Vegetation Surveys in Chihuahuan Desert Grassland and Shrubland Sites Associated with Coyote Scat Surveys at the Sevilleta National Wildlife Refuge, New Mexico (2008-2009)
This data set contains information regarding vegetation structure at sites in grama grassland and both creosote and mesquite shrubland habitats at the Sevilleta NWR. This information was collected at randomly selected sites throughout the refuge. Each site is within 100 meters of one of the 22 road-based transects(20 in 2008) that were used to carry out coyote scat surveys during three seasons (spring, summer and fall) in 2009 (see "Coyote scat surveys in grassland and shrubland sites at the Sevilleta NWR, spring, summer and fall 2009" data set). Data was collected within at total of 22 circular vegetation plots (40 in 2008), each of which is 30m in diameter. Each plot was surveyed a total of three times, specifically in: April (spring), July (summer), and October (fall) 2009. Variables were selected based on their relevance to patterns of coyote habitat use, as well as their utility in calibrating Landsat images of the study site and the likelihood that they would vary seasonally. Measured variables include: average percent live woody vegetation cover, average percent live grass cover, average percent live forb cover, and average woody plant height. Information on woody plant species with individuals greater than 0.5 m in height is also presented.

Data from: Public questions spur the discovery of new bacterial species associated with lignin bioconversion of industrial waste
A citizen science project found that the greenhouse camel cricket (Diestrammena asynamora) is common in North American homes. Public response was to wonder "what good are they anyway?" and ecology and evolution guided the search for potential benefit. We predicted that camel crickets and similar household species would likely host bacteria with the ability to degrade recalcitrant carbon compounds. Lignocellulose is particularly relevant as it is difficult to degrade yet is an important feedstock for pulp and paper, chemical, and biofuel industries. We screened gut bacteria of greenhouse camel crickets and another household insect, a hide beetle (Dermestes maculatus) for the ability to grow on and degrade lignocellulose components as well as the lignocellulose-derived industrial waste product black liquor. From three greenhouse camel crickets and three hide beetles, 14 bacterial strains were identified capable of growth on lignocellulosic components, including lignin. Cedecea lapagei was selected for further study due to growth on most lignocellulose components. The C. lapagei secretome was identified using LC/MS/MS analysis. This work demonstrates a novel source of lignocellulose-degrading bacteria and introduces an effective workflow to identify bacterial enzymes for transforming industrial waste into value-added products. More generally, our research suggests the value of ecologically-guided discovery of novel organisms.

Multiomics analyses reveal a Type III-associated immune response in immunotherapy-induced toxicity in melanoma and identifies potential new therapeutic targets
<p>Immune checkpoint inhibitors (ICIs) are standard-of-care for the treatment of advanced melanoma, but their use is limited by immune-related adverse events (irAEs). Proteomic analyses and multiplex cytokine/chemokine assays from serum at baseline and at irAEs onset in melanoma patients indicated aberrant T-cell activity with differential expression of Type I and III immune signatures. This was in line with an increase in the proportions of monocytes and decrease of IL-17A-producing CD4+ T-cells in the peripheral blood using single cell RNA sequencing. Multiplex immunohistochemistry and spatial transcriptomics on ICI-induced skin rash and colitis showed an increase in the proportion of CD4+ T-cells with IL-17A expression. </p> <p>Anti-IL-17A mAbs were administered in two patients with myocarditis, colitis and skin rash with resolution of the irAE. This study highlights the potential role of Type III CD4+ T-cells in irAEs development and provides proof-of-principle evidence for a corresponding clinical trial using anti-IL17A for treating irAEs.</p> <p>Code to generate the figures are located at https://github.com/pcheng84/AE_analysis/</p> <p> </p>
