Application of inverse theory for high spatial resolution reconstructions of thermospheric vector wind fields from Doppler shifts measured by a ground-based network of all-sky Fabry-Perot interferometers

Several types of all-sky viewing Fabry-Perot Interferometers (FPI) have been developed since the 1990s for ground-based remote sensing of thermospheric winds. The Scanning Doppler Imager (SDI) is one such instrument, which provides temporally simultaneous line-of-sight observations from hundreds of independent look directions per instrument exposure. A geographically distributed network of such instruments increases spatial coverage and, at many locations, also provides overlapping observations along multiple independent lines-of-sight. Together, these characteristics significantly increase the density and fidelity that is possible for reconstructed thermospheric vector wind fields, compared to a traditional narrow-field FPI, but at the cost of complexity and difficulty.

Presently, we describe an application of inverse theory to reconstruct three-component vector thermospheric neutral wind fields using data from multiple SDI instruments. The salient features of the method used here are the ability to reconstruct three-component winds on a dense grid that is sampled regularly in latitude, longitude, and time, without assuming any a-priori underlying structure of the winds. This requires solving an inverse problem that does not in general yield a unique solution unless additional constraints are enforced. We describe this step, also known also as regularization, along with the strategy used to maximize the spatial resolution of the derived wind fields by automatically determining the minimum level of regularization that can produce stable inversions. We present example results obtained from applying this technique to one night of data from a network of SDIs in Alaska, and discuss the implications of these results for current understanding of thermospheric dynamics.

The application of short and highly polymorphic microhaplotypes based on nonbinary-SNPs in kinship testing of extremely degraded samples

Kinship testing becomes more difficult in extremely degraded samples. But commonly used genetic markers cannot completely solved this problem. Microhaplotype combining the advantages of STR and SNP may be a promising genetic marker for kinship testing in extremely degraded samples. Therefore, in this study, 36 short and highly polymorphism microhaplotype loci with length smaller than 100 bp and Ae greater than 3.0 were developed, of which 29 loci met the Hardy-Weinberg and linkage equilibrium. The CPD and CPE of these 29 loci were 0.99999999999999999999999997036 and 0.9999999945, respectively. Allele dropout of these loci was not observed in extremely degraded samples. Through simulated kinship analysis, the effectiveness of paternity testing reached 98.39% at threshold of 4/-4, and effectiveness of full-sibling testing reached 93.01% at threshold of 2/-2, which were greater than that of 15 STR loci. After combining with other 50 short and highly polymorphic microhaplotype loci, the effectiveness of half-sibling testing also reached 82.42% at the threshold of 2/-2. Our developed short and highly polymorphic microhaplotype loci may be useful for paternity testing and full-sibling testing in extremely degraded samples, and after combining with other short and highly polymorphic microhaplotype loci, may be helpful to analyze the more distant kinship relationship.

Systemic application of the TRPV4 antagonist GSK2193874 induces tail vasodilation in a mouse model of thermoregulation

In humans, skin is a primary thermoregulatory organ, with vasodilation leading to rapid body cooling, whereas in Rodentia the tail performs an analogous function. Many thermodetection mechanisms are likely to be involved including transient receptor potential vanilloid-type 4 (TRPV4), an ion channel with thermosensitive properties. Previous studies have shown that TRPV4 is a vasodilator by local action in blood vessels, so here we investigated whether constitutive TRPV4 activity effects Mus muscularis tail vascular tone and thermoregulation. We measured tail blood flow by pressure plethysmography in lightly sedated Mus muscularis (CD1 strain) at a range of ambient temperatures, with and without intraperitoneal administration of the blood brain barrier crossing TRPV4 antagonist GSK2193874. We also measured heart rate and blood pressure. As expected for a thermoregulatory organ, we found that tail blood flow increased with temperature. However, unexpectedly we found that GSK2193874 increased tail blood flow at all temperatures, and we observed changes in heart rate variability. Since local TRPV4 activation causes vasodilation that would increase tail blood-flow, these data suggest that increases in tail blood flow resulting from the TRPV4 antagonist may arise from a site other than the blood vessels themselves, perhaps in central cardiovascular control centres.

A new tree-based methodological framework to infer the evolutionary history of Mesopolyploid lineages: An application to the Brassiceae tribe (Brassicaceae)

Whole genome duplication events are notably widespread in plants and this poses particular challenges for phylogenetic inference in allopolyploid lineages, i.e. lineages that result from the merging of two or more diverged genomes after interspecific hybridization. The nuclear genomes resulting from allopolyploidization contain homologous gene copies from different evolutionary origins called homoeologs, whose orthologs must be sorted out in order to reconstruct the evolutionary history of polyploid clades. In this study, we propose a methodological approach to resolve the phylogeny of allopolyploid clades focusing on mesopolyploid genomes, which experienced some level of genome reshuffling and gene fractionation across their subgenomes. To illustrate our methodological framework, we applied it to a clade belonging to the model Brassicaceae plant family, the Brassiceae tribe, that experienced a mesohexaploidy event. The dataset analysed consists of both publically available genomic sequences and new transcriptomic data according to taxa. The present methodology requires a well-annotated reference genome, for which the identification of the parental subgenome fragments has been performed (e.g. Brassica rapa and Brassica oleracea). Focusing on fully retained genes (i.e., genes for which all homoeologous gene copies inherited from the parental lineages are still present in the reference genome), the method constructs multilabelled gene trees that allow subsequent assignment of each gene copy to its diploid parental lineage. Once the orthologous copies are identified, genes from the same parental origin are concatenated and tree-building methods are used to reconstruct the species tree. This method allows resolving the phylogenetic relationships (i) among extant species within a mesopolyploid clade, (ii) among the parental lineages of a mesopolyploid lineage, and (iii) between the parental lineages and closely related extant species. We report here the first well-resolved nuclear-based phylogeny of the Brassiceae tribe.
