Homocysteine, folic acid, cyanocobalamin, and frailty in older people: findings from the "Invece. Ab" Study
<p><strong>The database includes</strong> the raw data of the article “Homocysteine, Folic Acid, Cyanocobalamin, and Frailty in Older People: Findings From the “Invece. Ab” Study”.</p> <p><strong>Aim of the work</strong> was to verify the association of homocysteine (HOcy), cyanocobalamin (B12), and folic acid (FO) plasma concentrations with frailty and to underline the influence of these biomarkers on frailty incidence in a community dwelling older population, studied longitudinally (2012, 2014 and 2018 waves).</p> <p><strong>The database contains</strong> data obtained by the evaluations of plasma levels of i) HOcy, ii) B12 and iii) FO using chemiluminescent microparticle immunoassay on an Architect 2000 analyzer (Abbott) within 3h of blood collection. Briefly, we found that frail subjects increased from the 2014 wave to the 2018 wave of the study. Moreover, the median concentration of HOcy increased with age only in males and it showed a significant positive correlation with the Frailty Index (FI) at all three waves of the study; no positive correlation was instead reported for B12 and FO. HOcy seems to be a predictor of cumulative incidence of frailty (during the six years of the study), independent of age, sex, and education, although HOcy80 (19.4μmol/l - cutoff for the 80th percentile) showed only a positive trend in this sense. On the contrary HOcy90 (23.3μmol/l - cutoff for the 90th percentile) significantly influenced the cumulative incidence of frailty. B12 and FO showed no relationship with the cumulative incidence of frailty. Finally, subjects with plasma HOcy concentrations in the top quintile (HOcy80) had five fewer frailty-free months; this finding was independent of age, sex, and education. A comparable frailty-free survival time, was found for plasma HOcy concentrations in the top decile (HOcy90).</p> <p><strong>To conclude</strong>, high plasma concentrations of HOcy, could be a factor that increases the likelihood of becoming frail and becoming frail sooner; these results showed a clear association with frailty. Moreover, the role of B12 and FO plasma concentrations was found to be marginal.</p>
FRAILTY PSYCOVID
<p>Submitted materials consist of study dataset (Dataset_Frailty_Psycovid.xlsx, Excel format) which includes socio-demographic data of 1258 adult Italian residents who completed the survey (71.5% females; mean age: 43±13.5; age range: 18-81) and the clinical and psychosocial variables used in the study (i.e., frailty Index and Social vulnerability Index; loneliness, empathic skills, coping strategies, alexithymia), and 4-item scale of perceived impact of Covid-19 outbreak.  </p> <p> </p> <p>Study results showed: 1) a linear increase of frailty index with age and greater scores in females than in males; 2) higher social vulnerability in young adults and elders compared to middle aged and older adults, and in males than females; 3) both frailty and social vulnerability contributed in explaining the individual perception of the impact of Covid-19 emergency on health, which was further influenced by proactive attitudes/behaviors and social isolation.</p>
Inflammation and cell-to-cell communication, two related aspects in frailty
<p>The database includes the raw data of the article “Inflammation and cell-to-cell communication, two related aspects in frailty”. Aim of the work was to investigate inflammation and altered intercellular communication, both mechanisms involved in the biology of frailty, to ultimately identify if extracellular vesicles (EVs), specifically large EVs (lEVs) could play a role in the immune function in frail subjects.</p> <p>The database contains data obtained by means the following evaluations: i) Cobas 6000 automated analysis for the detection of c-reactive protein (CRP) plasma levels; ii) Meso Scale Discovery analysis for detection of cytokines (IL-1β, IL-6, TNFα, IFNγ and IL-13) plasma levels; iii) Nano Tracking Assay analysis for the detection of size and concentration of lEVs; iv) flow cytometry analysis for the detection of immunological markers (TLR2, TLR4, CD40 and CD120B) and senescence markers (CD221 and IL-6R) on lEVs.</p> <p>We found that CRP plasma levels increases in Fr subjects, thus suggesting a role of CRP as a possible biomarker in subjects experiencing symptoms of frailty. Moreover, we observed an increase in IL-1β, IL-6 and TNFα plasma levels in frail subjects. All these evidences point the robust involvement of inflammaging in frailty and suggest the role of circulating cytokines in the identification of frailty. We also focused on the role of lEVs and pro-inflammatory and pro-senescent hallmarks in frailty. We reported that lEVs size and concentration is not affected by frailty, howeverTLR2, TLR4 , CD40 and CD120b (TNF Receptor II) are significantly more expressed in lEVs of frail subjects respect to nFr. These results are indicative of a first and clear involvement of lEVs in the activation of pro-inflammatory pathways in frailty.</p> <p>Finally, in frail individuals we reported a significant increase in lEVs positive to markers of senescence: the IGF1receptor (CD221) and the IL6 receptor (IL6R). These evidences suggest IGF1 as a new potential biomarker of frailty, although new studies are needed. Moreover, positivity for the IL6R and the concomitant increase in circulating IL6 suggest a possible crucial role of lEVs in the delivery of cytokines through extracellular release and absorption. In conclusion, lEVs may contribute to the creation of a pro-inflammatory and pro-senescent microenvironment that acts as one of the major contributors to frailty.</p>
