African Continent of Flags of Nations Animus Prime Research Collective

<p>In a remarkable show of unity and diversity, the nations of Africa came together to proudly display their national flags. This captivating sight was witnessed at an event that celebrated the rich cultural heritage and shared aspirations of the continent. The vibrant display of flags served as a visual reminder of the strong bond that unites the African nations. Representatives from each country unfurled their flags, each boasting a unique design and symbolism that reflects their individual history, traditions, and values. From the bold colors of Ghana's flag representing the country's vibrant spirit, to the symbol of peace and harmony depicted on the flag of Rwanda, the diversity of African culture was proudly showcased. The event was not only a celebration of diversity but also a demonstration of solidarity. As the flags fluttered in unison, it served as a powerful reminder of the collective strength and determination of the African nations to overcome challenges and work towards a brighter future. The flags also symbolized the shared purpose of the African nations to foster peace, stability, and development on the continent.</p><p>&nbsp;They stood as a testament to the commitment of African leaders and citizens to create a better tomorrow, one that is characterized by progress, prosperity, and mutual respect. During the event, attendees marveled at the array of flags on display, recognizing the rich tapestry of cultures and identities that make up the African continent. Visitors had the opportunity to learn about the unique stories and symbolism behind each flag, gaining a deeper appreciation for the values and aspirations of the respective countries. In addition to celebrating unity and diversity, the event aimed to promote cultural exchange and strengthen diplomatic ties among the African nations. Representatives engaged in fruitful discussions, forging new partnerships, and exploring avenues for collaboration in various fields, including trade, tourism, and education. As the event drew to a close, the resonance of this display of flags lingered in the hearts and minds of all who attended. The unity and diversity showcased through this visual spectacle served as a powerful reminder of what can be achieved when nations come together to celebrate their unique identities and work towards a common goal. As the flags were gently folded and put away, the spirit of unity continued to resonate, leaving a lasting impression on all who witnessed this magnificent display. The message was clear: when the nations of Africa stand together, they stand strong, united, and poised to overcome any challenge that comes their way. In the spirit of celebrating African culture and progress, let us honor this display of unity by embracing the values of diversity, solidarity, and collaboration. Together, we can build a brighter future for Africa and the world at large.</p>

mRNA prime–boost evolves precursors toward VRC01-like broadly neutralizing antibodies in preclinical humanized mouse models

<p>Germline-targeting (GT) protein immunogens to induce VRC01-class broadly neutralizing antibodies (bnAbs) to the CD4 binding site (CD4bs) of the HIV Envelope have shown promise in clinical trials. Here, we preclinically validated the mRNA-LNP delivery of one such immunogen, eOD-GT8, as a soluble self-assembling 60mer nanoparticle in humanized mouse models. In a model with three humanized B cell lineages bearing distinct VRC01-precursor B cell receptors (BCRs) with similar affinities for eOD-GT8, all lineages could be simultaneously primed and undergo diversification and affinity maturation with no sign of exclusionary competition. Boosts drove precursor B cell participation in germinal centers, the accumulation of somatic hypermutations, including in key VRC01-class positions, and affinity maturation to boost and native-like antigens in two of the three precursor lineages. We have preclinically validated a prime-boost regimen of soluble self-assembling nanoparticles delivered by mRNA-LNP, demonstrating that multiple lineages can be primed, boosted, and diversify along the bnAb pathway.</p>

Parasite resistance and parasite tolerance: insights into transgenerational immune priming in an invertebrate host

<p>Parasites impose different selection regimes on their hosts, which respond by increasing their resistance and/or tolerance. Parental challenge with parasites can enhance the immune response of their offspring, a phenomenon documented in invertebrates and termed transgenerational immune priming. We exposed two parental generations of the model organism <em>Daphnia magna</em> to the horizontally-transmitted parasitic yeast <em>Metschnikowia bicuspidata</em>, and recorded resistance- and tolerance-related traits in the offspring generation. We hypothesized that parentally-primed offspring will increase either their resistance or their tolerance to the parasite. Our susceptibility assays revealed no impact of parental exposure on offspring resistance. Nonetheless, different fitness-related traits, which are indicative of tolerance, were altered. Specifically, maternal priming increased offspring production and decreased survival. Grandmaternal priming positively affected age at first reproduction and negatively affected brood size at first reproduction. Interestingly, both maternal and grandmaternal priming significantly reduced within-host parasite proliferation. Nevertheless, <em>Daphnia </em>primed for two consecutive generations had no competitive advantage in comparison to unprimed ones, implying additive maternal and grandmaternal effects. Our findings do not support evidence of transgenerational immune priming from bacterial infections in the same host species, thus emphasizing that transgenerational immune responses may not be consistent even within the same host species.</p>

All-atom molecular dynamics simulations of synaptic vesicle fusion I: a glimpse at the primed Synaptotagmin-SNARE-complexin complex

<p>Synaptic vesicles are primed into a state that is ready for fast neurotransmitter release upon Ca2+-binding to Syt1. This state likely includes trans-SNARE complexes between the vesicle and plasma membranes that are bound to Syt1 and complexins. However, the nature of this state and the steps leading to membrane fusion are unclear, in part because of the difficulty of studying this dynamic process experimentally. To shed light into these questions, we performed all-atom molecular dynamics simulations of systems containing trans-SNARE complexes between two flat bilayers or a vesicle and a flat bilayer with or without fragments of Syt1 and/or complexin-1. Our results need to be interpreted with caution because of the limited simulation times and the absence of key components, but suggest mechanistic features that may control release and help visualize potential states of the primed Syt1-SNARE-complexin-1 complex. In particular, the simulations suggest that SNAREs alone induce formation of extended membrane-membrane contact interfaces that may fuse slowly, and that the primed state contains macromolecular assemblies of trans-SNARE complexes bound to the Syt1 C2B domain and complexin-1 in a spring-loaded configuration that prevents premature membrane merger and formation of extended interfaces but keeps the system ready for fast fusion upon Ca2+ influx.</p>
