Radical change: Temporal patterns of oxidative stress during social ascent in a dominance hierarchy

<p>Dominant individuals have priority access to mates and resources. However, high rank can be costly too, especially when it is maintained by intense agonistic behavior. Oxidative stress has been proposed as a potential cost of social dominance. However, social dominance hierarchies can be dynamic, and few studies have examined the cost of social dominance when males are changing status. We studied temporal changes in markers of oxidative stress during social ascent in the East African cichlid fish <i>Astatotilapia burtoni</i>. After removing the dominant male, males ascended from subordinate to dominant status. On the first day of social ascent, immediately after the dominant male removal, the newly dominant male showed lower levels of plasma total antioxidant capacity (TAC). However, we found that liver TAC and liver superoxide dismutase, an enzymatic antioxidant, were significantly upregulated on day 1 and 2 of social ascent, respectively. By day 14, all markers of oxidative stress were similar to those observed in stable dominant males, which has higher levels of reactive oxygen metabolites (ROM) compared to subordinate males. We conclude that markers of oxidative stress vary dramatically during social ascent in a time- and tissue-sensitive manner. Our study provides a more nuanced look at the oxidative cost of social dominance and highlights the importance of considering temporal changes in markers of oxidative stress during important life-history events.</p>

Data from: Evolutionary conserved neural signature of early life stress affects animal social competence

In vertebrates the early social environment can persistently influence behaviour and social competence later in life. However, the molecular mechanisms underlying variation in animal social competence are largely unknown. In rats, high-quality maternal care causes an upregulation of hippocampal glucocorticoid receptors (gr) and reduces offspring stress responsiveness. This identifies gr regulation as a candidate mechanism for maintaining variation in animal social competence. We tested this hypothesis in a highly-social cichlid fish, Neolamprologus pulcher, reared with or without caring parents. We find that the molecular pathway translating early social experience into later-life alterations of the stress axis is homologous across vertebrates: fish reared with parents expressed the glucocorticoid receptor gr1 more in the telencephalon. Furthermore, expression levels of the transcription factor egr-1 (early growth response 1) were associated with gr1 expression in the telencephalon and hypothalamus. When blocking glucocorticoid receptors (GR) with an antagonist, mifepristone (RU486), parent-reared individuals showed more socially appropriate, submissive behaviour when intruding a larger conspecific's territory. Remarkably, mifepristone-treated fish were less attacked by owners and had a higher likelihood of territory takeover. Our results indicate that early social-environment effects on stress axis programming are mediated by an evolutionary conserved molecular pathway, which is causally involved in environmentally-induced variation of animal social competence.

Data from: Repetitive concussive traumatic brain injury interacts with post-injury foot shock stress to worsen social and depression-like behavior in mice

The debilitating effects of repetitive concussive traumatic brain injury (rcTBI) have been increasingly recognized in both military and civilian populations. rcTBI may result in significant neurological, cognitive, and affective sequelae, and is often followed by physical and/or psychological post-injury stressors that may exacerbate the effects of the injury and prolong the recovery period for injured patients. However, the consequences of post-injury stressors and their subsequent effects on social and emotional behavior in the context of rcTBI have been relatively little studied in animal models. Here, we use a mouse model of rcTBI with two closed-skull blunt impacts 24 hours apart and social and emotional behavior testing to examine the consequences of a stressor (foot shock fear conditioning) following brain injury (rcTBI). rcTBI alone did not affect cued or contextual fear conditioning or extinction compared to uninjured sham animals. In the sucrose preference test, rcTBI animals had decreased preference for sucrose, an anhedonia-like behavior, regardless of whether they experienced foot shock stress or were non-shocked controls. However, rcTBI and post-injury foot shock stress had synergistic effects in tests of social recognition and depression-like behavior. In the social recognition test, animals with both injury and shock were more impaired than either non-shocked injured mice or shocked but uninjured mice. In the tail suspension test, injured mice had increased depression-like behavior compared with uninjured mice, and shock stress worsened the depression-like behavior only in the injured mice with no effect in the uninjured mice. These results provide a model of subtle emotional behavioral deficits after combined concussive brain injury and stress, and may provide a platform for testing treatment and prevention strategies for social behavior deficits and mood disorders that are tailored to patients with traumatic brain injury.

Data from: Modulation of social space by dopamine in Drosophila melanogaster, but no effect on the avoidance of the Drosophila stress odorant

Appropriate response to others is necessary for social interactions. Yet little is known about how neurotransmitters regulate attractive and repulsive social cues. Using genetic and pharmacological manipulations in Drosophila melanogaster, we show that dopamine is contributing the response to others in a social group, specifically, social spacing, but not the avoidance of odours released by stressed flies (dSO). Interestingly, this dopamine-mediated behaviour is prominent only in the day-time, and its effect varies depending on tissue, sex and type of manipulation. Furthermore, alteration of dopamine levels has no effect on dSO avoidance regardless of sex, which suggests that a different neurotransmitter regulates this response.

