Semisynthetic 6-Lattice Diffraction Data Re-binned

This macromolecular crystallography diffraction data set was derived from the six-lattice "acegik" data set published at https://doi.org/10.5281/zenodo.10820. Please see that entry for details. The original entry was authored by Richard Gildea and Graeme Winter at Diamond Light Source and also licensed under the Creative Commons Attribution 4.0 International Licence.

The number of images was reduced from 100 to 25 by merging them in groups of four using the program dials.merge_cbf. The resulting data set still represents at 10° rotation, but each individual image now covers 0.4°.

Alongside the images, the files refined.expt and refined.refl are provided, which contain the refined experimental geometry and strong spot locations in a format compatible with, at least, DIALS 3.6.

This data set has many overlapping spots and is provided for testing of overlap handling algorithms in diffraction data processing software.

Raw diffraction images of apo-trypsin

Trypsin is an enzyme in the first section of the small intestine that starts the degestion of protein molecules by cutting these long chains of amino acids into smaller pieces.

We used datasets to investigate the protocol of detecting polymorphs using Hierarchical clustering (Acta D., submitted). All diffraction data were collected at BL32XU, SPring-8, using an automated data collection system ZOO. Data were acquired from four crystals for apo-form, 4-Methoxybenzamidine(benzamidine)-bound, and 5-Chlorotryptamine(tryptamine)-bound trypsin crystals, respectively. All datasets were collected using a continuous helical scan scheme for 360º oscillation with the following experimental parameters; Beam size: 10 µm × 15 µm, Wavelength: 1.0000 Å, Total dose/crystal: 10 MGy, Detector: EIGER X 9M (DECTRIS Co. Ltd.). All crystals belonged to the space group P212121 with the unit cell parameters roughly corresponding to a=54.5, b=58.4, 66.8 Å.

Raw diffraction images of Spirochaeta YeeE of SeMet derivative

Crystal structure of thiosulfate transporter YeeE from Spirochaeta thermophila (PDB code 6LEO).

The complete datasets were obtained by merging multiple small-wedge datasets using EIGER X 9M detector at a wavelength of 0.979 Å with a multi-crystal data collection scheme using 15×10 μm beam on BL32XU, SPring-8. For the SeMet dataset, diffraction images of 7° rotation range with a pitch of 0.1° oscillation range were collected from each of the 326 crystals.
