Data from: Functional antibodies against Plasmodium falciparum sporozoites are associated with a longer time to qPCR-detected infection among schoolchildren in Burkina Faso
Background: Individuals living in malaria-endemic regions develop naturally acquired immunity against severe malarial disease, but it is unclear whether immunity that affects the establishment of infections develops following continuous natural exposure. Methods: We cleared schoolchildren in Burkina Faso of possible sub-patent infections and examined them weekly for incident infections by PCR. Plasma samples collected at enrolment were used to quantify antibodies to the pre-eryhrocytic-stage antigens circumsporozoite protein (CSP) and liver stage antigen. Sporozoite gliding inhibition by naturally acquired antibodies was assessed using Plasmodium falciparum NF54 sporozoites; hepatocyte invasion was assessed using the human HC-04 hepatoma cell line and NF54 sporozoites. The associations between these functional pre-erythrocytic immunity phenotypes and time to PCR-detected infection were studied. Results: A total of 51 children were monitored; the median time to first detection of infection by PCR or development of clinical symptoms was 28 days. Anti-CSP antibody titres showed a strong positive association with sporozoite gliding motility inhibition (P<0.0001, Spearman's ρ=0.76). In vitro hepatocyte invasion was inhibited by naturally acquired antibodies (median invasion inhibition, 19.4% [IQR 15.2-40.9%]), and there was a positive correlation between gliding and invasion inhibition (P=0.02, Spearman's ρ=0.60). Survival analysis indicated longer time to infection in individuals displaying higher-than-median sporozoite gliding inhibition activity (P=0.01). Conclusions: In summary, functional antibodies against the pre-erythrocytic stages of malaria infection are acquired in children who are repeatedly exposed to Plasmodium parasites. This immune response does not prevent them from becoming infected during a malaria transmission season, but might delay the appearance of blood stage parasitaemia and consequently needs to be considered in the evaluation of malaria vaccines.

Data from: Detection of airborne genetically modified maize pollen by real-time PCR
The cultivation of genetically modified (GM) crops has raised in the European Union and other parts of the world numerous concerns about their environmental and economic impact. Especially outcrossing of genetically modified organisms (GMO) was from the beginning a critical issue as airborne pollen has been considered an important way of GMO dispersal. Here, we investigate the use of airborne pollen sampling combined with microscopic analysis and molecular PCR analysis as an approach to monitor GM maize cultivations in a specific area. Field trial experiments in the European Union and South America demonstrated the applicability of the approach under different climate conditions, in rural and semi-urban environment, even at very low levels of airborne pollen. The study documents in detail the sampling of GM pollen, sample DNA extraction and real-time PCR analysis. Our results suggest that this ''GM pollen monitoring by bioaerosol sampling and PCR screening" approach might represent an aid in managing the co-cultivation of GM and non-GM crops, especially in the surveillance of GM-free areas, centres of origin, and natural reserves.

Data from: Ready to detect a reversal of time's arrow: a psychophysical study using short video clips in daily scenes

It is generally believed that time flows in one direction and that a reversal of time's arrow would render the external world nonsensical. We evaluated our ability to tell the direction of time's arrow in a wide range of dynamic scenes in our daily life by presenting 360 video clips in the correct or incorrect direction. Participants, who judged the direction in a speeded manner, erred in 39% of trials when a video was played in reverse, but in only 9% when it was played normally. Due to the bias favouring the "forward" judgement, reaction was generally faster for the forward response. However, the reaction became paradoxically faster and more synchronous for the detection of reversal in some critical occasions such as forward motion, free fall, diffusion, division, and addition of materials by hand. Another experiment with a fraction of the video clips revealed that reversal replay of these videos provided instantaneous evidence strong enough to overtake the forward judgment bias. We suggest that our brain is equipped with a system that predicts how the external organisms behave or move in these critical occasions and that the prediction error of the system contributes to the fast "reversal" detection.

Real-time Welding Sparks Detection on Construction Sites us-ing Contour Detection and Deep Learning

One of the primary causes of fires at construction sites is welding sparks. Fire detection systems utilizing computer vision technology offer a unique opportunity to monitor fires in construction sites. However, little effort has been made to date in regards to real-time tracking of small sparks that can lead to major fires at construction sites. In this study, a novel method is proposed to detect welding sparks in real time contour detection with deep learning parameter tuning. An automatic parameter tuning algorithm employing a convolutional neural network was developed to identify the optimum hue-saturation-value. Additional filtering methods regarding non-welding zone and contour area-based filter, were also newly developed to enhance prediction accuracy of welding sparks. The method was evaluated using 230 welding sparks images and 104 videos. The results obtained from the welding images indicate that the suggested model for detecting welding sparks achieves a precision of 74.45% and a recall of 63.50% when noise images, such as flashing and reflection light, were removed from the dataset. Furthermore, our findings demonstrate that the proposed model is effective in capturing the number of welding sparks in the video dataset, with a 95.2% accuracy in detecting the moment when the number of welding sparks reaches its peak. These results highlight the potential of automated welding sparks detection for enhancing fire surveillance at construction sites.