Dominance hierarchies are a fundamental part of social systems in many species and social rank can influence access to resources and impact health and physiology. While social subordination is a profound stressor, few studies consider the social stress experienced by dominant males due to constantly needing to defend their dominance status through costly aggressive displays. Recent studies suggest that in species that use body coloration to signal status, these costs may also be color morph-specific. Our study examines the link between the social rank, intensity of territorial defense, body coloration, and oxidative stress in males of the color polymorphic cichlid fish Astatotilapia burtoni where males are either blue or yellow. We studied behavior in naturalistic communities and examined circulating reactive oxygen metabolites and antioxidant defenses. We found that dominant males experience higher concentrations of circulating reactive oxygen metabolites without notably increasing their antioxidant defenses, but this effect was not related to color morph. Aggression and social network ties predicted oxidative stress in a morph-specific manner, with yellow but not blue males showing signs of increased oxidative damage with increasing agonistic effort. In contrast to expectation, oxidative stress was not influenced by cortisol or testosterone levels. We conclude that oxidative stress is instrumental to understanding the costs and benefits of high social rank.

Data from: Social buffering of stress in a group-living fish

Living in groups affords individuals many benefits including the opportunity to reduce stress. In mammals, such 'social buffering' of stress is mediated by affiliative relationships and production of the neuropeptide oxytocin, but whether these mechanisms facilitate social buffering across vertebrates remains an open question. Therefore, we evaluated whether the social environment influenced the behavioural and physiological recovery from an acute stressor in a group-living cichlid, Neolamprologus pulcher. Individual fish that recovered with their social group displayed lower cortisol levels than individuals that recovered alone. This social buffering of the stress response was associated with a tendency towards lower transcript abundance of arginine vasotocin and isotocin in the preoptic area of the brain, suggesting reduced neural activation of the stress axis. Individuals that recovered with their social group quickly resumed normal behaviour but received fewer affiliative acts following the stressor. Further experiments revealed similar cortisol levels between individuals that recovered in visual contact with their own social group and those in visual contact with a novel but non-aggressive social group. Collectively, our results suggest that affiliation and familiarity per se do not mediate social buffering in this group-living cichlid, and the behavioural and physiological mechanisms responsible for social buffering may vary across vertebrates.

Data from: Developmental stress predicts social network position

The quantity and quality of social relationships, as captured by social network analysis, can have major fitness consequences. Various studies have shown that individual differences in social behaviour can be due to variation in exposure to developmental stress. However, whether these developmental differences translate to consistent differences in social network position is not known. We experimentally increased levels of the avian stress hormone corticosterone (CORT) in nestling zebra finches in a fully balanced design. Upon reaching nutritional independence, we released chicks and their families into two free-flying rooms, where we measured daily social networks over five weeks using passive integrated transponder tags. Developmental stress had a significant effect on social behaviour: despite having similar foraging patterns, CORT chicks had weaker associations to their parents than control chicks. Instead, CORT chicks foraged with a greater number of flock mates and were less choosy with whom they foraged, resulting in more central network positions. These findings highlight the importance of taking developmental history into account to understand the drivers of social organization in gregarious species.

Measures of glucocorticoid stress hormones (e.g. cortisol) have often been used to characterize conflict between subordinates and dominants. In cooperative breeders where subordinates seldom breed in their natal group and assist in offspring rearing, increases in subordinate glucocorticoid levels may be caused by conflict among subordinates as well as by the energetic costs of helping behavior and fluctuations in food availability may exacerbate these effects. During a 6-year study of Kalahari meerkats (Suricata suricatta), we investigated how social, environmental, and individual characteristics influenced subordinate plasma cortisol levels. Subordinate females, who are often the target of aggression from dominant females, had higher cortisol levels when the dominant female in their group was pregnant while the cortisol levels of subordinate males were unaffected by the reproductive state of dominant females. Subordinates of both sexes had higher cortisol levels if they belonged to groups 1) where neither of the dominant breeders in the group were their parents, 2) that contained a high proportion of subordinate females, or 3) that were either very large or very small, especially when the weather was cold and dry. Subordinates in groups containing young pups had higher cortisol levels. Finally, cortisol levels were higher in subordinates of both sexes if they were lighter for their age or had lost little body mass the night prior to sampling. Our results show that both social conflict and cooperative behavior can elevate glucocorticoid levels in subordinates and that both effects can be modified by variation in weather and food availability.
