. Four and five species responded significantly to SWP in terms of growth or mortality, respectively. Growth (but not mortality) responses were positively related to species distributions along the SWP gradient; species with a more positive (negative) growth response to SWP were more abundant at higher (lower) SWP, i.e. at wetter (drier) sites. In addition, wet distributed species grew faster on the wet end of the SWP gradient than on the dry end ('best at home') and grew faster on the wet end than dry distributed species ('home advantage'). Mortality rates declined with seedling size for all species. Thus, seedling growth responses to SWP indirectly shaped local species distributions by influencing seedling size and thereby mortality risk. 4. Synthesis. By demonstrating how growth responses to spatial variation in soil water availability affect species distributions, we identified a demographic process underlying niche differentiation on hydrological gradients in tropical forests. Recognizing the role of these growth responses in shaping species distributions should improve understanding of tropical forest composition and diversity along rainfall gradients and with climate change.

Data from: Phylogeny, traits and biodiversity of a neotropical bat assemblage: close relatives show similar responses to local deforestation

If species' evolutionary pasts predetermine their responses to evolutionarily novel stressors, then phylogeny could predict species survival in an increasingly human-dominated world. To understand the role of phylogenetic relatedness in structuring responses to rapid environmental change, we focused on assemblages of Neotropical bats, an ecologically diverse and functionally important group. We examined how taxonomic and phylogenetic diversity shift between tropical forest and farmland. We then explored the importance of evolutionary history by ascertaining whether close relatives share similar responses to environmental change and which species traits might mediate these trends. We analyzed a 5-year data set (5,011 captures) from 18 sites in a countryside landscape in southern Costa Rica using statistical models that account and correct for imperfect detection of species across sites, spatial autocorrelation, and consideration of spatial scale. Taxonomic and phylogenetic diversity decreased with deforestation, and assemblages became more phylogenetically clustered. Species' responses to deforestation were strongly phylogenetically correlated. Body mass and absolute wing loading explained a substantial portion of species variation in species' habitat preferences, likely related to these traits' influence on maneuverability in cluttered forest environments. Our findings highlight the role that evolutionary history plays in determining which species will survive human impacts and the need to consider diversity metrics, evolutionary history, and traits together when making predictions about species persistence for conservation or ecosystem functioning.

Forest spider data to study the response of assemblages to regional and local processes

<p>Understanding species richness variation among local communities is one of the central topics in ecology, but the complex interplay of regional processes, environmental filtering and local processes hampers generalization on the importance of different processes. Here, we aim to  unravel drivers of spider community assembly in temperate forests by analyzing two independent data sets covering gradients in elevation and forest succession. We test the following four hypotheses: (H1) Spider assemblages within a region are limited by dispersal; (H2) Local environment has a disproportionate influence on species composition; and (H3) resources and (H4) biotic interactions both affect species richness patterns. Spider data were collected by pitfall traps in two independent projects within a region along elevation and canopy cover gradients.</p>

Data from: Local diversity in phenological responses of migratory lake sturgeon to warm winters

<p>Rich intraspecific diversity in traits that shape responses to environmental conditions implies that effects of climate change will differ within species or even populations. Nevertheless, few studies investigate how different groups within species respond to climatic fluctuations, and most risk assessments rely upon species-wide generalizations. We studied effects of among-year variation in air temperature on the spring migratory phenology of a metapopulation of lake sturgeon (<em>Acipenser fulvescens</em>) within waters connecting Lake Huron and Lake Erie of the Laurentian Great Lakes. Sturgeon here express multiple migratory phenotypes that all spawn in either the St. Clair River or Detroit River but differ in their use after spawning of more than 86,000 km<sup>2</sup> of accessible lake and river habitat. Acoustic tracking over nine years (2012-2020) revealed mixed phenological responses to late-winter air temperatures, with three migratory groups arriving at rivers earlier during warm years and one whose arrival was consistent across years regardless of temperature. Notably, two groups that spawn in the same river but overwinter in different lakes entered the river with greater synchrony during warm years because one advanced its phenology while the other did not. The results indicated warm weather could alter the dynamics of the metapopulation and broader community, and exemplify the complexity hidden beneath broadscale generalizations of species' response to climate change.</p>

A comprehensive experimental analysis of the local passive response across the healthy porcine left ventricle

<p>This work provides a comprehensive characterization of porcine myocardial tissue, combining biaxial (Bx), simple triaxial shear (STS) and confined compression (CC) tests to analyze its elastic behavior under cyclic loads. We expanded this study to different zones of the ventricular free wall, providing insights into the local behavior along the longitudinal and radial coordinates. The aging impact was also assessed by comparing two age groups (4 and 8 months). Resulting data showed that the myocardium exhibits a highly nonlinear hyperelastic and incompressible behavior. We observed an anisotropy ratio of 2-2.4 between averaged peak stresses in Bx tests and 1-0.59-0.41 orthotropy ratios for normalised fiber-sheet-normal peak stresses in STS tests. We obtained a highly incompressible response, reaching volumetric pressures of 2-7 MPa for perfused tissue in CC tests, with notable differences when fluid drainage was allowed, suggesting a high permeability. Regional analysis showed reduced stiffness and anisotropy (20-25%) at the apical region compared to the medial, which we attributed to differences in the fiber field dispersion. Compressibility also increased towards the epicardium and apical regions. Regarding age-related variations, 8-month animals showed stiffer response (at least 30% increase), particularly in directions where the mechanical stress is absorbed by collagenous fibers (more than 90%), as supported by a histological analysis. Although compressibility of perfused tissue remained unchanged, permeability significantly reduced in 8-month-old animals. Our findings offer new insights into myocardial properties, emphasizing on local variations, which can help to get a more realistic understanding of cardiac mechanics in this common animal model.</p>

In Situ-Forming Gels Loaded with Stimuli-Responsive Gated Mesoporous Silica Nanoparticles for Local Sustained Drug Delivery

<p>A novel combination of in situ-forming hydrogels of hyaluronic acid with gated mesoporous materials was developed to design depots for local sustained release of chemotherapeutics. The depot consists of a hyaluronic-based gel loaded with redox-responsive mesoporous silica nanoparticles loaded with safranin O or doxorubicin and capped with polyethylene glycol chains containing a disulfide bond. The nanoparticles are able to deliver the payload in the presence of the reducing agent, glutathione (GSH), that promotes the cleavage of the disulfide bonds and the consequent pore opening and cargo delivery. Release studies and cellular assays demonstrated that the depot can successfully liberate the nanoparticles to the media and, subsequently, that the nanoparticles are internalized into the cells where the high concentration of GSH induces cargo delivery. When the nanoparticles were loaded with doxorubicin, a significant reduction in cell viability was observed. Our research opens the way to the development of new depots that enhance the local controlled release of chemotherapeutics by combining the tunable properties of hyaluronic gels with a wide range of gated materials.</p>
